Objective: To research the expression of PTEN and its influence on biological ability in breast cancer cell in vivo. Methods: PTEN-shRNA plasmid was transtected into M231 breast cancer cells to knock down the expression of PTEN. The changes of PTEN expression, proliferation, adhesion and metastasis of PTEN knocked down cell were tested by western-blot, colony formation, adhesion and invasion assay. Results: PTEN-shRNA was successfully transfected into M231 cells and it inhibited PTEN expression efficiently. The capabilities of colony formation, migration and invasion of transfected cell were much greater than those of the controlling cell line. But the transfected cells were more difficulty in adhesion than the scrambled ones. Conclusion: PTEN genecan enhance the adhesion, but restrict the proliferation, migration of breast cancer cells in some degree, so that inhibit the development of the breast cancer. PTEN loss can be a prognostic factors for the patients with breast cancer.

Background Establising the culture model of Müller cells for obtaining the highly putified target cells is essential for the study about the physiology and pathology of retinal Müller cells. The exsiting purifing method for culturing Müller cells is dissatisfactory. Objective This study was to establish a method to obtain high purifing Müller cells. Methods The retina from 5 clean newborn SD rats were isolated and digested by 0. 01% trypsin and cultured in DMEM containing 10% fetal bovine serum. The cellular suspension was then prepared, and the target cells were screened using flow cytometry based on the size and the quantity of cells. Cultured and passaged cells were identified by transmission electron microscope and light microscope. Immunocytochemistry was used to detecte the expression of GFAP in cultured cells for the determination of type and purity of the cells. Results The cells showed the similar shape to retinal Müller cells after primarily culture with the large volume, and some small other types of cells could been seen. The growth of cells was quickly 3 weeks later. The fibroblasts were removed using sticking-wall by steps, and neurons were eliminated following passage. Aboundent of cellular organs were seen under the transmission electron microscope. The positive response rate of the cells for CFAP was 100%. Conclution Flow cytometry offer a rapid and feasible approach for purifying Muller cell and it builds the foundation for further study about Müller cells.

Chinese Journal of Orthopaedic Trauma 2004;0(07):-
Vertebral compression fractures, the most common complication of the osteoporosis, results in significant morbidity of prolonged and intractable pain in some patients. Vertebroplasty procedure that involves percutaneous injection of bone cement into a collapsed vertebra has recently been introduced to treat osteoporotic patients who have prolonged pain following vertebral compression fracture. To determine the details of the procedure and to gather information on its safety and efficacy, we performed a MEDLINE search using the terms ‘vertebroplasty’. We reviewed reports of these procedures in patients with osteoporosis. Several reports suggest that the vertebroplasty is associated with pain relief in 67% to 100% of the cases. Short-term complications, mainly the result of extravasation of cement, can increase pain and damage from heat or compression to the spinal cord or nerve roots. Proper patient selection and good technique can decrease such complications so that the decompression surgery is rarely needed. But long-term complications, which include foreign-body reaction at the cement-bone interface, the wear of the cement, and increased risk of fracture in the adjacent vertebrae due to changes of mechanical stresses, have not been fully evaluated. In short, there is still a long way to go for the vertebroplasty.
