1.Genetic analysis of gonadotropin-gonadal axis in boys: a twin study.

OBJECTIVETo analyse the heritabilities of serum luteinizing hormone(LH), follicle-stimulating hormone (FSH), testosterone (T) and estradiol (E2) in twin boys, and to study the genetic contributions to gonadotropin-gonadal axis.
METHODSA total of 51 pairs of male twins, 35 monozygotic (MZ) and 16 dizygotic(DZ) aged 5 to 11 years, were investigated. Serum gonadotropin and sex hormone were measured by radioimmunoassay. The twin zygosity was verified by determination of short tandem repeat amplified fragment length polymorphism systems. The genetic analysis was performed using intraclass correlation coefficient method.
RESULTSThe intraclass correlation coefficient was greater in the MZ twins than in the DZ twins. The estimated heritabilities were respectively LH 0.51, FSH 0.32, T 0.81, E2 0.41.
CONCLUSIONGenetic factors are major determinants of gonadotropin-gonadal axis in boys.

2.Study on the sexual development of adolescent male.

OBJECTIVESThe investigation of the testicular volume, the penis length and the T, FSH, LH, PRL levels in serum were taken in 289 adolescent males to provide the valuable data for andrology.
METHODSThe adolescent males were grouped according to their age. The testicular volume was measured with testicular model and the T, FSH, LH, PRL levels in serum were determined by immunoenzymetric assay.
RESULTSThe male sexual development was rapid from age 11 to 16 and close to that of adult at age 18. Serum PRL of adolescent males was higher than that of adult males.
CONCLUSIONSThe age 11 to 16 is a period of rapid growth in sexual maturation. PRL may play an important role in sexual maturation.

3.Effects of serum testosterone level on plasma lipid and apolipoprotein spectrum in orchiectomized rabbits.

AIMTo investigate the effects of serum testosterone level on plasms lipid and apolipoprotein spectrum in orchiectomized rabbits.
METHODS40 male New Zealand rabbits were divided randomly into five groups (n = 8): control group, sham (castration), castration group (castration), hypotestosteronemia group (castration plus low dose testosterone replacement), physiological level group (castration plus moderate testosterone replacement), and hypertestosteronemia group (castration plus large dose testosterone replacement). The rabbits in hypotestosteronemia, physiological level and hypertestosteronemia group were injected testosterone undecanoate 3 mg/kg, 6 mg/kg, and 12 mg/kg, respectively and the remaining control, and castration rabbits, vehicle only. 8 weeks after initiation of the experiment, blood was collected for determination of serum levels of total testosterone (TT), estradiol (E2), lipids, lipoproteins, apolipoproteins.
RESULTSAfter castration, the serum level of TT decreased significantly to the lowest level, and TU replacement increased serum TT level depending on dose. The low dose of TU increased serum TT level slightly and produced a hypotestosteronemia model, the moderate dose of TU increased serum TT level near to normal level and produced a physiological model, the large dose of TU increased serum TT level significantly to the highest level and produced a hypertestosteronemia model. The tatol cholesterol (TC), triglycerides (TG), low density lipoprotein cholesterol (LDL-C), apolipoprotein B (ApoB) were significantly increased in hypotestosteronemia and hypertestosteronemia rabbits and significantly decreased in physiological serum testosterone rabbits.
CONCLUSIONTestosterone, at physiological level has good effect on the serum lipids, lipoproteins, apolipoproteins in the castration rabbits, and the hypotestosteronemia and hypertestosteronemia has bad effect.

4.Sexual development characteristics and sex hormone levels in obese male adolescents.

OBJECTIVETo investigate the characteristics of sexual development and sex hormone levels in obese male adolescents.
METHODSWe included 156 obese male adolescents with micropenis and microorchidia in an observation group and 50 healthy ones in a control group. We measured the body mass index (BMI), penile natural length and testicular volume, investigated the incidence of spermatorrhea and the age of the first spermatorrhea, detected the levels of serum luteinizing hormone (LH), follicle stimulating hormone (FSH), prolactin (PRL), total testosterone (TT), free testosterone (FT), progesterone (P) and estradiol (E2) using radioimmunoassay, and calculated TT/E2 and testosterone secretion index (TSI).
RESULTSCompared with the healthy controls, the obese adolescents showed significantly higher BMI ([20.4 +/- 1.6] vs [27.1 +/- 2.2] kg/m2, P < 0.05), but shorter penile natural length ([6.7 +/- 2.1] vs [5.6 +/- 1.7] cm, P < 0.05) and lower testis volume ([9.9 +/- 3.1] vs [7.6 +/- 2.3] cm3, P < 0.05). The incidence of spermatorrhea was significantly decreased in the observation group in comparison with that of the control (chi2 = 17.335, P < 0.05), but there was no significant difference in the age of the first spermatorrhea between the two groups (P > 0.05). The levels of LH, E2 and P were remarkably higher in the observation group than in the control ([7.82 +/- 2.14] vs [5.39 +/- 1.76] mIU/ml, P < 0.05; [48.57 +/- 8.34] vs [8.61 +/- 4.08] pg/ml, P < 0.01; and [1.25 +/- 0.58] vs [0.64 +/- 0.19] ng/ml, P < 0.05), while TT and FT were markedly lower in the former than in the latter ([0.73 +/- 0.20] vs [1.47 +/- 0.41] ng/ml, P < 0.01 and [5.09 +/- 2.60] vs [11.28 +/- 4.72] pg/ml, P < 0.01), and so were the TT/E2 ratio and TSI (0.015 +/- 0.004 vs 0.173 +/- 0.037 and 0.098 +/- 0.026 vs 0.272 +/- 0.084, P < 0.01). BMI was correlated positively to PRL and E2, but negatively to TT, FT, TT/E2 and TSI (P < 0.05); the penile natural length positively to TT, FT, TT/E2 and TSI, but negatively to E2 (P < 0.05); and the mean testis volume positively to TT, FT, TT/E2 and TSI, but negatively to LH, PRL and E2 (P < 0.05).
CONCLUSIONTestis dysplasia and alteration of sex hormone levels exist in obese male adolescents. Obesity and fat accumulation lead to increased E2 and decreased TT and FT, particularly the reduction of TT/E2 and TSI, which suggest that the body fat content has an important influence on the development of the male reproductive system.

5.Effect of sequential method of nourishing yin and tonifying yang on serum sex hormone levels in rats with polycystic ovary syndrome.

OBJECTIVETo observe the effect of sequential method of nourishing yin and tonifying yang on serum sex hormone levels in rats with polycystic ovary syndrome (PCOS), and to explore its possible mechanisms.
METHODSPCOS rat models were established by injecting dehydroepiandrosteron (DHEA) subcutaneously. A total of 70 21-day female SD rats were randomly divided into seven groups, ten in each group: the normal control, the model control, the yin nourishing, the yang tonifying, the sequential method, the Westem medicine, and the combined medication. Rats in all groups were treated by gastrogavage for twelve successive days. Serum testosterone (T) and estradiol (E2) were detected by chemoluminescence method. Serum androstenedione (A), follicle stimulating hormone (FSH), luteinizing hormone (LH), insulin (INS), and insulin-like growth factor-I (IGF-I) were detected by radioimmunoassay (RIA).
RESULTSSerum levels of A, T, and E2 were significantly higher in the model control group than in the normal control group (P < 0.05, P < 0.01). Serum levels of A and T were obviously lower in the sequential group than in the model control group with significant difference (P < 0.01). The serum E2 level was obviously lower in the sequential group than in the model control group (P < 0.05). Serum levels of INS and IGF-I were significantly higher in the model control group than in the normal control group (P < 0.05). Serum levels of INS and IGF-I were significantly lower in the sequential group than in the model control group (P < 0.01).
CONCLUSIONSThe sequential method of nourishing yin and tonifying yang can lower serum levels of A, T, E2, INS, and IGF-I, and reverse its endocrine disturbance in PCOS rats. One of its mechanisms in decreasing serum levels of A, T, E2 might be possibly through lowering serum levels of INS and IGF-I.

6.Effects of guizhi fuling capsule on sex hormone levels and breast issue morphology of mammary gland hyperplasia model rats.

OBJECTIVETo research the effects of Guizhi Fuling capsule on sex hormones levels in blood serum and breast issue morphology of hyperplasia of mammary glands model rats.
METHODThe unpregnancy SD rat models of hyperplasia of mammary glands were established by injecting 0.5 mg x kg(-1) benzoate estradiol. After five weeks doses,the effects of Guizhi Fuling capsule 2.0, 1.0, 0.5 g x kg(-1) and Rupixiao tablet 0.5 g x kg(-1) on the changes of papilla diameter, height and breast issue morphology of the naimal models were explored, and sex hormones levels in blood serum were measured.
RESULTGuizhi Fuling capsule can inhibitnipple swell, improve breast tissue morphology pathological profiles of the animal models, and decrease oestradiol (E2) level and increase progesterone (P) level in blood serum.
CONCLUSIONThese results suggested that Guizhi Fuling capsule could, improve mammary gland pathological profiles. Regulating sex hormone levels may be its important mechanism for treatment of hyperplasia of mammary glands.

7.Effect of chaishu sijun decoction on the gonad axis of gan-qi stagnation, Pi deficiency, and gan-qi stagnation pi deficiency model rats.

OBJECTIVETo observe changes of gonad functions of Gan-qi stagnation (GS), Pi deficiency (PD), Gan-qi stagnation Pi deficiency (GSPD) model rats, and the effect of Chaishu Sijun Decoction (CSD) on them.
METHODSRats were randomly divided into 7 groups according to romdom digit table, i.e., the normal control group, the GS model group, the GS medication group, the PD model group, the PD medication group, the GSPD model group,and the GSPD medication group, 10 in each group. Rats in the GS model group, the PD model group, and the GSPD model group were treated with chronic restraint, improper diet +excessive fatigue, chronic restraint +improper diet +excessive fatigue. The model was established for 4 successive weeks. Starting from the 15th day of modeling, CSD at the daily dose of 3.57 g/kg was given by gastrogavage to them for 14 successive days. Equal volume of distilled water was given by gastrogavage to rats in each model group and the normal control group for 14 successive days. The blood contents of gonadotrophin releasing hormone (GnRH), follicular stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), and testosterone (T) were detected in rats of each group.
RESULTSCompared with the normal control group, there was statistical difference in GnRH, T, E,, and FSH in the GS, PD, and GSPD model groups (P < 0.05, P < 0.01). The content of LH was elevated in the GS model group (P < 0.05) and declined in the GSPD model group (P < 0.01). Compared with the GS model group, the contents of FSH, LH, and T decreased and E2 increased in the PD model group (all P < 0.05); the contents of FSH and LH also declined in the GSPD model group (P < 0.05). Compared with the PD model group, the T content increased and FSH decreased in the GSPD model group (all P < 0.05). Compared with each corresponding model group, the FSH content decreased (P < 0.01) and LH increased in the GS medication group; the T content increased, E2 and LH decreased (P < 0.05, P < 0.01) in the PD medication group; the T content decreased (P < 0.01), GnRH, E2, FSH, and LH increased (P < 0. 05, P < 0.01) in the GSPD medication group.
CONCLUSIONSThere exist different degrees of abnormal function of the gonad axis in the GS, PD, and GSPD models. CSD had certain regulatory effect on the 3 syndromes. Of them, it showed a more comprehensive role in improving the gonad function axis. Results of this experiment had provided the experimental evidence for higher correlation between CSD and GSPD syndrome.

8.Sex hormones in female patients with systemic lupus erythematosus.

OBJECTIVE:
To explore the levels of sex hormones in female patients with systemic lupus erythematosus (SLE) and its role in the genesis of SLE.
METHODS:
The serum levels of estradiol, estriol, testosterone, progesterone, and prolactin were determined by electro-chemiluminescence immunoassay in 98 female patients with SLE and compared with those of 38 healthy women.
RESULTS:
The serum levels of estradiol, estriol, progesterone, and prolactin were significantly higher than those in the healthy women (P <0.05). The serum levels of estradiol, progesterone, and prolactin in patients with SLE in 25 to 34 year old group were higher than the other age groups and the control group (P < 0.05), and the serum levels of estradiol and prolactin in patients with active phase of SLE were significantly higher than those in patients with stable phase of SLE (P <0.05).
CONCLUSION
The levels of sex hormones have a close corretation with the genesis and development of SLE.
