OBJECTIVETo provide the basic guidance for seed breeding and cross-breeding of Paris polyphylla var. yunnanensis.

METHODThe floral behavior and pollinators were observed; 0.5% TTC solution was used for the pollen viability test and benzidine and -H2O2 was used for estimation of the stigma receptivity. The mating systems were tested by out crossing index (OC1), and pollination experiment was carried out by bagged and emasculated test in the field.

RESULTCommonly, stigma lobes spread slightly, and anthers started presenting the pollen from the outer ring while the flower was just beginning to open. Consequently, the distance between the stigma and its own pollen was relatively far, this "floral behavior" may be conducive to outcrossing. Then the flower entered the later period, while the stigma lobes spread widely, anthers all split, and this "floral behavior" shortened the stigma and its own pollen's distance, which may be conducive to selfing. P. polyphylla was partly protogynous. Stigma life-span was about 10-12 d. After anther dehiscence, the pollen viability maintained about 10% within 2 days, and 20% within 10 days. The value of out crossing index (OC1) was 4. By pollination experiment and pollinators observed, P. polyphylla was self-compatible, but no capacity for autonomous self-fertilization; In natural circumstances, outcrossing fructification rate was low, and mainly anemophilous. Assisted selfing-fertilization fructification rate was higher, spider was the main pollinators.

CONCLUSIONP. polyphylla has a mixed mating system with self-pollination and cross-pollination characteristics. Floral behavior has important adaptive significance in avoiding female and male interference, outcrossing, and delayed selfing. P. polyphylla is ambophily (a combination of both wind and insect pollination), pollinators changes due to environment. Pollen limitation is the main cause of low fructification rate under natural conditions.

OBJECTIVETo determine the self-compatibility of Platycodon grandiflorum and the location of microspore germination on stigma.

METHODThe microspore germination of self-pollination, self-plant-pollination and cross-pollination and the pollination microspore germination in and outside of stigma were observed with fluorescence microscopy.

RESULTMost pollens of self-pollination, self-plant-pollination and cross-pollination can germinate on the stigma, and after 24 hours, pollen tube entered into the ovary successfully. Pollinated on the outer-surface of stigma, microspores could not germinate, but on the inner-surface of the stigma when it dehisced most microspores can germinate.

CONCLUSIONThe compatibility of self-plant-pollination of Platycodon grandiflorum is high. The microspore germination loci is on the inner-surface of the dehisced stigma.

The endoplasmic reticulum quality control (ER-QC) is a conserved mechanism in surveillance of secreted signaling factors during cell-to-cell communication in eukaryotes. Recent data show that the ER-QC plays important roles in diverse cell-to-cell signaling processes during immune response, vegetative and reproductive development in plants. Pollen tube guidance is a precisely guided cell-cell communication process between the male and female gametophytes during plant reproduction. Recently, the female signal has been identified as small secreted peptides, but how the pollen tube responds to this signal is still unclear. In this review, we intend to summarize the role of ER-QC in plants and discuss the recent advances regarding our understanding of the mechanism of pollen tube response to the female signals.

OBJECTIVETo study the pollen germination in vitro and pollen tube growth of Scutellaria baicalensis.

METHODTwo years-old S. baicalensis that in the experiment field of Shangluo university were used as the materials. The effects of culture medium, sucrose, PEG, pH value, culture time and temperature on the pollen germination and tube growth of S. baicalensis were investigated using the method of liquid culture.

RESULT AND CONCLUSIONThe results showed that 100 g x L(-1) sucrose was helpful for the germination and growth of pollens. However, higher concentration of sucrose would inhibit strongly pollen germination and the growth of pollen tubes. PEG could accelerate pollen germination and tube growth, and the optimal concentration was 150 g x L(-1). The optimum culture medium was BK and BK + 10% sucrose + 150 g x L(-1) PEG (pH 5.8) , and the latter was more benefit for pollen germination and tube growth. The pollen of germination was better at 25-35 degrees C, and it was the best at 30 degrees C for 2-3 hours, while pollen tube growth was slower at 25 degrees C.

Mei (Prunus mume Sieb.et Zucc.) is traditionally not only a famous special ornamental plant but also a fruit tree origined in China. In order to conserve and ultilize scientifically the germplasm resources of wild mei, we identified and analysed the germplasm of mei flower in 65 samples collected from the habitat, using AFLP makers in combination with morphological anaylsis. This study amplified clearly 1 728 polymorphic bands, using the 8- pair-primer of Mse I -EcoR I screened totally from 64 -primer combination. According to the Nei' 72 distance coefficient clustering, all of the formas and varieties used in this study, including Prunus mume var. mume, P. mume, P. mume var. goethartiana, P. mume var. pallescens, P. mume var. microcarpa, P. mume var. cenrnus-sempervirens (newly recoeding variety), P. mume var. cernua, P. mume var. pallidus, P. mume var. taomei, were identified at the point of Nei' 72=0.26. Due to the genetic difference obviously among the formas and varieties, we suggested that wild germplasmtypes of all formas and varieties in P. mume should be conserved in their habitats in the furture.

OBJECTIVETo study four kinds of germplasm resources of Codonopsis pilosula and provide the basal mating systems data for the breeding and cultivation of C. pilosula.

METHOD0.5% TTC (2,3,5-triphenyl tetrazolium chloride) solution was used for the pollen viability test and benzidineand-H2O2 [1% benzidine in 60% ethanol,hydrogen peroxide (3%), and water, 4:11:22] was used for estimation of the stigma receptivity. The mating systems were tested by out crossing index (OC1), pollen-ovule ratio (P/O) and pollination by bagged and emasculated in the field.

RESULTThe pollen viability of C. pilosula reached highly about 80% when the pollen staying in the anther, 2-3 days before the petals opening. The anther began scattering pollen before the day of the petals opening, the pollen viability was the highest about 95%, the pollen stick thickly aroud the stigma and quickliy lost in the next day. The stigma life-span was about 4-5 days, the optimal time for pollination was the first day of the petals opening, when the stigma was highly sticky and yellow. The value of out crossing index (OC1) was 4, pollen-ovule ratio was between 104.84-185.75. The natural fructification rate of cross-pollination by emasculated-treatment was 25.6% 42.4%. The fructification rate and compatible index of self-pollination by bagged- treatment were about 3.3%-6.7% and 3.0-21.8.

CONCLUSIONThe mating system of C. pilosula is mixed with self-pollination and cross-pollination, prone to cross-pollination. The compatibility of self-pollination is high. The difference of maturing period of pistil and stamen and the lack of polen amount cause low fructification rate of self-pollination.

OBJECTIVETo study the relationship between Cibotium barometz and its companion species, Arachniodes falcate and Alpinia japonica, we used aqueous leachates of the two companions to deal with C. barometz spores.

METHODSpores of C. barometz were translated on MS culture which contained different concentration of aqueous extracts of the two companions, the germination and gametophyte development were observed and recorded.

RESULTAll extracts inhibited and delayed the C. barometz spores germination and rhizoid elongation was inhibited. It also had obvious inhibition to the prothallus formation and sexual differentiation. And the higher concentration, the more obvious inhibition of aqueous extracts of the two companion species.

CONCLUSIONThe two companion species have allelopathic effects on the spore germination and gametophyte development of C. barometz. And it may have an influence on sporogon ontogenesis and the population expansion.

Lectins are glycoproteins that specifically bind carbohydrate structures and may participate in the biodefense mechanisms of fish. In this study, the binding of three lectins, Dolichos biflorus agglutinin (DBA), soybean agglutinin (SBA), Bandeiraea simplicifolia BS-1 (isolectin B4), Triticum vulgaris (WGA), Arachis hypogaea (PNA) and Ulex europaeus (UEA-I) were studied in the gill, liver, intestine, kidney, heart, and spleen of the flat fish Paralichthys olivaceus. DBA was detected in intestinal mucous cells, as well as in gill epithelial and mucous cells. It was weakly detected in renal tubule epithelial cells and in bile duct epithelial cells. The strong SBA staining was seen in the intestinal club cells, in bile duct epithelial cells and renal tubule epithelial cells. There were intense positive reactions for isolectin B4 in gill epithelial and mucous cells, and the strong isolectin B4 staining was seen in epithelial cells of the bile duct and intestine. The strong WGA staining was seen in the gill mucosal cells, sinusoid, renal tubule epithelial cells and mucosal cells of the intestine. UEA-I was detected in the gill epithelial and mucosal cells, bile duct epithelial cells and renal tubular epithelial cells. These results suggest that the six lectins examined were localized in the covering epithelia of the various organs of the flat fish and they may participate in the biodefense mechanism of the intra body surface in which is exposed to various antigens.

Recently the safety of Heshouwu become a focus, but the reasons of its hepotoxicity are confused. On the basis of literature research, some findings on species and usage custom maybe supply some clues to explain the reasons of its hepotoxicity. Heshouwu had red Heshouwu (male) and white Heshouwu (female) in ancient literature, and traditional usage was use of the male and female together. The Latin name of red Heshouwu is Fallopia multiflora, and that of the white one is F. multiflora var. multiflora.
