2.Characteristics and risk factor analysis of femoral supracondylar fractures combined with meniscal and?ligamentous knee?injuries

Objective:To analyze the characteristics and related risk factors of femoral supracondylar fractures combined with meniscus and ligament injuries.Methods:A retrospective case series analysis was performed on clinical data of 100 patients with femoral supracondylar fractures combined with ipsilateral meniscal and?ligamentous knee?injuries?admitted to Tianjin Hospital from January 2016 to December 2020. There were 55 males and 45 females,with age of 22-78 years[(38.5 ± 3.3)years]. Causes of injury included traffic accidents in 45 patients,falls in 45 and others in 10. According to AO classification,type A1 was in 56 patients,A2 in 33 and A3 in 11. Sixty of these patients presented fractures on the left side and forty on the right side. All patients accepted MRI examination to estimate meniscal and ligamentous knee injuries concerning its prevalence,locations and severity. Locations of injury included the medial meniscus(MM),lateral meniscus(LM),anterior cruciate ligament(ACL),posterior cruciate ligament(PCL),medial collateral ligament(MCL)and lateral collateral ligament(LCL). Meniscal knee injuries were measured using Stoller four-level method,and ligamentous knee injuries by Ruiz three-level method. The patients were subdivided based on age,sex,causes of injury,injury side and fracture AO classification to measure the prevalence of meniscal and?ligamentous injuries,and to identify the factors related to injuries using logistic regression analysis.Results:Seventy-six patients(76.0%)were complicated with meniscal or ligamentous injuries in 151 different parts. Twenty-nine patients(29.0%)had both meniscal and ligamentous injuries. Fifty-six patients had meniscal injuries,including 11 with bilateral meniscus injuries. MM injuries were noted in 32 patients(32.0%),with severity of grade Ⅰ in 19 patients,grade Ⅱ in 12 and grade Ⅲ in 1. LM injuries were found in 35 patients(35.0%),with severity of grade Ⅰ in 19 patients,grade Ⅱ in 14 and grade Ⅲ in 2. Forty-nine patients had ligamentous injuries,among which 22 patients were with single ligamentous injuries,19 with double ligamentous injuries and 8 with triple ligamentous injuries. ACL injuries were found in 33 patients(33.0%),with severity of grade Ⅰ in 22 patients,grade Ⅱ in 10 and grade Ⅲ in 1. PCL injuries occurred in 4 patients(4.0%),with severity of grade Ⅰ in 3 patients and grade Ⅱ in 1. MCL injuries occurred in 28 patients(28.0%),with severity of grade Ⅰ in 18 patients,grade Ⅱ in 9 and grade Ⅲ in 1. LCL injuries occurred in 19 patients,with severity of grade Ⅰ in 12 patients,grade Ⅱ in 5 and grade Ⅲ in 2. There were significant differences in prevalence of meniscal injuries among subgroups of different gender,causes of injury and fracture types( P < 0.05).Logistic regression analysis suggested that gender and fracture types showed significant correlation with meniscal injuries,especially factors of males( P < 0.05)and type A3 fractures( P < 0.01). There were significant differences in prevalence of ligamentous injuries among subgroups of different causes of injury and fracture types( P < 0.05). Logistic regression analysis showed that causes of injury and fracture types were significantly related to ligamentous injuries,especially factors of traffic accidents( P < 0.01)and type A3 fractures( P < 0.01). Conclusions:Ipsilateral meniscal or ligamentous knee injuries have a high incidence in patients with supracondylar femoral fractures. There exist multiple-site injuries,particularly in ligamentous injuries. Males,type A3 fractures and traffic injuries are risk factors,which needs to be paid much attention in clinical diagnosis and treatment.

ObjectiveTo characterize the neural progenitor cell in the human amnion mesenchyme and epithelial layer with specific mark proteins of neural stem cell.MethodsExpressions of specific mark proteins of neural stem cell including nestin, glial fibrillary acidic protein (GFAP), musashi-1, vimentin and PSA-NCAM in human amnion tissue and cultured amniotic cells were determined by immunohistochemistry and immunofluorescence staining.ResultsExpressions of pluripotent neural stem cell specific makers (nestin, musashi-1, vimentin and PSA-NCAM) were detected in the human amnion mesenchyme and epithelial layer. In addition, cultured amniotic cells were expressed several neural stem cell specific markers including nestin, GFAP and PSA-NCAM. Nestin+ and GFAP+ double positive cells were identified in the human amnion tissue and cultured amniotic cells by immunohistochemistry and immunofluorescence staining.ConclusionSpecific mark proteins of neural stem cell are expressed in human amnion tissue and cultured amniotic cells.
5.Rhizosecretion of HIV-1 Recombinant Capsid Protein from Transgenic Lycium barbarum L.Hairy Roots

HIV p24 core protein can induce both cellular and neutralizing antibody responses.HIV-1 CA-virus-like particles(VLPs)vaccines provide a promising approach for the development of an effective vaccination strategy against HIV infection.Rhizosecreion of the recombinant proteins provides a new manufacturing platform that can simplify the extraction and purification procedure.Lycium barbarum L.was transformed by Agrobacterium tumefaciens EHA105 harboring the plant expression vector pCAMBIA1305.2-MA4-CA with a GRP signal peptide and MA4-CA fusion gene.Transgenic hairy roots were induced and cultivated in hydroponic culture.Western blotting indicated that the recombinant CA proteins were present in two forms,a glycosylated monomer(37 kDa)and a dimer(50 kDa)in the roots and hydroponic medium.It appeared from the present immunohistochemical data that the recombinant CA proteins fused with GRP signal peptide were confined to the cytoplasm,cell wall and intercellular space,indicating targeting into the secretory pathway.It demonstrated for the first time the rhizosecretion of HIV-1 recombinant capsid protein in Lycium barbarum L.hairy roots,and may offer a novel method for expressing HIV-1 CA-VLPs vaccines in plants.
6.The rate-adaptive response study of integrated sensor pacemakers

Objective To observe the rate-adaptive response of dual sensor pacemakers integrated with activity sensor and minute ventilation sensor.Methods Fifty patients with chronotrepic incompetence were implanted with pacemakers of integrated sensor.The patients with pacemakers implanted were arranged to take upstairs test,downstairs test and tapping test under the monitor of integrated sensor,minute ventilation sensor and activity sensor respectirely.The rate-adaptive responses were tracked down every 15 seconds with remote heart rate monitor.The results were compared with the control group patients with normal sinal chronotropic function(n=15).Results In activity sensor group,pacing rates were significantly higher than the normal control group in downstairs test and tapping test.In minute ventilation sensor group,pacing rates increasement was significantly slower in the beginning of upstairs test.Pacing rates of integrated group were similar to the normal control group in upstairs and downstairs tests.Conclusion The rate-adaptive pacing response of dual sensor integrated with activity sensor and minute ventilation sensor,which is mostly close to the normal chronotropic response,better than that of single sensor.
7.Relationship Between Septal Myocardial Metabolism and Left Ventricular Mechanical Synchronization in Patients With Dilated Cardiomyopathy and Left Bundle Branch Block

Objective: To explore the relationship between septal myocardial metabolism and left ventricular mechanical synchronization in patients with dilated cardiomyopathy (DCM) and left bundle branch block (LBBB) by gated 18F-FDG myocardial metabolic imaging.
Methods: A total of 20 consecutive patients diagnosed for DCM with LBBB from 2010-10 to 2013-05 were enrolled, there were 11 male and 9 female at the mean age of (54±11) years. All patients received gated 18F-FDG myocardial metabolic PET imaging. TrueD software was used to determine the maximal standardized 18F-FDG uptake value (S-SUVmax) and the average standardized uptake value (S-SUVavg). QGS software was applied to conduct left ventricular phase analysis and to detect the cardiac function, left ventricular bandwidth (BW), standard deviation of bandwidth (SD), left ventricular end-diastolic volume (LVEDV), LVESV and LVEF. The relationship between 18F-FDG uptake in septal myocardium with the indexes of phase analysis and the indexes of cardiac function was analyzed.
Results: S-SUVmax and S-SUVavg were respectively negatively related to BW (r=-0.44, P<0.05 and r=-0.48, P<0.05);they were also respectively negatively related to SD (r=-0.57, P<0.01 and r=-0.51, P<0.05). While S-SUVmax and S-SUVavg were not really related to LVEDV, LVESV and LVEF, all P>0.05.
Conclusion: In patients of DCM with LBBB, reduced septal myocardial metabolism was closely related to left ventricular mechanical synchronization, gated 18F-FDG myocardial metabolic PET imaging may simultaneously detect both functions, which was important for prognostic evaluation and therapeutic monitoring in clinical practice;phase analysis.

10.Expression and identification of recombinant P-selectin and P-selectin glycoprotein ligand-1.

P-selectin, one of the membrane proteins, expresses on platelet and endothelia and interacts with P-selectin glycoprotein ligand-1 (PSGL-1) on leukocyte membrane. This interaction mediates leukocytes rolling on endothelial membrane and then induces leukocyte recruitment to the site of infection or tissue injury. In the present study, we constructed the recombinant wild type human P-selectin, its calcium-binding sites mutants and recombinant PSGL-1-globulin (PSGL-1-Rg). They expressed in Sf9 cells by using the baculovirus expression system and were purified by TalonTM metal or Protein A affinity chromatography. The results showed that the recombinant PSGL-1-Rg interacted with recombinant wild type P-selectin and two P-selectin mutants with 2 calcium-binding sites mutation respectively, but could not bind to the P-selectin mutant with all 4 calcium-binding sites mutation. Therefore, we verified the importance of P-selectin calcium-binding sites for its interaction with PSGL-1.
