AIM:To investigate the influence of tumor necrosis factor-? (TNF-?) induced apoptosis through knocking down Bax gene by RNA interference (RNAi) in human alveolar type II epithelial cells (A549). METHODS: A549 cells were cultured in vitro and divided into 4 groups according to RNAi treatment: control group,TNF-? treated group,Bax siRNA group and control siRNA group. Chemically synthesized small interfering RNA (siRNA) directed against human Bax gene was transfected into A549 cells by cationic liposome. 2.Experimental study of three kinds of intravenous stents with different materials and different types

Objective To select a kind of stent with better property and spread its clinical application. Methods Three kinds of stents including “Z” type stainless steel stent(SSS), mesh Ni Ti alloy stent(AS), and gild stent(GS) were placed into dogs′ vena cava. Angiography was performed 2 weeks, 4 weeks, 8 weeks, 6 months, and 9 months after stent placements (according to the grouping time), then the animals were sacrificed to get the veins with stents for histopathology examinations (including macro pathology, microscope and scanning electron microscope). Data of displacement rate, expand rate, patency rate of stents, patency rate of tributary veins, endothelial thickness, and course of endothelialization were acquired to indicate stent application property. Measurement data were analyzed by analysis of variance and q test. Enumeration data were analyzed by ? 2 test. Ranked data were analyzed by Ridit analysis. Results The displacement rates of SSS, AS, and GS were 13 3%, 13 3%, and 7 7%, respectively(? 2=0 281, P =0 869); The incomplete expanding rates of SSS, AS, and GS were 6 7%, 46 7%, and 0.0% respectively. There was statistical difference(? 2=12 194, P 0 05); The patency rates of SSS, AS, and GS were 93 3%, 66 7%, and 84 6%, respectively. There was no statistical difference among the rates(? 2 =3 649, P =0 161). Endothelialization reached about 100% 4 weeks after the stents placements. Desquamating of GS plating layer was found 4 weeks after stent placement, surface erosion of SSS wire and AS wire was not found in 9 months. Conclusion Z type stainless steel stent has better anticoagulative capability, anticorrosive capability, and biocompatibility, and it is now an ideal intravenous stent.

METHODS: A549 cells were cultured in vitro and divided into 4 groups according to RNAi treatment: control group, TNF-α treated group, Bax siRNA group and control siRNA group. The effect of RNAi was investigated by reverse transcription PCR, Western blotting and immunohistochemisty, and the rate of apoptosis was investigated by flow cytometry. RESULTS: Bax gene was knocked down effectively in Bax siRNA group (P<0.05). Apoptosis was induced by TNF-α in TNF-α treated group and control siRNA group. However, it was abolished in Bax siRNA group by the downregulation of Bax gene compared to TNF-α treated group and control siRNA group (P<0.05). CONCLUSION: This result suggests that Bax gene has a significant role of pro-apoptosis in A549 cells and knocking down Bax gene by RNAi can effectively inhibit TNF-α-induced apoptosis in A549 cells.
6.Dose-effect relationship of donor antigenic specificity CD4~+ CD25~+ Treg cells prolonging survival of rat kidney allograft

Objective To investigate the dose-effect relationship of donor antigenic specificity CD4~+ CD25~+ Treg cells prolonging the survival of rat kidney allograft.Methods Sixty allograft kid- ney transplantation animal models were established with SD rats as donors and Wistar rats as recipi- ents.CD4~+ CD25~+ T cells were sorted from Wistar rats'spleens by way of MACS and the phenotypes of donor antigenic specificity were induced in vitro.According to the quantities of prepared CD4~+ CD25~+ T cells injected through tail vein in kidney transplantation,models were divided into four ex- perimental groups:2?10~5(group I),5?10~5(groupⅡ),1?10~6(groupⅢ),2?10~6(groupⅣ).The models which had no injection served as control group(n=12).The survival of the transplanted kid- ney was observed.The levels of blood serum creatinine were determined and the histopathological changes in the transplanted kidney were observed at day 4,9 and 15.The results of histopathology were evaluated according to the standard of Banff Schema and the semi-quantitative scores were gained by way of Watanabe.The reaction indexes of receptor spleen cells to the donor antigens were tested by way of MTT at day 15.Results The mean survival time of transplanted kidneys was the highest in groupⅢ[(31.4?4.6)days]and lowest in the control group[(11.7?6.2)days].There was signif- icant difference between groupⅢand control group in levels of serum cre'atinine(P

Objective To investigate the expression of nerve growth factor(NGF) and slouble Fas(sFas) in children with viral myocarditis(VM),and the relationship with immunity,with the aim to provide evidence and basis for the diagnosis and treatment of VM.Methods The serum levels of NGF and sFas were detected by two-site enzyme-linked immunosorbent assy(ELISA) in 43 children with VM,and 20 normal heathy children as controls.Results The serum levels of both NGF and sFas in children with the acute stage(course in less than month and 1-2 months)and the procrastinate and chronic stages of VM were significantly higher than those of healthy children(P0.05).Conclusion The increased serum expression levels of NGF and sFas are related to the process of viral myocarditis,which confirms the relationship between the onset of VM and immune factors.
