1.EXPRESSION OF c-fos IN THE MEDULLA OBLONGATA AFTER CAROTID BARORECEPTOR ACTIVATION BY ELEVATED INTRASINUS PRESSURE AND ADENOSINE

Expression of c-fos protein in the medulla oblongata after baroreceptor activation by elevated intrasinus pressure (ISP) and perfusion of adenosine (Ado) was examined in 14 vascularly isolated carotid sinus perfusion rats. The results showed that Fos-like immunoreactive(FLI) neurons were distributed throughout nucleus tractus solitarius, area postrema, rostral ventrolateral medulla and nucleus raphe pallidus, and the number of FLI was increased with the elevation of ISP. Furthermore, perfusing the carotid sinus with Ado at a given ISP markedly increased the FLI in the above regions. From the results obtained, it is concluded that the c-fos expression in baroreflex pathway in medulla oblongata may be enhanced by elevated ISP and intrasinus perfusion of Ado, and Ado is capable of facilitating the baroreflex.
2.A study on the characteristics and influencing factors of heterosexual intercourse among MSM population

Objective To explore the heterosexual intercourse characteristics and its influencing factors among MSM population. Methods Voluntary counseling/testing and questionnaire survey were carried out at the bathhouses where MSM usually assemble. Results Among 342 MSM surveyed,52. 05% were married. 76 people( 22. 22%) were completely homosexual,while 266 people(77. 78%)were bisexual. 24(31. 57%)MSM who answered″ totally gay″ in self-assessment of sex orientation were married,significantly lower than those answered bisexual(57. 89%)(p<0. 05). 197 of participants had sex with women ,and 123 MSM had sex with women during the last six months,49 MSM of which used condoms at each heterosexual activity. MSM who were married or bisexual were easy to have heterosexual intercourse. Among the MSM surveyed,the HIV and syphilis infection rates were 11. 40% and 16. 67% respectively. There was no significant difference between the use of condoms and infection( both p>0. 05). In the recent six months,there was no significant difference in the use of condoms between infected( 43. 34%)and non-infected persons( 43. 34%)( p >0. 05). Conclusion Among MSM population,the prevalence of heterosexual intercourse is high,while condom use rate is relatively low. It is necessary to strengthen the behavioral intervention and promote condom use.
3.Establishment of osteoblast primary cilia model removed by chloral hyrate.

OBJECTIVETo establish osteoblast model, primary cilla model was removed by chloral hyrate, observe effects of osteoblast primary cilla moved on enhancing ALP staining and calcified nodules staining in electromagnetic field.
METHODSThree 3-day-old male SD rats weighed between 6 and 9 g were killed, cranial osteoblast was drawed and adherencing cultured respectively. Cells were subcultured and randomly divided into 4 groups until reach to fusion states. The four groups included chloral hydrate non-involved group (control group), 2 mM, 4 mM and 8 mM chloral hydrate group, and cultured in 37 °C, 5% CO2 incubator for 72 h. Morphology of primary cilla was observed by laser confocal scanning microscope, and incidence of osteoblast primary cilia was analyzed by Image-Pro Plus 6.0 software. Cells in the correct concentration group which can removed cillia most effectively were selected and divided into 3 groups, including control group (C), Electromagnetic fields group (EMFs), and EMFs with 4 mM chloral hydrate group. DMEM nutrient solution contained 10%FBS were added into three groups and cultured for 9 days and formation of ALP were observed by histochemical staining of alkaline phosphatase. After 12 days' cultivation, formation of mineralization nodes was observed by alizarin red staining.
RESULTSCompared with control group and 2mM chloral hydrate group,4 mM chloral hydrate group could effectively remove osteoblast primary cilla (P<0.01). Removal of osteoblast primary cilla could weaken the formation of ALP and mineralization nodes in osteoblast in EMFS. Compared with EMFs group, the area of ALP and mineralization nodes in EMFs with 4 mM chloral hydrate group were decreased obviously (P<0.01).
CONCLUSION4mM chloral hydrate could effectively remove osteoblast primary cilia. Primary cilla participate in EMFs promoting formation of ALP and mineralization nodes in osteoblast and provide new ideas for exploring mechanism of EMFs promoting osteoblast maturation and mineralization.

4.Ocular manifestations of Alport syndrome

·AIM: To analyze the clinical manifestation of Alport syndrome, especially the ocular features.·METHODS: The physical, ophthalmologic and audiologic examination results of thirty two patients with Alport syndrome were analyzed retrospectively.·RESULTS: Thirty (93.7%) patients had some family history. All patients had renal disease: eighteen(56.3%) patients with chronic renal failure, four(12.5%) patients with renal insufficiency, and the other ten(31.3%) patients with hematuria. Twenty (62.5%) patients had sensorineural deafness. Thirteen (40.6%) patients had ocular deformity, five(15.6%) patients had typical ocular changes: three patients with anterior lenticonus, and two patients with macular flecks.·CONCLUSION: Ocular anomalies are not requisite for the diagnosis of Alport syndrome. But its typical ocular features should be recognized by the ophthalmologists which supports the diagnosis.
5.Effects of MRI in the diagnosis and prognosis of cervical spinal cord injury without fracture or dislocation

BACKGROUND: In the elderly, the cervical vertebrae will appear with different degrees of spinal stenosis caused by various factors. Notably, the incidence of cervical spinal cord injury without fracture or dislocation is high, accounting for 3%-16% of cervical spinal cord injury.OBJECTIVE: To explore the efficacy of MRI in the diagnosis and prognosis of cervical spinal cord injury without fracture or dislocation, and to analyze the influencing factors for surgical effectiveness.METHODS: Fifty-one patients with cervical spinal cord injury without fracture or dislocation in Zibo Central Hospital from August 2014 to June 2016 were analyzed retrospectively. All the patients underwent anterior, posteror, or anterior-posterior fusion, respectively, and then the intraoperative ligament injury and MRI images were compared. The age, injury type, spinal cord injury type revealed on MRI, time from injury to hospital, signal length of spinal cord injury,American Spinal injury Association grade, degree of cervical stenosis, whether hormone used or not within 8 hours after injury, time from injury to surgery, and surgical approaches were analyzed based on univariate analysis and multivariate analysis by Logistic regression analysis.RESULTS AND CONCLUSION: (1) MRI showed more comprehensive damage of the spinal cord and intervertebral disc ligament complex than the intraoperative findings (χ2=5.966,P < 0.05), and the cervical spinal cord injury was related to the injury of the corresponding intervertebral disc ligament complex (χ2=1.53, P > 0.05). (2) Univariate analysis results showed that the spinal cord injury type revealed on MRI, signal length of spinal cord injury, American Spinal injury Association grade, degrees of cervical stenosis, whether hormone used or not within 8 hours after injury, and time from injury to surgery were related to the functional recovery of spinal cord (P < 0.01). (3) Multivariate analysis results indicated that the type revealed on MRI, signal length of spinal cord injury and degrees of cervical stenosis were significantly related to the functional recovery of spinal cord (P < 0.05). (4) These results suggest that MRI has a high tissue resolution, so it plays an important role in the diagnosis of cervical spinal cord injury without fracture or dislocation.Moreover, the degrees of spinal cord injury and spinal stenosis, as well as operation time are main factors for curative efficacy.

7.NADPH-diaphorase activity and Fos expression in brainstem nuclei involved in cardiovascular regulation following intracarotid injection of capsaicin

The present study was undertaken to define whether intracarotid injection of capsaicin induces Fos expression associated with the activation of NOS-containing neurons in brainstem nuclei by combining the immunocytochemical method for Fos with NADPH-d histochemical technique for NOS. The results obtained are as follows: (1) Intracarotid injection of capsaicin caused a significant increase of Fos-like immunoreactive neurons in area postrema (AP), nucleus tractus solitarius (NTS), paragigantocellularis lateralis (PGL) and locus coeruleus (LC), without influence upon the neurons of raphe nuclei (RN) and periaqueductal gray (PAG). (2) NO-containing neurons in PGL and NTS and the double-labeled neurons in PGL were also increased significantly following intracarotid injection of capsaicin. Small numbers of NO-containing neurons were found in LC, but there was no change in the number of NO-containing neurons in RN and PAG. No NADPH-d histochemical activity could be found in AP. (3) The above responses to capsaicin were significantly inhibited by pretreatment with either a capsaicin receptor antagonist ruthenium red or a NMDA receptor antagonist MK-801. The above results indicate that intracarotid injection of capsaicin may activate the neurons in brainstem nuclei involved in cardiovascular regulation, and that NO only plays an indirect role in the modulation of the responses of brainstem nuclei to capsaicin. These effects of capsaicin are mediated by capsaicin receptors with involvement of glutamate.
8.Protective effect of Quinacrine on striatum neurons from heat treatment injury.

AIMTo study the protective effect of Quinacrine(QA) on rat striatum neurons from the injury caused by heat environment treatment, to probe the relationship between cell membrane injury and cellular injury protection, and to seek the possibility of QA as a preventive agent to heat injury.
METHODSPrimary cultured striatum neurons from newborn rats were pretreated with QA at different concentration for 1 h, and then heat-treated at 43 degrees C for another 1 h. Cell necrosis was detected by Trypan blue staining, and apoptosis was evaluated through Activated Caspase-3 dye and TdT dye.
RESULTSHeat treatment effected the survival of striatum neurons and resulted in great number of cell death, which was mainly mediated by cell necrosis process. It was shown that treatment of QA itself had little effect on the survival of striatum neurons, while QA pretreatment decreased cellular necrosis caused by following heat treatment.
CONCLUSIONQA protects striatum neurons from heat environment injury at about 20 pmol/L, and the protection may mediated by reduction of necrosis.

9.CD133 in identifying perivascular niche of glioma stem cells

Objective To investigate the expressions of the putative brain tumor stem cell (BTSC) marker CD133, nestin and proliferating cell nuclear antigen (PCNA) in formalin-fixed and paraffin-embedded gliomas, and discuss the BTSC microenvironment: the composition, morphology and distribution of the niche. Methods The samples of 74 patients performed resection of the glioma in our hospital from January 2007 and October 2008 were chosen. According to WHO 2008 classification of nervous system tumors, they were assigned into low-grade glioma group (grade Ⅱ, 22) and high-grade glioma group(grade Ⅲ, 27 and grade Ⅳ,25). Immunohistochemistry was used to detect the expression of CD 133 in 74 cases of brain gliomas. Double immunofluorescence staining was employed to detect the co-expressions of CD133/nestin or CD133/PCNA. The percentages of the CD133+cells, CD133+ blood vessels and CD 133+ niches were calculated, and correlation analysis was also performed on their percentage and the pathological grading of the tumor. Results Accumulated CD 133+ cells grew in the niches. CD 133+ niches could be observed in all grade gliomas; low-grade group showed lower level expression of the CD 133+ niches, fewer proliferating cells, clearer boundary between the 2 neighboring niches and fewer surrounding blood vessels as compared with high-grade group. Besides CD113+/nestin+ BTSCs, some subsets as CD133+/nestin-cells and CD133-/PCNA+ cells were noted in the niches in the high-grade group. The percentages of CD 133+ cells, CD 133+ blood vessels and CD 133+ niches were different in different grades of gliomas and the higher the grade of gliomas was, the higher their expressions were. Positive correlation was observed between the percentage of CD 133+ niches and the percentage of CD133+ blood vessels (r=0.425, P=0.000). The expression of CD133+ niches in high-grade glioma were higher than that in low-grade tumors (F=5.324, P=0.002). Conclusion Glioma tissues have niche structures, which composed of CD133+/nestin+ BTSCs and some subsets. CD133+ blood vessels were playing a key role in maintaining the niche structure, and the expression of CD133+ niches is statistically different in different pathological grading tumors.
10.Effect of multiple micronutrients supplementation on anti-oxidative activity and oxidized DNA damage of lymphocytes in children.

OBJECTIVETo examine the effect of multiple micronutrients supplementation on anti-oxidative activity and decreasing oxidized DNA damage of lymphocytes in Chinese children.
METHODS82 healthy children in a rural areas, aged 9-11 years, were selected and randomized allocated into group receiving supplements and control group with each of them 41. 24-hour dietary recall was used to collect data on daily nutrient intakes of the research subjects. The subjects in the supplement group were given vitamin A (VA) 600 microg, beta-carotene (beta-C) 1.0 mg, vitamin E (VE) 100 mg, vitamin C (VC) 300 mg and Na2SeO3(Se) 200 microg in a tablet on daily base while those in the control group took a same-sized color placebo tablet. The trial lasted 8 weeks. 5 ml blood samples from each subject were taken during 7 to 9 o'clock in the morning. DNA damage of lymphocytes and levels of plasma VA, VE, VC, beta-C, Se, malondialdehyde (MDA), activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were then analyzed twice before and after the 8-week of trial.
RESULTSThe low intakes of VA, VC and Se only accounted for 50.6%, 65.6% and 67.3% of their recommended nutrient intake (RNI) respectively. After the trial, levels of plasma beta-C, VA, VE, VC and Se in the supplemented group increased by 13.4%, 32.8%, 11.5%, 46.9% and 24.6% respectively, compared with the control group, indicating that nutritional status regarding antioxidant nutrients had largely been improved. GSH-Px activity had a significant increase in the supplement group than before the supplement and in the control group (P < 0.01). GSH-Px before the trial (the 100.4 U/ml) also showed significant increase after the trial (161.7 U/ml) (P < 0.01). However, the values of SOD and MDA significantly decreased after the trial. Analysis of DNA damage indicated that there was no significant difference in the intrinsic damage of DNA (P > 0.05). Significant decreases of oxidized DNA damage induced by 5 micromol/L, 10 micromol/L and 25 micromol/L H2O2 were found more in peripheral lymphocytes of the supplemented group, than in pre-supplement and the control group after the trial (P < 0.01).
CONCLUSIONSupplementation of multiple micronutrients could effectively increase the levels of beta-C, VA, VE, VC and Se in plasma, and GSH-Px activity. In the meantime, MDA and oxidized DNA damage induced by a low level H2O2 decreased significantly after the trial. The reason accounted for the decrease of SOD activity after the trial needs to be further studied.
