1.Radiosensitization of artemisinin and its derivatives in tumors

Artemisinin and its derivative is a kind of efficient,quick and low toxicity of anti-malarial drug.In recent years,we find that artemisinin drugs can not only against malaria,but also have pharmacological effects of immunosuppression,antiviral and anticancer.A number of studies have confirmed that artemisinin and its derivatives have the radiosensitization effects in nasopharyngeal carcinoma,cervical cancer,lung cancer and glioma,and their mechanisms are related to decreasing Weel protein expression,increasing Cyclin B1 protein expression,blocking G2-M phase and cell apoptosis.
2.Effect of propofol pretreatment against hepatic ischemia-reperfusion injury on mitochondrial permeability transition pore in rats

Objective To investigate the protective effect of propotol against hepatic ischemiareperfusion injury in rats on mitochondrial permeability transition pore (MPTP) and the mechanism of GSK-3β.Methods Thirty SD rats were randomly assigned into five groups (n =6):sham operation group (S group),ischemia reperfusion group (I/R group),CsA pretreatment group (C group),propofol pretreatment group (P group),and propofol plus atractyloside pretreatment group (A + P group).Nauta liver ischemia-reperfusion rat model was used.Liver lobes were subjected to warm ischemia for 60min and then reperfusion for 120 min.In P group,propofol [12 mg/(kg · h)] was administered in the femoral vein for 30 min before ischemia until the end of reperfusion.In C group,CsA (2 mg/kg) was administered in the femoral vein for 20min before ischemia.In A + P group,20 μmol/kg of atractyloside was given through the femoral vein 10min before the injection of propofol.Rats were sacrificed at the end of reperfusion,and venous blood and hepatic tissue specimens from the same part of ischemia were obtained from different groups.Results Compared with S group,the AST and ALT levels were increased significantly,mitochondrial swelling were increased and mitochondrial membrane potential were decreased significantly in I/R group and A + P group.Casepase-3 were increased significantly and p-GSK3β Ser9 were decreased significantly in I/R group and A + P group.Compared with I/R group,the content of AST and ALT were decreased significantly,mitochondrial swelling were decreased and mitochondrial membrane potential were increased significantly,casepase-3 release were decreased significantly and p-GSK3β Ser9 were increased significantly in P group and C group.GSK-3β in each group displayed no significant difference.Conclusions Propofol can significantly reduce hepatic ischemia-reperfusion injury.The protective effect of propofol may be achieved via the inhibition of GSK-3β activation,increased p-GSK-3β Ser9 level,suppressing MPTP opening and decreasing hepatocytes apoptosis.
3.Isolation and identification of Neisseria gonorrhoeae strain from a balanoposthitis patient and drug resistance mechanism of the isolate

We isolated and identified the bacterial pathogen in a pyogenic balanoposthitis patient and investigated the drug resistance and its mechanism of the isolate.Urethral secretions and balanus pustule liquids were collected for microscopic examination after Gram-staining and detection of mycoplasma using Mycoplasma IST 2 kit.The two samples were inoculated on Columbia blood plate,N.gonorrhoeae selective plate and chromID Candida plate for isolation.The obtained colonies were identified by VITEK 2-compact automatic bacterial detection and analysis system.Moreover,PCR was performed to detect 16S rRNA gene of N.gonorrhoeae in the samples and colonies.KB method was applied for detecting susceptibility of five common antibiotics against the isolate.The β-lactamase and extended spectrum β-lactamase confirmatory tests were used to investigate the enzyme production of the isolate as well as drug resistance-associated tetM,TEM,mefA and ermF genes in the isolate were detected by PCR.Results showed that all the clinic samples showed negative for mycoplasma.All the isolating cultivation results of urethral secretions were negative while the balanus pustule liquids provided positive isolating cultivation in the blood and selective plates.The VITEK 2-compact system and 16S rRNA-PCR revealed that the isolated strain belongs to N.gonorrhoeae.The isolate can produce β-lactamases and resist to penicillin G,ciprofloxacin and tetracycline.The tetM,TEM,mefA and ermF genes could be found in the isolate's genome.The patient's balanoposthitis is caused by infection of N.gonorrhoeae.The multidrug resistance of Neisseria gonorrhoeae isolate is closely associated with its carried resistant genes.
4.Application of two dietary survey methods in type 2 diabetes patients

Objective To analyze and compare the differences in application of 24-hour dietary recall and dietary balance index (DBI) in dietary survey and evaluation of patients with type 2 diabetes mellitus,meanwhile investigate their nutrition status.Method This was a cross-sectional study.A total of 100 type 2 diabetes inpatients at the age of 19-59 were recruited from Shanghai Jiaotong University Affiliated Sixth People's Hospital from December 2013 to February 2014.They were surveyed and evaluated by 24-hour dietary recall and DBI respectively.Result The micronutrient intake in patients with type 2 diabetes was not sufficient.Compared with RNI,the intake of vitamin B1,B2,and calcium was less than 50%.The average of DBI lower bound score (DBI-LBS) of the 100 patients was 32.1±6.1,65%; the patients were in moderate or severe deficit of food intake.DBI higher bound score (DBI-HBS) was 4.4±2.8.No significant excess intake problem was found.Dietary quality distance was 36.4±6.9.Eighty-seven percent of them had a dietary patterns of mode B.Conclusion Dietary patterns in type 2 diabetes patients were not reasonable.Nutrition education and nutrition intervention for type 2 diabetes should be emphasized.The two methods can be used to evaluate dietary quality independently,but it would be better to evaluate the quality of the patients' diet using two dietary survey methods together.
5.Research progress of static magnetic field targeting drug delivery system in tumor diagnosis and therapy

Chemotherapy is one of the traditional tumors treatment solutions.Chemotherapy has the feature of tissue non-specificity,which can cause side effects on normal cells while inhibiting tumor cell growth.Magnetic targeting drug delivery system (MTDDS) employs biocompatible and stable magnetic nanoparticles (MNP) as drug carries to transport and accumulate anticancer drugs to the specific tumor tissues under the guidance of external magnetic field.This technology not only improves the efficiency of drug delivery and antitumor activity,but also reduces the drug dosage and side effects.The properties of drug-loaded MNPs and the applied external magnetic field are the main factors that affecting the MNPs targeting to the tumor tissues.The effectiveness of the targeted delivery of the drug-loaded magnetic nanoparticles mainly depends on the form and strength of the magnetic field at the target site.That is,whether there is sufficient strength to attract and retain NMPs,and to promote antitumor drug release at the tumor region.In this paper,the research progress of static magnetic field targeting drug delivery system in tumor diagnosis and therapy was summarized,which can provide some basic information for the relative scientific researches.
6.Distribution characteristics and diversity of virulence genes in clinical isolates of vancomycin-resistant enterococci

The aim of this study is to determine the diversity of virulence genes carried by different vancomycin-resistant enterococci (VRE),which will provides a basis for studying pathogenic mechanism of VRE.Microdilution-based drug sensitivity test was applied to detect the vancomycin resistance of 490 Enterococcus faecium isolates and 862 Enterococcus faecalis isolates in Zhejiang area.The seven virulence genes (ace,asa1,cylA,efaA,esp,gelE and hyl) in the isolates of VRE were detected by PCR.According to the results of drug sensitivity test,10% of the E.faecium isolates (49/490) and 0.8% of the E.faecalis (7/862) were identified as VRE.In the vancomycin-resistant E.faecium isolates,five isolates were negative for any of the target genes and the other 44 isolates were positive for asa1,esp,gelE and hyl genes alone,in which the esp (73.5%,36/49) and hyl (53.1%,26/49) were the predominant genes and single or double virulence genes acted as the major carrying models.Except for the hyl gene,the vancomycin-resistant E.faecalis isolates were positive for the other six pathogenic genes,and the isolates could carry 3-6 pathogenic genes.All the data indicate that E.faeciurn is the major species of VRE in the local area,and the carrying rate,types and models of virulence genes in the vancomycin-resistant E.faecium and E.faecalis isolates are obviously different.

8.Analysis of the relationship between different types of placental implantation after cesarean section and previous cesarean section

Objective To analyze the correlation between different types of placental implantation after cesarean section and previous cesarean section.Methods The data of 7 286 pregnant women with second pregnancy after cesarean section from January 2010 to October 2018 were retrospectively analyzed.Among them,842 cases had placental implantation.Five hundred and ninetyseven cases were adhesion type,204 cases were implantation type and 41 cases were penetration type.The relationship between the type of placental implantation in this pregnancy and emergency operation and postpartum hemorrhage during previous cesarean section and the time from this pregnancy was analyzed.Results The total incidence of placental implantation and the incidence of three types of placental implantation in the transfer operation group was higher than that of non-transfer operation group,total:27.4% (419/1528) vs.7.3% (423/5 758);adhesion type:18.0% (275/1 528) vs.5.6% (322/5 758);implantation type:7.9% (121/1 528) vs.1.4% (83/5 758);penetration type:1.5% (23/1 528)vs.0.3% (18/5 758),and there were significant differences (P<0.01).The total incidence of placental implantation and the incidence of three types of placental implantation in postpartum hemorrhage group was higher than that in the non-postpartum hemorrhage group,total:33.9% (74/218) vs.10.9% (768/7 068);adhesion type:22.5% (49/218) vs.7.8% (548/7 068);implantation type:9.6% (21/218) vs.2.6% (183/7 068);penetration type:1.8% (4/218) vs.0.5% (37/7 068),and there were significant differences (P<0.01).There was no statistical significance between the time of pregnancy and the incidence and total incidence of three types of placental implantation (P>0.05).Conclusions Placental implantation after cesarean section may be closely related to emergency operation and postpartum hemorrhage during the previous cesarean section.Pregnant women with second pregnancy after cesarean section should strengthen the management of pregnancy.
9.Research advances in hereditary epilepsy and precision drug therapy.

Epilepsy is a common nervous system disease. It has been found that the pathogenesis of epilepsy is associated mutations in various genes, including genes encoding voltage-dependent ion channel, genes encoding ligand-gated ion channel, and solute carrier family genes. Different types of epilepsy caused by different mutations have different responses to drugs, and therefore, diagnosis and medication guidance based on genes are new thoughts for developing therapies. With the application of next-generation sequencing technology, more and more genes will be determined, which helps to further study the pathogenic mechanism of mutant genes and provides a basis for precision drug therapy for epilepsy.

10.Effect of transcatheter arterial chemoembolization combined with radiofrequency ablation in the treatment of primary liver cancer and its influence on the levels of nuclear factor kappa B and p53

Objective To investigate the effect of transcatheter arterial chemoembolization combined with radiofrequency ablation in the treatment of primary hepatocellular carcinoma (HCC) and its influence on the levels of nuclear factor kappa B (NF-B) and p53.Methods 90 patients with primary HCC were selected,and they were randomly divided into control group (n =45) and observation group (n =45) according to the digital table.The control group received chemoembolization treatment,the observation group was given combined radiofrequency ablation treatment.The patients were continuously treated for 6 months.Immunohistochemical assay was used to detect the expression of NF-B and p53 in two groups.The clinical curative effect and influence on the expression of NF-B and p53 were compared between the two groups.Results After 6 months of treatment,the Karnofsky score of the observation group was (84.32 ± 12.31)points,which was higher than (64.32 ± 11.24) points of the control group (t =21.295,P < 0.05).After treatment for 6 months,the AFP level of the observation group was (121.23 ± 1.43) μg/L,which was lower than (189.44 ± 36.42) μg/L of the control group (t =19.853,P < 0.05).After treatment,the positive expression rates of NF-B and p53 of the observation group were 24.44% and 20.00%,respectively,which were lower than 40% and 33.33 % of the control group (x2 =6.597,5.784,all P < 0.05).The recurrence rate of the observation group was 15.56%,which was lower than 28.89% of the control group (x2 =6.126,P < 0.05).The 6-month and 12-month survival rates of the observation group were 86.67% and 82.22%,respectively,which were higher than 75.56% and 64.44% of the control group (x2 =4.927,7.192%,all P<0.05).Conclusion The effect of radiofrequency ablation combined with radiofrequency ablation for patients with primary liver cancer is satisfactory,which is worthy of popularization and application.
