1.EFFECTS OF GINSENOSIDES AND PANTOCRINE ON THE FUNCTION OF REPRODUCTIVE ENDOCRINE SYSTEM IN MALE RATS
Chinese Pharmacological Bulletin 1986;0(04):-
In vitro experiment, results showed the that the LH secretion of adenohypophysis incubated with Ginsenosides ( G ) and Pantoc-rine ( P ) was significantly increased, similar to the exciting effect of LRH which stimulats LH secretion. By addition of G or P and Indomethacin (an inhibitor of PGs synthesis) at the same time, however, the LH secretion was significantly decreased. It appears that PGs may be involved in the regulation of the exciting effect of G and P on LH secretion. The experiment in vivo also demonstrated that in healthy young male rats injected with P by S.c.,thetestosterone content in plasma had significant increase, that the binding capacity of testicular tissue to 125I-HCG (receptor function) did not change clearly.

Objective To construct the recombinant human metapneumovirus(hMPV) (defined as rhMPV NL/1/00 GFP) in vitro by reverse genetics technique. Methods BSR-T7 cells were transfected using LipofectAMINE 2000 with the full-length cDNA plasmid, and four major protein expressing plasmids, pCITE-N, pCITE-P, pCITE-M2.1 and pCITE-L. After 3 d, cells were subjected to one -80℃ freeze-thaw cycle to prepare lysates. The supernatant of lysate was used to inoculate Vero-E6 cells. After 1-4 d, cells were found for the obvious development of cytopathic effects under light microscope and green fluoroscopic signals under fluorescence microscope, and were observed up to 10 d. The supernatant were collected to de-tect virus titer. Viral RNA was extracted from the supernatant and reverse transcriptase polymerase chain re-action (RT-PCR) was used to amplify N, F and G genes of rescued virus. Results Cytopathic effects and green fluoroscopic signals was readily and obviously observed after 1-4 d post-inoculation in Vero-E6 cells, then cytopathic effects got worse and green fluoroscopic signals became stronger gradually up to 10 d. The ti-ters of the 1st, 5th, 10th,15th and 20th generation virus ranged from 105.0 to 106.5 TCID50/ml. Amplicons with size of 910 bp (N), 450 bp (F) and 980 bp (G) by RT-PCR were accordant with expectant. Nucleotide sequence analysis of above cDNA fragments showed 100% similarity with reported sequence of hMPV NI/1/00 strain. The recombinant virus was genetically constant and GFP-labeled after 20 passages in Vero-E6 cells. Conclusion Recombined hMPV was successfully rescued by reverse genetics technique. This study lays ground for exploring pathogenesis of hMPV infection and development of hMPV attenuated vac-cines.

OBJECTIVE To study multiple factors on bone graft healing of mandibular reconstruction. METHODS Patients were divided into two groups: healing group(31 cases) and healing delayed group(10 cases).By comparison between two groups, the factors on wound healing after bone grafting were studied. RESULTS In patient′s age and sex, bone graft type and size, and history of operation on bone graft done, there were no differences between two groups.Extraoral and intraoral passage was the important factor to two groups,P=0.000. CONCLUSIONS It is important for us to treat wound carefully while bone graft is carried with extraoral and intraoral passage.

Objective To analyze the outcomes and prognostic factors in patients with esophageal cancer after concurrent chemoradiotherapy. Methods A total of 135 patients with esophageal squamous cell carcinoma were enrolled in the clinical study from January 2008 to June 2015.The patients were treated with two-dimensional radiotherapy (56 patients) or three-dimensional radiotherapy (79 patients). The radiotherapy was delivered at a total dose of 60-64 Gy (1.8-2.0 Gy per fraction). The concurrent chemotherapy regimen consisted of fluorouracil plus cisplatin or paclitaxel plus cisplatin and was performed on days 1 and day 29 of radiotherapy. The Kaplan-Meier method was used to calculate overall survival (OS)and progression-free survival (PFS) rates, the log-rank test was used for survival difference analysis and univariate prognostic analysis, and the Cox model was used for multivariate prognostic analysis. Results The 1-,3-,and 5-year sample sizes were 96,31,16,respectively.The 1-,3-,and 5-year OS rates were 74.0%,39.0%,and 28.6%,respectively;the median OS time was 25 months. The 1-,3-,and 5-year PFS rates were 57.3%,27.3%,and 16.6%,respectively;the median PFS time was 15 months. The univariate analysis indicated that clinical stage, radiotherapy method, and M stage were prognostic factors for OS and PFS (P =0.006,0.000,and 0.032;P=0.017,0.004,and O.000).The multivariate analysis showed that clinical stage and radiotherapy method were independent prognostic factors for OS and PFS (P=0.006 and 0.000;P =0.033 and 0.023).Conclusions For non-surgical treatment of patients with esophageal cancer, concurrent chemoradiotherapy is a preferred strategy and has proven to be effective and tolerable.

ObjectiveTo evaluate the effectiveness and safety of high frequency electronic associating stents insertion and subsequent radiotherapy guided by fiberoptic bronchoscope in treating tracheobronchial stenosis caused by lung and esophagus carcinoma.MethodsFifty-two patients which were ascertained with serious stenosis in trachea or bronchi by fiberoptic bronchoscope were subjected to the study.Fiberoptic bronchoscope was used to investigate the location, extent, blood supply and the degree of tracheobronchial stenosis before the study. After the focus in the airways was cleared up with high frequency electronic, electrocoagulation and snare under the guide of the euthyphoria of fiberoptic bronchoscope,Ni-Ti memory alloy stents were placed into the stenosis airways. The 52 patients were divided into two groups with 26 cases each:the stent group and the stent associating radiotherapy group(radiotherapy group). The later group accepted subsequent radiotherapy after the therapy. The following indexes of the two groups were investigated:short-time clinical effect, dyspnea index class, rate and average time of airway restenosis, life span and survival rate.ResultsAfter therapy, the 52 patients had obvious improvements in dyspnea and the diameter of the stenosis airways. The short-time total effective rate was 100.0% (52/52),but there was not significant difference between the two groups(P> 0.05 ).After therapy, there was significant difference in two groups in dyspnea index class(P < 0.01 ).Although there was not significant difference in the rate of airway restenosis between the two groups(P>0.05),the average time of airway restenosis in the radiotherapy group was obviously longer than that in the stent group(P< 0.01 ).The survival rate of the radiotherapy group in the 6th and 9th month after therapy was higher than that in the stent group (P < 0.05),but there was no significantdifferenee in the 3rd and 12th month (P >0.05).ConclusionHigh frequency electronic associating stents insertion and subsequent radiotherapy guided by fiberoptic bronchoscope is an effective and safe treatment for the patients suffering tracheobronchial stenosis caused by lung and esophagus carcinoma.

Objective To investigate the expression and effect of secondary lymphoid tissue chemokine(SLC)in experimental ulcerative colitis(UC)in rats. Methods Thirty Spague-Dawley rats were divided into control group and UC group.SLC expression in colon tissues was detected by RT-PCR and immunohistochemistry, respectively. Results The transcriptional level of SLC in UC tissues was significantly higher than that in the control group(0.846?0.07 vs 0.312?0.12,P<0.01).The positive expression of SLC was concentrated mainly on submucosa, and the positive rate of SLC protein expression in UC group significantly higher than that in control group(P<0.01).Conclusion SLC overexpression could contribute to the pathological processes in UC rats, thus SLC may be an ideal ther- apeutic target for UC.
