1.Research progress of novel tyrosine kinase inhibitor AC220 in the treatment of acute myeloid leukemia

Acute myeloid leukemia (AML) is a highly heterogenous disease with multiple signaling pathways contributing to its pathogenesis.A key driver of AML is the mutation of FMS-like tyrosine kinase receptor-3 (FLT3) gene.Mutations in FLT3,primarily the FLT3-internal tandem duplication (FLT3-ITD),are associated with decreased progression-free and overall survival.Due to the importance of FLT3-ITD and its pathway in the prognosis of patients with AML,it has stimulated efforts to develop therapeutic inhibitors of FLT3.Although the FLT3 inhibitors show a certain degree of antileukemia activity,their clinical effect has some limitations as a single drug.Now,Quizartinib (AC220) is the potent second generation FLT3 selective inhibitors.Based on the resistance mechanism for FLT3 inhibitom,more and more scholars has been studying the inhibition of FLT3-ITD+ cell lines and primary leukemia cells by AC220 alone and in combination with cytotoxic chemotherapies/protein kinase inhibitors as well.However,the clinical effects of a variety of combination plan remain to be further investigated.
2.Application of Enterprise Master Patient Index in Hospital Clinical Information Integration

Taking Children's Hospital of Fudan University as an example, the paper establishes a patient identity matching model based on the historical treatment data according to features of pediatric patients.It also sets up a uniform indexing mechanism of the his-torical treatment and hospitalization records of the patients by improving the existing business system, providing support patient identity recognition and laying a foundation for establishment of the clinical data center in future.

Replacement therapy of stem cells transplantation represents a potential treatment for neural retinal diseases.Despite the encouraging results in laboratory,the clinical application of cells replacement therapy is still difficult because the limitation of seed cells,immunologic rejection,oncogenicity and ethical problems,etc.Recent breakthrough in somatic reprogramming provides a promising solution overcoming these obstacles.Further researches on virus-free reprogramming will make the clinical application of stem cell replacement therapy possible.

Trauma has been one of the public haza rds in the modern society.About 1/2t o 2/3of modern traumas involve the musculoskeletal system.The priority in an em ergency care of a severely injuried p a-tient is to save his life and preserve major functions of his body as much as possible.To minimize the rate of mortality or disability,it is of prime importance to start timely emergency resuscitation process,control of life -threatening conditions(such as hypovolemic shock),hemostasis,adequate ventilation,restoring normal perfu-sion,and prompt diagnosis and skill ful evaluation of the general and local significant risks so as to adopt fu rther interventions,especially for multiply injured patients.After satisfactory recovery of vital signs and stable main-tenance of adequate perfusion press ure,surgical procedures,such as debridement,fracture fixation,repair of soft tissue and management of associated lesions are advisable at an early sta ge.[
6.Study on the Heredity Stability of a Recombinant Plasmid pMal-2X in Escherichia coli

The strain of TB1 from the original stock which harboring a recombinant plasmid pMal-2X including apc gene from cyanophyceae had been subcultured serialyto 100~(th) generation in Amp~(+) plate. The growth characteristics and the morphology of the cultures did not show differences among the generations. The chromatogram of the plasmids in different generations digested by EcoRⅠ restriction enzymes were same, as well as DNA sequences showed no difference among generations. In addition to that, origin strain and 10~(th), 20~(th), 50~(th) and 100~(th) generation strain had a similar rAPC expression level. The immunology activity of the rAPC and SDS-PAGE of the proteins expressed in different generations were also same. From the above results, it is indicated that recombinant stain P1 has a high hereditable stability.
7.Laparoscopic transcystic common bile duct exploration

Objective To investigate the feasibility of transcystic removal of common bile duct stones during laparoscopic cholecystectomy. Methods Clinical data of 18 cases of transcystic cholangiography during laparoscopic cholecystectomy from January 2003 to July 2004 were retrospectively reviewed. Before the resection of the gallbladder, the cystic duct was opened to insert a catheter for cholangiography. After the common bile duct stones were detected, a wire basket was introduced transcystically to remove the stones. Results Transcystic cholangiography was conducted in 18 cases, 11 of which were detected as having common bile duct stones. Of the 11 cases, laparoscopic transcystic common bile duct exploration was successfully completed in 6 cases and failed in 2 cases with stones significantly larger than the diameter of the bile duct, 2 cases of failure of transcystic insertion of catheter, and 1 case with stones embedded in the hepatopancreatic ampulla. Follow-up with B-ultrasonography for 6~18 months in the 6 cases revealed no residual stones,biliary stricture,or bile duct dilatation. Conclusions Laparoscopic transcystic common bile duct exploration (LTCBDE) provides an effective option for the diagnosis and treatment of secondary common bile duct stones during laparoscopic cholecystectomy without opening the common bile duct.
8.Therapeutic drug monitoring in patients using cyclospone A following renal transplantation

OBJECTIVE: To evaluate the value as well as correlation between blood drug level and blood routine results in patients using cyclospone A (CsA) following renal transplantation via analyzing varies of immunosuppressive agents used in renal transplantation. METHODS: Retrieval-related contents were collected by computer from Chinese Journal Full-Text Database with of "renal transplantation, CsA, and therapeutic drug monitoring". Simultaneously, data of patients who received CsA treatment after renal transplantation in the Department of Laboratory, Liaoning Provincial Electrical Hospital were selected. All the literature was retried and analyzed. RESULTS: A total of 717 patients were included in the final analysis. It demonstrated that 6 sorts of immunosuppressive agents, such as mizoribine, sirolimus, zenapax, mycophenolate, and basiliximab play an important role in immune suppression after organ transplantation. However, there still exist problems need to be further explored. CsA was more suitable for clinical application with lower price and good curative effect, which worked better combined with other immunosuppressive agents. CONCLUSION: A best curative effect of CsA can be achieved by timely adjusting therapeutic regimen under the patient's condition. Meantime, a high efficiency and low toxicity drug with specific tolerance is the ultimate goal of immunosuppressive agents.
9.Utilization of Oral Antihypertensive Drugs in Emergency Department of Our Hospital

OBJECTIVE: To investigate the utilization of oral antihypertensive drugs in the emergency department of our hospital.METHODS: By using drug utilization index,the oral antihypertensive drugs used in the emergency department of our hospital from Mar to Aug in 2006 were analyzed statistically in respect of the drug categories,DDDs,DDC etc.RESULTS: Leading the list in terms of DDDs and DDC were calcium antagonists,ACEI and compound preparations.CONCLUSION: The utilization of oral antihyperetensive drugs in the emergency department of our hospital was basically in line with that of the currently common antihypertensive drugs,however,the application categories should be extended,esp.for those with prolonged action.Furthermore,individualized medication should be advocated and great importance should be attached to ADR.
10.EVALUATION OF LH, FSH AND PRL RIA IN SECONDARY AMENORRHEA

RIA of LH and FSH in 437 cases and PRLin 287 cases of secondary amenorrhea were performed. According to the results they can be divided into 5 groups: hypergonadotropic amenorrhea 132 cases (30.2%), hypogonadotropic amenorrhea 44 cases (10.1%), normogonadotropic amenorrhea 40 cases (9.1%), amenorrhra with increased LH/FSH ratio 94 cases (21.5%) and hyperprolactinemic amenorrhea 127 cases of which 117 cases (92.1%) had galactorrhea. The clinical features, laparoscopy findings and results of treatment were presented.
