1.Construction of X IAP-3′UTR-luciferase reporter vector and its activity analysis

Objective To construct the recombinant X-linked inhibitor of apoptosis protein(XIAP) gene 3′untranslational region (3′UTR)-luciferase reporter vector ,and analyze the microRNA(miRNA) which possibly regulate the expression of XIAP gene . Methods Polymerase chain reaction (PCR) was employed to amplify X IA P-3′UTR sequences from human cDNA ,in which luciferase reporter vector pGL3-Ctrl was inserted ,and the recombinant vector pGL3-Ctrl/XIAP was gained .Target Scan 6 .2 soft-ware was adopted to predict the miRNA which possibly combined with the X IA P-3′UTR .pGL3-Ctrl/XIAP recombinant plasmids and the miRNA were co-transfected into A549 cells ,and the X IA P-3′UTR-luciferase activity was measured .Results Confirmed by digestion and DNA sequencing ,the X IA P-3′UTR-luciferase reporter recombinant was successfully constructed .Prediction of miRNA target sites indicated that X IA P gene may be the target of miR-200b ,miR-200c and miR-429 .Compared with miRNA mim-ic ctrl group ,miR-200b ,miR-200c and miR-429 significantly reduced the luciferase activity of pGL 3-Ctrl/XIAP with statistically significant difference(P<0 .05) .Conclusion X IA P-3′UTR-luciferase reporter vector is successfully constructed .miR-200b ,miR-200c and miR-429 can obviously decrease the luciferase activity .
2.Emotional experience and processing characteristics of long-term plateau immigrant undergraduates

Objective:To investigate the subjective experience and processing characteristics of positive and negative emotions in undergraduates who have immigrated to the plateau for a long time.Methods:SPSS 22.0 was used for data analysis.Using independent sample t-test to compare various data from the plateau group(58 undergraduates who had moved to an altitude of 3 658 m for over 2 years after reaching adulthood) with the plain group (58 undergraduates who had never been to the plateau), including the state anxiety inventory(SAI), self-rating depression scale(SDS), positive and negative affect schedule (PANAS), and the affective valency and arousal on the positive, neutral, and negative emotional pictures.And χ 2 test was conducted for the positive detection rates of state anxiety and depression of the two groups.Then, Pearson correlation analysis was performed on SAI, SDS and PANAS scores. Results:The positive detection rates(state-anxiety: 67.24%, depression: 56.90%) and scores of state anxiety(44.45±8.57) and depression(50.12±10.11) in the plateau group were all higher than those in the plain group(46.55%, 32.76%, (40.19±9.58), (44.64±9.17) respectively, χ 2=5.06, 8.12, t=2.52, 3.06, all P<0.05). In addition, positive affect scores of the plateau group (28.16±5.39) tended to be lower than those of the plain group (30.28±6.51), but the difference was not statistically significant( t=-1.91, P=0.059). SAI, SDS and PANAS scores were moderately correlated each other( r= 0.69, -0.52, 0.67, -0.62, 0.53, all P<0.01). Positive affect was mildly negatively correlated with negative affect( r=-0.27, P<0.01). The correlation between positive emotion and depression( r=-0.62) was greater than that between positive emotion and state anxiety( r=-0.52, t=2.57, P<0.05). The correlation between negative emotion and state anxiety( r=0.67) was greater than that between negative emotion and depression( r=0.53, t=1.71, P<0.05). The valency score of neutral images in the plateau group (3.06±0.20) was higher than that in the plain group ((2.99±0.20), t=2.01, P=0.047). Conclusion:The long-term plateau immigrant undergraduates are prone to state anxiety and depression, with depression being the primary and anxiety secondary, and exhibit a positivity bias to neutral stimulus processing.
3.The Concept of Interoception in Modern Psychology and Its Clinical Applications from the Perspective of Traditional Chinese Medicine

This study critically examines the current status of interoception research in modern psychology, taking into account perspective of traditional Chinese medicine (TCM), encompassing both foundational studies and clinical applications. The investigation highlights a deep interconnection between psychosomatic theories in TCM, such as the TCM emotions and viscera theory and the view of body-spirit syncretism, and the concept of interoception at the cognitive level. Moreover, philosophical principles, particularly the “Zhi Yi Lun” in TCM, align naturally with the three-dimensional interoception model. Importantly, this comparative analysis emphasizes that Western and Chinese medicine place different emphasis on interoception research with the former focusing on diagnosis and the latter on treatment. These differing approaches complement each other and provide opportunities for cross-fertilization. At the practical level of interoception application, “five-state personality classification” demonstrates promising potential for further clinical application. In addition, TCM has valuable insights for contemporary medical practices in terms of emotional regulation. TCM methods for emotional regulation, known as “Daoyin”, provide important knowledge in this area. One concept developed by TCM for clinical intervention targeting interoception is “Tiao Qi” (regulating qi). This concept is used during diagnosis and treatment in TCM and plays a crucial role in managing interoceptive states. This paper aims to explain the cognitive and applied aspects of interoception in TCM, offering a perspective for future research and clinical application. By integrating modern psychology's psychosomatic medicine model, TCM can enhance its relevance and efficacy in contemporary medical contexts. This integration promotes a balanced relationship between the mind and body. TCM researchers have a responsibility to explore the inherent strengths and practical value of TCM in interoception research, and contribute to the advancement of TCM.
4.MiR-503 regulates cisplatin resistance of human gastric cancer cell lines by targeting IGF1R and BCL2.

BACKGROUNDStudies have shown that the drug resistance of gastric cancer cells can be modulated by abnormal expression of microRNAs (miRNAs). We investigated the role of miR-503 in the development of cisplatin resistance in human gastric cancer cell lines.
METHODSMiR-503 expression was measured by quantitative real-time PCR. MTT (3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide) and clonogenic assays were used to examine changes in cell viability and the drug resistance phenotype of cancer cells associated with upregulation or downregulation of the miRNA. A dual-luciferase activity assay was used to verify target genes of miR-503. Immunohistochemistry, Western blotting analysis, and a flow cytometric apoptosis assay were used to elucidate the mechanism by which miR-503 modulates drug resistance in cancer cells.
RESULTSMiR-503 was significantly downregulated in gastric cancer tissues and several gastric cancer cell lines. Additionally, downregulation of miR-503 in the cisplatin (DDP)-resistant gastric cancer cell line SGC7901/DDP was concurrent with the upregulation of insulin-like growth factor-1 receptor (IGF1R) and B-cell lymphoma 2 (BCL2) expression compared with the parental SGC7901 cell line. An in vitro drug sensitivity assay showed that overexpression of miR-503 sensitized SGC7901/DDP cells to cisplatin. The luciferase activity of reporters driven by IGF1R and BCL2 3'-untranslated regions in SGC7901/DDP cells suggested that IGF1R and BCL2 were both direct target genes of miR-503. Enforced miR-503 expression in SGC7901/DDP cells reduced expression of the target proteins, inhibited proliferation, and sensitized the cells to DDP-induced apoptosis.
CONCLUSIONOur findings suggest that hsa-miR-503 modulates cisplatin resistance of human gastric cancer cells at least in part by targeting IGF1R and BCL2.
