1.A STUDY ON THE MAIN PHARNACODYNAMICS OF KUNTAIAN

The pharmacoclynamics of Kuntaian was studied. The results showed that it exerts an inhibiting effect on normal uterine smooth muscle in a dosedependent manner. It can counteract the violent uterine contraction induced by pituitrin, promote the growth of uterus of young mice, dilate the capilaries and accelerate blood circulation. It also exerts analgesic antiphlogistic antioneotie effects, suggesting that this drug may be applicable to the treatment of dysmanorrhea, irregular menstruation and pelvic infection.
2.The incidence of the main adverse events of thoracic/abdominal aortic coated graft: a systematic review and meta-analysis

Objective The purpose of this study was to review all published reports on thoracic/ abdominal stent graft and investigate the incidence rates of six main adverse events(MAEs):myocardial infarction,paraplegia,renal failure,respiratory failure,stroke and all-cause mortality.Methods Electronic databases(PubMed、Embase、OVID、ProQuest、Elsevier and The Cochrane Library) were searched from inception through May 2014 to identify studies that assessed the safety of thoracic/abdominal aortic coated stents on MAEs.The incidence rates with 95% confidence intervals(95% CIs) were derived using a random effects model,considering the heterogeneity between the included studies.Subgroup and sensitivity analyses were applied to explore heterogeneity.Results A total of 152 studies were included in the analysis with 264 arms.For thoracic stent graft,meta-analysis yielded a combined estimated incidence rates of 12.2%,and the most common MAEs was all-cause mortality (8.1%),followed by respiratory failure (6.5 %).For abdominal stent graft,meta-analysis yielded a combined estimated incidence rates of 4.6%,and the most common MAEs was all-cause mortality(4.0%),followed by myocardial infarction(2.1%).For thoracic and abdominal stent graft,subgroup analysis stratified by age and proportion of males indicated that middle-aged and females have a higher incidence rates of all-cause mortality.Besides,subgroup analysis stratified by follow-up time indicated that the longer follow-up time,the higher incidence rate of all-cause mortality.Conclusion The current evidence indicates that the incidence of MAEs of thoracic and abdominal stent graft is high,and we should pay more attention to the patients and follow up them as long as possible.
3.Comparison of fluor-mica glass-ceramics made by melt-casting and sintering methods

Objective: To study the effects of the melt casting and the sintering methods on the machinability and other characteristics of K_2O-MgO-MgF_2-SiO_2 glass-ceramics. Methods: X-ray diffraction(XRD) and scanning electron microscopy (SEM)were used to examine the main crystal and the microstructure of the glass ceramics of the same composition which were made by melt casting and sintering methods, respectively. The transmissivity, flexural strength and brittleness index were observed respectively. Results: The glass ceramics made by two different methods have the same crystal, KMg_(2.5)Si_4O_(10)F_2, while the specimen prepared by the sintering method had lower transmissivity,better machinability and mechanical properties due to the higher mica volume percent. Conclusion; For production of fluoro-mica glass ceramics, sintering method has superiority compared to melt-casting method.
4.Variations of basic glomerular filtration function in experimental rabbits with partial unilateral ureteral ;obstruction

Objective To investigate the variations of glomerular function in rabbits with partial unilateral ureteral obstruction (PUUO). Methods Thirty healty adult Japanese White Rabbits were randomly divided into the control group (6 rabbits) and the obstruction group (24 rabbits). The right PUUO model was established according to Cheng′s method. According to the PUUO duration, they were eaqually divided into four subgroups:the 7 day PUUO group, the 14 d PUUO group, the 28 d PUUO group and the 56 d PUUO group. The dynamic renal imaging was performed on all rabbits, and the glomerular filtration rate (GFR) was also calculated. The correlation between the renal function and PUUO duration was analyzed , and functional variations in the healthy kidneys were observed. Results (1)To the control group, no significant difference in the renal function was found between the left and the right kidneys (t = -0.145, P = 0.894). However, to the PUUO group, the right renal GFRs were significantly lower than the left renal GFRs (P < 0.05). (2) To the right renal GFR, significant difference was observed between the 7 day PUUO group and the control group (t = 5.066, P = 0.001),but not among the 7 day PUUO group, the 14 day PUUO group, the 28 day and the 56 day PUUO group (P > 0.05), (3)No significant differences were found in the left renal GFRs among the five groups. Conclusions The function of PUUO kidneys is characterized by an obvious decrease in the early stage, followed with a steady state. The basic function of the opposite healthy kidney does not present the compensation phenomenon.
5.Transplantation of neonatal cardiomyocytes plus fibrin sealant restores myocardial function in a rat model of myocardial infarction.

BACKGROUNDMost cardiac regenerative approaches can restore injured heart muscles. In this study, we investigated if fibrin sealant could help neonatal cardiomyocytes restore myocardial function in a rat model of myocardial infarction.
METHODSThe left anterior descending artery in adult female Sprague-Dawley (SD) rats was ligated to make a myocardial infarction model. Neonatal ventricular cardiomyocytes from one-day male SD rats were isolated, labeled and cultured. The cells were injected into the infarcted area three weeks later. The animals were randomized into four recipient groups: (1) cardiomyocytes plus fibrin sealant (group CF, n = 10); (2) cardiomyocytes alone (group C, n = 10); (3) fibrin sealant recipients alone (group F, n = 10); (4) control group (n = 10). Four weeks after transplantation, echocardiography and Langerdoff model were used to assess heart function. Immunohistochemical staining and polymerase chain reaction (PCR) were performed to track the implanted cardiomyocytes and detect the sex-determining region Y gene on Y chromosome.
RESULTSEchocardiography showed the fraction shortening (FS) in groups CF, C, F and control group was (27.80 +/- 6.32)%, (22.29 +/- 4.54)%, (19.24 +/- 6.29)% and (20.36 +/- 3.29)% respectively with statistically significant differences in group CF compared with the other groups (P < 0.05). The Langendoff model revealed that the left ventricular development of peak pressure (LVDPmax, mmHg) in groups CF, C, F and control group was 104.81 +/- 17.05, 80.97 +/- 21.60, 72.07 +/- 26.17 and 71.42 +/- 17.55 respectively with statistically significant differences in group CF compared with the other groups (P < 0.05). Pathological examination and PCR indicated that transplanted cardiomyocytes in group CF survived better than those in the other groups.
CONCLUSIONTransplanted neonatal cardiomyocytes plus fibrin sealant can survive in myocardial infarctioned area and improve heart function greatly in rat models.

7.Expression of midkine and vascular endothelial growth factor in bone marrow of patients with multiple myeloma and its significance.

The purpose of this study was to explore the expressions of midkine (MK) and vascular endothelial growth factor (VEGF) in multiple myeloma (MM), and to evaluate their relation with angiogenesis and prognosis. The expression levels of MK and VEGF in bone marrow mononuclear cells of 31 MM patients in different stages and 20 controls were detected by real-time fluorescent quantitative RT-PCR. The results showed that the MM patients had significantly higher MK and VEGF expression level than control group (p < 0.05, p < 0.01), and there was a linear relationship between MK and VEGF (r = 0.692, p < 0.01); The expression levels of MK and VEGF in stage III was significantly higher than those in stage I and stage II (p < 0.05, p < 0.01), but there was no difference between stage I and stage II (p > 0.05); MK and VEGF levels were significantly decreased in MM patients after treatment than those before treatment (p < 0.05, p < 0.01). It is concluded that the high expression of MK and VEGF is correlated with angiogenesis and prognosis of MM, and there is synergistic effect between MK and VEGF. It is supposed that the monitoring MK and VEGF expression levels may contribute to guide the treatment and estimate prognosis for MM.

8.Significance of color Doppler ultrasonography in therapy of tuberculous epididymitis

Objective To study the clinical value of color Doppler ultrasonography in typing tuberculous epididymitis.Methods The appearances of color Doppler ultrasound and the findings on operation were analysed in 33 patients with tuberculous epididymitis.Results Of the 33 patients,epididymis appeared as diffusely and heterogeneously enlarged lesions with increased flow in 2 cases,appeared as nodular lesions in 13 cases including nodi with echofree space in 3 cases, nodi with high-level echo patches in 3 cases, and low echo-level nodi in 7 cases. Multiple lesions in scrotum were detected in 17 cases, of whom epididymis up to 11 cases appeared as diffusely enlarged heterogeneous lesions with flow increased.The sonographic appearancs of tuberculous epididymitis could be divided into 3 types:diffusion type, nodus type and complicated type. Nodus type included 3 subtypes: purulence type, calcification type, and cheese type.The accuracy rate of ultrsound diagnosis was 87.9%.Conclusions Testis is easy to be involved when epididymitis appears as diffusion type, so surgical treatments should be early.Purulence type and complicated type need surgical treatments while calcification type does not. Antituberculous drug treatments can be tried before surgical treatments in cheese type.Sonography of urinary system is helpful for the diagnosis of asymptomatic tuberculosis in urinary system when tuberculous epididymitis is first suspected on sonography.
9.Clinical characteristics and efficacy of anti-Aspergillus therapy in patients with hematological malignancies and invasive aspergillosis.

This study was aimed to analyze the clinical features, anti-fungal therapeutic efficacy and safety in hematological malignancy patients with invasive aspergillosis (IA) after hematopoietic stem cell transplantation (HSCT) or chemotherapy. The patients with hematological malignancies received chemotherapy or HSCT were analyzed retrospectively, then the clinical characteristics and diagnosis were analyzed according to the diagnostic criteria for IA. The efficacy and safety of anti- Aspergillus therapy, and the factors influencing therapeutic response were evaluated. The results showed that out of 30 cases with IA, 2 were proven, 19 were probable, 9 were possible, and 19 were diagnosed after HSCT, most in the late period after-HSCT (> 40 d). 8 cases received fluconazol only, 6 received caspofungin only, 7 received combined therapy. The efficacy and time interval from the first day of treatment to successful response (TTR) were 87.5%, 50% and 85.7% and 38, 20 and 36 days, respectively. Combined therapy is better than single drug treatment (p < 0.05) while the TTR was not significantly different between them. The factors influencing the therapeutic efficacy were as follows: age, HSCT, GVHD and CMV, previous IFI and so on (p < 0.05). All the anti- Aspergillus medicines resulted in some injury of hepatic and renal function. However, there were no significant difference between the drugs or between combination and single drug therapy (p > 0.05). It is concluded that IA is also the major and severe complication in the patients with hematological malignancies or received HSCT. Combined therapy for anti- aspergillus is better than single drug in efficacy and safety, without increasing the adverse drug reactions for hepatic and renal function. The efficacy of anti- aspergillus may be related to age, HSCT, GVHD and CMV, previous IFI and so on.

10.Cytotoxic effect of cisplatin with proteasome inhibitor on osteosarcoma cells.

OBJECTIVETo observe the cytotoxic effect of cisplatin with proteasome inhibitor on osteosarcoma cells.
METHODSCell survival was tested by MTT, apoptotic morphology was observed by electron microscopy, apoptotic rate was analyzed by flow cytometry, the transcription level of excision repair cross complementation-1 (ERCC-1) was tested by reverse transcription polymerase reaction.
RESULTSCompared with cells treated with cisplatin alone, cells treated with cisplatin and proteasome inhibitor showed a decreased survival rate, more typical apoptotic morphology, higher apoptotic rate [(14.37 +/-2.37)% vs. (50.93 +/-4.84)%, P<0.01)], and lower transcription level of excision repair cross complementation-1.
CONCLUSIONProteasome inhibitor could increase the cytotoxic effect of cisplatin on osteosarcoma cells and promote cisplatin-induced osteosarcoma apoptosis. These effects may be associated with the decreased transcription of excision repair cross complementation-1.
