1.Stable expression of programmed cell death 4 gene in glioma cell line and its influence on proliferation of tumor cells

Objective:To establish a glioma cell line U251 stably expressing programmed cell death 4(PDCD4)gene,and to observe the influence of exogenous PDCD4 gene on the proliferation and cell cycle of U251 cells.Methods:Recombinant eukaryotic expression vector pEGFP-PDCD4 was transfected into human glioma cell line U251 by Lipofectamine 2000,and the U251 cells stably expressing PDCD4 were established by G418 selection.Reverse transcription polymerase chain reaction(RT-PCR)and Western blotting were employed to detect the expression of PDCD4 mRNA and protein.Furthermore,cell proliferation and colony forming ability were determined by cell counting and colony formation assay;the cell cycle was detected by FACS.Results:High expression of PDCD4 mRNA and protein was observed in U251 cells transfected with pEGFP-PDCD4,whereas no PDCD4 mRNA and protein expression was detected in the non-transfected and vector-transfected cells.Further,cells transfected with pEGFP-PDCD4 grew more slowly and had lower colony formation rate than cells of the other two control groups(P
2.Artificial Abortion Has Important Effect on the Development of Adenomyosis

3.Design and clinical application of modified fixed Maryland Bridge for restoration of geriatric anterior dentition defects

Objective The anterior dentition defects of geriatric patients were restored with modified fixed Maryland Bridge.The clinical design,tooth preparation,fabrication of prosthesis and clinical indication were summarized.Method 24 cases of geriatric patients with anterior dentition defects were restored with modified fixed Maryland Bridge.All selected cases were accompanied with different degree of loosening of neighboring teeth,gingival atrophy and exposure of root.The dentition defects were successfully restored and followed up for 3 to 5 years.Results The overall success rate was 87.5%.Three patients had the prosthesis loosened and re-bonding was necessary.No loosening was found in other patients.There was no swelling or inflammation of gingival underneath the prosthesis.All patients were quite satisfactory with the function and appearance of the prosthesis.Dental calculus was observed in 12 patients,and 2 patients complained of a feeling of foreign body on the side of tongue.Conclusion The modified fixed Maryland Bridge,when used in restoring the dentition defect of geriatric patients,showed the advantages of less tooth reduction,perfect esthetic effect,good fixation of loosening teeth as well as restoration of missing teeth with its splint wings.However,the long-term effect of this technique needs to be further observed.
4.Effect of AGEs on expression of mitochondrial fusion proteins Mfn1 and Mfn2 in cultured human aortic endothelial cells

AIM: To clarify whether advanced glycation end products (AGEs) can influence the expression of mitochondrial fusion proteins Mfn1 and Mfn2 in cultured human aortic endothelial cells (HAECs) in vitro.METHODS:AGE-BSA was used as AGEs.Purchased primary human aortic endothelial cell line was multiplied, and transferred to dif-ferent passages for subsequent grouping.For dose-dependent experiment, HAECs were divided into 4 groups, and the con-centrations of AGE-BSA in each group were 0 mg/L (control group), 50 mg/L, 100 mg/L and 200 mg/L, respectively. For time-dependent experiment, HAECs were divided into 5 groups with the same concentration (100 mg/L) of AGE-BSA, but the intervention time was 0 h (control group), 6 h, 12 h, 24 h and 48 h, respectively.The mRNA and protein expres-sion levels of Mfn1 and Mfn2 in the HAECs were detected by real-time PCR and Western blot, respectively.RESULTS:Exposure of the HAECs to AGEs at different concentrations for 24 h all down-regulated the mRNA and protein expression levels of Mfn1 and Mfn2.Except for 6 h intervention group, 100 mg/L AGEs intervention for 12 h, 24 h and 48 h all down-regulated the mRNA and protein expression levels of Mfn1 and Mfn2 in cultured HAECs.CONCLUSION: AGEs down-regulates the expression of mitochondrial fusion proteins Mfn1 and Mfn2 in cultured HAECs, indicating that AGEs may influence mitochondrial dynamics of human aortic endothelial cells.
5.Artificial Abortion Has Important Effect on the Development of Adenomyosts

Objective To study the factors such as artificial abortion frequency, pregnancy time and so on on development of adeno-myosis (AM). Methods A retrospective analysis was carried in 462 patients who were from the department of gynaecology and obstetrics in Affiliated Hospital of Nantong University and Beijing Aviation Center Hospital from January 2004 to December 2008. The patients were divided into two groups: AM and no - AM. The two groups were compared for abortion frequency, pregnancy time, age, menarche age and so on. Results There were no significant differences between the two groups in age, menarche age, age of the first artificial abor-tion, bearing frequency, IUD with pregnancy and so on(P > 0.05). Objective To observe the apoptosis of neuron and protein kinase B (Akt) expression after intracerebral hemorrhage (ICH)in rats, and investigate the interference effect and mechanism of ?-aeacine sodium on ICH injury.Methods All Sprague - Dawley male rats were randomly divided into normal group, pseudo -operation group,model group and therapeutic group treated with ?-sodium aeacine.The latter three groups were divided into five subgroups respectively :6 h,12h,1d,3d and 7d.ICH models were performed by injection Ⅶ type collagenase into the the right globus pallidus.By the methods of in situ terminaldeoxynucleotidyl transferase- mediated dUTP nick end labeling (TUNEL) and immunohistochemistry, The dynamic changes of apoptosis and the expression of AKt protein were observed in the damaged cortex.Results TUNEL-positive cells appeared at 6h after collagenase injection in the model group and the expression of AKt increased at 12 h.They reached their peak at 3rd day, and were still present during 1 week. The numbers of TUNEL-positive cells in 1 d, 3 d, 1 week after ICH decreased markerly in treatment group than those of in model group ( P
8.Treatment of Closed Abdominal Trauma with Craniocerebral Trauma:34 Cases Report

Objective To summarize the clinical feature and treatment of 34 cases with closed abdominal trauma and craniocerebral trauma. Methods 34 patients with closed abdominal trauma and craniocerebral trauma were analyzed retrospectively. Results 4 cases died in hospital. Conclusion For the patients with closed abdominal trauma and craniocerebral trauma, shock and acute intracranial hypertension should be dealt with in time. Early diagnosis and operation is important to avoid secondary brain injury and to reduce the death rate.

10.Comparison of Preoperative Screening for Patients with Obstructive Sleep Apnea Hypopnea Syndrome by Cinema Magnetic Resonance and Fiber Optic Laryngoscope

Objective To localize the upper airway obstruction of patients with obstructive sleep apnea hypopnea syndrome (OSAHS)with the Cinema Magnetic Resonance(Cine-MR) and fiber optic laryngoscope with Müller maneuver(FLMM)before operation and discuss the clinical application values.Methods Before operation, FLMM and Cine-MR were applied to 22 patients diagnosed as OSAHS by Polysomnography(PSG).Medical examinations conducted in this study from September 2015 to April 2016 to examine the obstruction of the soft palate region, the lingual region and epiglottis.Results There were complete agreements between the Cine-MR and FLMM at locating obstruction sites of the soft palate (n=22/n=22),and there were moderate agreements between the Cine-MR and FLMM in locating obstruction sites of the retroglottal region(n=13/n=6),epiglottal region (n=4/n=2)and multiple level(n=13/n=6), respectively.Conclusion For those moderate and severe OSAHS patients with multiple sites obstruction , the preoperative application of the Cine-MR and FLMM together will be better in locating the obstruction sites.
