1.Effect of neuropeptides on periodontium and oral bone repair

BACKGROUND:With the development of researches on periodontal disease etiology and neuroendocrine factors, the role of neuroendocrine factors on the pathogenesis of periodontal disease and tissue restoration has become a hot spot.
OBJECTIVE:To summarize the biological effect of neuropeptides on periodontal tissues and the changes of periodontal tissue after nerve injury.
METHODS:A computer-based search of articles was performed in CNKI, Wanfang and PubMed databases using the key words ofneuropeptides, calcitonin gene related peptide, substance P, vasoactive intestinal peptide, neuropeptide Y and periodontal tissuein Chinese and English. The included articles focused on the effect of various neuropeptides on periodontal tissue.
RESULTS AND CONCLUSION:Calcitonin gene related peptide, substance P, vasoactive intestinal peptide, and neuropeptide Y positive nerve fibers are widely distributed in periodontal support tissues. When sensory nerves are damaged, the target cel s in periodontal tissues are influenced due to changes of nerve fibers distribution and neuropeptide release. They also play a role in the reconstruction of alveolar bone, immune function of periodontal ligament and periodontal tissue. This evidence shows that neuroendocrine factors are closely linked with periodontal tissue. However, these studies concentrated in the animal models for orthodontic and fracture, the effect of the neurotransmitters such as neuropeptides on periodontal disease and the mechanisms need further exploration.
2.Analysis of differential urinary proteome in experimental minimal change nephropathy

Objective To screen early urine protein markers for minimal change nephropathy.Methods Adriamycin nephropathy was employed as minimal change nephropathy model.Urinary protein and ConA captured glycoproteins were respectively profiled.Results By profiling urine proteome,25 differential proteins were identified.These differential proteins were from leaked plasma proteins,secreted proteins from immuno-and inflammatory cells,specifically asecreted proteins from urinary tract,and so on.They took part in different pathogenic process,eg.hemodynamic changes,podocytes injury,immunological disorder and so on.By profiling ConA-enriched urinary glycoproteome,21 differential proteins were identified,among which 12(57%) were different from the above 25 differential proteins.This indicates that the knowledge of urine glycoproteome is complementary to urine proteome in understanding kidney condition.Conclusion These differential proteins can be potential indicators of minimal change nephropathy,and can help better understand the pathogenesis by further studying their functions.
3.Construction of The Antigen Expression Vector Used for Library-library Screening

Selectively infective phage (SIP) technology was developed for screening interacting protein-protein pairs. The in vivo SIP strategy would in principle be suitable for??library-library??selections, and the co-packaged polyphage may be a suitable approach. In order to construct the antigen expression vector which can be co-packaged into polyphage with phage displaying vectors, plasmid TG10 was chosen as the basic vector which is compatible with antibody display vector. The interval sequence of phage genome was amplified with PCR and cloned into TG10 to provide the packaging signal. It was named pTMI and it can be packaged into phage particles in 1011 level. The N1N2 region of gene ¢? was amplified and cloned into pTMI under the control of lac promoter to give pTMIN. Promoter trc was synthesized and replaced the lac promoter to give pTTMIN which permits the fusion expression of antigen with N1N2. To test its ability for fusion expression, gene code for ten-peptide of c-myc was synthesized and inserted into pTTMIN downstream to N1N2. After induction expression, the results of ELISA and SDS-PAGE showed that it has been expressed successfully. When pTTMIN was transfected into cell carrying antibody display vector p3MHHB3, it was copackaged into phage particles in 0.3% to 55% after rescuing with helper phage VCSM13.From the results it can concluded that the antigen expression vector was constructed successfully and it can be used for library-library screening in theory.
4.Construction of helper virus VCSM13N1 with amber mutation

Objective:To construct the helper virus VCSM13N1 with amber mutation,so as to provide a platform for the selective infective phage(SIP) technique.Methods: A pair of primers with amber stop codon were synthesized and introduced into N1 region of helper virus VCSM13 protein by overlapping extension PCR,and the products were identified by ristriction enzyme digestion and sequencing.The switch on/off efficiency of amber codon was detected with E.coli XL1-Blue and HB2151.The feasibility of using the amber mutant phage as helper virus was determined with anti-HBsAg phage antibody.Comprehensive analysis was also made on the performance of the amber mutant phage.Results: The titer of mutant phage was 2.5?10~(11) when detected with E.coli XL1-Blue,comparable with that of wild phage.The titer of mutant phage was at 10~6 level when E.coli HB2151 was used,being 10 000 folds lower than that of wild phage,when the mutant phages from HB2151 were used to infect E.coli XL1-Blue,the titer peaked at 5?10~(9),being 50 folds lower than that of wild phage.The activity of phage antibody prepared with mutant virus was comparable with the phage antibody prepared with wild phage.Conclusion: The constructed amber mutant phage can be used as helper virus,and it can switch off efficiently in non-suppressing strain HB2151.
5.Rhinoscleroma disease accompanied with laryngotracheal stenosis: a case report.

Rhinoscleroma disease accompanied with laryngotracheal stenosis as a main clinical manifestation is seldom. Here one case is reported. At the beginning, the patient had pharyngalgia and his swallowing was not smooth. After that, he had difficulty in breathing. Surgical therapy, antibiotics treatment and nutritional support were performed. The patient was cured.

6.Analysis on allergens in patients with allergic rhinitis in Urumqi Xinjiang.

OBJECTIVE:
Understand the main types and characteristics of inhaled allergens in allergic rhinitis patients in Urumqi, Xinjiang, and provide theoretical basis for diagnosis, treatment, prevention of local allergic rhinitis and epidemiological investigation.
METHOD:
Inhaled allergen detecting on suspected patients with allergic rhinitis in clinic by the allergopharma skin prick test. Group the skin prick test positive patients of 340 samples according to their nationality, age and gender, then compare the inhaled allergen differences between them.
RESULT:
The total positive rate with allergic rhinitis prick test in 340 cases is 94.7%, of which the highest proportion is Artemisia (66.2%), followed by Chenopodium album (51.2%), poplar (42.6%), Ulmus (40.9%), Liu (33.8%), Timothy (27.6%), dust mite (21.2%), house dust mite (17.9%), ragweed (15.9%), dog (11.2%), cat (10.6%), cockroach (8.5%), Alternaria (5.6%) and Candida albicans (3.5%). The sensitivity of poplar, willow, Timothy and Alternaria of the minority group is higher than the Han group (P<0.05). The allergen positive rate difference among different age group has statistical significance (Χ2=59.076, P<0.01).
CONCLUSION
Artemisia, Chenopodiaceae, poplar and elm are the main allergens in Urumqi area. There is a difference in allergen sensitivity between Minority and Han nationality patients. There is no statistical significance on inhaled allergen positive rate between different genders. Specific allergens can provide basis for the prevention and control of allergic diseases in Urumqi area.

7.Construction of a packaged strain XL-pⅢ and preparation of defective helper phage

Objective To construct a packaged bacteria strain and, with it, to prepare gene III-defective helper phage. Methods Whole gene III and chloramphenicol-resistant gene were amplified and joined to form a fragment flanking 36bp homologous sequence of hisBCD using overlapping extending PCR, and it was then integrated into chromosome of E. coli XL1-Blue to replace the hisBCD gene using Red recombination system. The recombinant strain was identified with chloramphenicol-resistant screening, PCR and histidine-phenotype analysis, and named as XL-pⅢ. The gene III-deleted genome of phage was constructed with PCR and transfected into the recombinant strain XL-pⅢ to prepare the defective helper phage. It was quantified by infecting E. coli XL1-Blue and formed colonies were counted in kanamycin plate. Results The recombinant strain could grow in chloramphenicol-resistant plate, but couldn't grow in M9 medium without histidine, implying that it had lost its histidine phenotype. The aimed gene fragment could be amplified as designed. The constructed recombinant was named as XL-pⅢ. The prepared defective helper phage could infect E. coli XL1-Blue only once and the CFU was 3.7?108. Conclusions The packaged strain XL-pⅢ is successfully constructed and used to prepare the gene III-deleted helper phage. It is expected to be used in SIP system.
8.Observation and nursing with different intravenous depressors for perioperative hypertension in Intensive Care Unit

ObjectiveTo observed the clinical result of 3 kinds of commonly used depressor in Intensive Care Unit (ICU) and attend to the main point in nursing.MethodsNitroglycerin, sodium nitroprosside and urapidil hiydrochloride were used among 60 patients with perioperative hypertension in ICU respectively.ResultsSodium nitroprosside showed obviously depressed result, but needs to be observed tightly. Nitroglycerin and urapidil hiydrochloride depressed the blood pressure gentlely, and were suitable to old people particularly.ConclusionThe nurse can carry on corresponding clinical observation and nursing according to the pharmacology function of 3 kinds of medicines.

Through study on Bianque Shenying Zhenjiu Yulong Jing (Bianque's Classic of Effective Acupuncture Prescriptions), acupuncture experiences and ideology of WANG Guo-rui, an acupuncture physician of the Yuan Dynasty (1271-1368), is discussed in this article. Inherited and carried forward DOU's acupuncture theory, WANG Guo-rui encouraged to use experiential points and to expand extra points. Penetrating needling method was created, and the application of acupoints according to needling opportunities and seasonal factors were enriched by him. Great importance was also attached on treatment according to differentiation of syndromes, which including the simultaneous acupuncture and moxibustion, simultaneous reinforcing and reducing, reducing after reinforcing, reinforcing after reducing, predomination of reducing and predomination of reinforcing. The flexible treating methods are still extensively adopted in clinic nowadays. WANG Guo-rui is a master of clinical acupuncture who made great contribution to the development of acupuncture and moxibustion.

10.Significance of global end-diastolic volume index in acute kidney injury after septic shock

Objective To analyze the significance of global end-diastolic volume index (GEDVI) in acute kidney injury (AKI) after septic shock.Methods A retrospective analysis of 61 patients was performed.The patients were diagnosed of septic shock in emergency ward of Shenyang Military Hospital from 2012 March to 2013 May and were monitored by pulse indicator continuous cardiac output (PiCCO).The patients were divided into two groups:low GEDVI group (GEDVI < 700 ml/m2,29 cases) and high GEDVI group (GEDVI≥700 ml/m2,32 cases) by evaluating GEDVI of 24 hour after PiCCO.Several physiologic and biochemical indexes were recorded,including the hemodynamic parameters at the beginning and the 24 h of PiCCO monitoring,Scr,BUN,lactic acid,incidence and mortality of AKI,baseline glomerular filtration rate,baseline Scr,APACHE Ⅱ scores,mortality during the period of emergency ward or within 28 d after the diagnosis.Results A total of 26 cases in high GEDVI group (81.3%) were attacked with AKI,while 16 cases in low GEDVI group (55.2%) were attacked with AKI,the incidence of AKI in high GEDVI group was significantly higher than that in the low GEDVI group.A COX regression analysis of mortality was performed between the patients staying at emergency ward and during 28 d after diagnosis.The results indicated that AKI and GEDVI had no relation with patients' death.Therefore,AKI and GEDVI could not be considered as the risk factors for the prognosis.Conclusions High GEDVI can significantly increase the incidence of AKI after septic shock,therefore high GEDVI should be avoided as much as possible in the course of clinical treatment.
