Objective:To investigate the effects of oxaliplatin (OXA) and teniposide (VM-26) on proliferation and apoptosis of gastric cancer cell line BGC-823.Methods:MTT assay was used to examine the inhibition rate of cell growth when cells were treated at various concentrations of OXA and VM-26 alone or in combination. The apoptosis was examined by flow cytometry (FCM). The protein expression of cell apoptosis associated proteins caspase-9 and livin were examined by immunocytochemistry. Results:OXA or VM-26 effectively inhibited the growth of BGC-823 gastric cancer cells in a concentration-dependent manner at certain range of concentrations. The inhibitory rate of combined treatment with OXA and VM-26 was significantly higher than that of OXA or VM-26 alone (P<0.05). The value of combination index (CI) was 0.46. The rate of apoptosis cells induced by 1.25 μg/mL OXA was 6.13%, 13.86% and 21.48% at 12, 24 and 48 h. The apoptosis rate induced by 0.625 μg/mL VM-26 was 4.60%, 10.72% and 17.07% for 12, 24 and 48 h. In combined treatment group the apoptosis rate was 11.73%, 24.14% and 44.75% at 12, 24 and 48 h, respectively. The difference was significant between combined treatment group and single drug treatment group (P<0.05). Immunnocytochemical analysis showed that the expression of caspase-9 protein was up-regulated after being exposed to OXA (1.250 μg/mL) or OXA plus VM-26 (0.750 μg/mL), while the expression rate of livin protein was down-regulated. There were significant differences among different treatment groups as well as between treatment groups and control group (P<0.05).Conclusion:OXA combined with VM-26 has synergistic effects in inhibiting the growth and inducing apoptosis of gastric cancer BGC-823 cells.

Objective To investigate the relationship between the expressions of maspin gene in colorectal membrane,and their roles in the occurrence,progression,metastasis and prognosis of colorectal adenocarcinoma.Methods Immunohistochemistry technique was used to detect the expression of maspin in tissues of normal controls,adenoma,colorectal adenocarcinoma primary tumor,liver metastasis,lymph-node metastasis respectively.Results The expressions of maspin in normal controls,adenoma and colorectal adenocarcinoma primary tumor are down-regulated successively; the expression of maspin in colorectal adenocarcinoma with metastasis is lower than that in colorectal adenocarcinoma without metastasis(P＜0.05) ; the down-regulation of maspin expression is significantly correlated with the depth of invasion,differentiation,liver metastasis and lymph node metastasis of colorectal adenocarcinoma(P＜0.01,P＜0.05,P＜0.01,P＜0.05).Conclusion The down-regulation of expression of maspin probably plays an important role in the occurrence,progression and metastasis of colorectal adenocarcinoma.maspin is probable a predictive index for reflecting the metastasis of colorectal adenocarcinoma and provide a clue for early diagnosis of metastasis of colorectal adenocarcinoma.

Objective To discuss the clinical value of united detection of myocardial enzymes and troponin in the diagnosis of hypothyroidism.Methods 27 patients with hypothyroidism were chosen as the observation group.26 healthy volunteers were selected as the control group.CK-MB,LDH,AST,FT3,FT4,TSH,cTnT,cTnI were detected.Results The levels of serum AST,LDH,CK-MB of the observation group were significantly increased,the mean value was(52.67 ± 7.32)U/L,(265.23 ± 101.34)U/L,(26.75 ± 18.75)U/L respectively,which were significantly higher than the control group,the differences were statistically significant(P ＜ 0.05).The c TnT and c TnI positive rate of patients in observation group were 70.4％ and 81.5％,compared with the control group,the rates were much higher (P ＜ 0.05).Conclusion The united detection of myocardial enzymes and troponin had high clinical value in the diagnosis of hypothyroidism.

Objective:To construct pIRES-I-Ad?? bicistronic eukaryotic expression vector. Methods: Total RNA was acquired by TRIZOL method, the genes of I-Ad ?? chains were amplified through RT-PCR, respectively. The target genes were connected to pGEM-T vector and sequenced. Then the target genes were subcloned into pIRES bicistronic eukaiyotic expression vector and NIH3T3 cell line was transfected. Transfectant was screened by G418 antibiotics. Total RNA of transfectant was obtained by TRIZOL method, mRNA of foreign gene was examined by RT-PCR. Flow cytometry( FCM) was used to detennine foreign gene expression in protein level and surface expression in NIH3T3 cell line. Results: Bicistronic eukaryotic expression vector pIRES-I-AdaB was established. Foreign gene in mRNA level in transfectants was examined. Verified Ⅰ-Ad ?? chain wa3 expressed in high level expressed on transfected NIH3T3 cell line surface by FCM. Conclusion:Bicistronic eukaryotic expression vector pIRES-I-Ad ?? was constructed successfully. It is useful for studying antigen presentation and interaction between epitopes and MHC- Ⅱ molecule of BALB/c mouse.

Voltage-gated sodium channels (VGSCs) are known to be involved in the initiation and progression of many malignancies, and the different subtypes of VGSCs play important roles in the metastasis cascade of many tumors. This study investigated the functional expression of Nav1.5 and its effect on invasion behavior of human breast cancer cell line MDA-MB-231. The mRNA and protein expression of Nav1.5 was detected by real time PCR, Western Blot and immunofluorescence. The effects of Nav1.5 on cell proliferation, migration and invasion were respectively assessed by MTT and Transwell. The effects of Nav1.5 on the secretion of matrix metalloproteases (MMPs) by MDA-MB-231 were analyzed by RT-PCR. The over-expressed Nav1.5 was present on the membrane of MDA-MB-231 cells. The invasion ability in vitro and the MMP-9 mRNA expression were respectively decreased to (47.82+/-0.53)% and (43.97+/-0.64)% (P<0.05) respectively in MDA-MB-231 cells treated with VGSCs specific inhibitor tetrodotoxin (TTX) by blocking Nav1.5 activity. It was concluded that Nav1.5 functional expression potentiated the invasive behavior of human breast cancer cell line MDA-MB-231 by increasing the secretion of MMP-9.

BACKGROUND:In recent years, it has been a hot topic that stem cel transplantation is used to improve cardiac insufficiency after acute myocardial infarction by inducing regeneration of cardiomyocytes in the infarction regions. OBJECTIVE:To observe the effect of rosuvastatin combined with umbilical cord blood mesenchymal stem cel s transplantation on rat cardiac function after acute myocardial infarction. METHODS:Forty-five Sprague-Dawley rats were enrol ed to prepare myocardial infarction models by ligaturing the left anterior descending coronary artery. Then they were equivalently divided into model group, transplantation group and combination group. At 7 days after modeling, rats in the combination group were given injection of 300μL umbilical cord blood mesenchymal stem cel s (15.0×108) via the tail vein and by gavage once a day for 28 days with 1 mg/kg rosuvastatin;rats in the transplantation group and model group were injected with 300μL umbilical cord blood mesenchymal stem cel suspension through the tail veins or the same amount of LG-DMEM medium, respectively, fol owed by intragastrical administration of the same amount normal saline. At 5 weeks after modeling, indexes of cardiac function, level of plasma Lp-PLA2 and heat shock protein 70 in the infarction regions were detected by color Doppler ultrasound, enzyme-linked immunosorbent assay and western blot assay, respectively. In addition, pathological changes of myocardial tissues were observed using hematoxylin-eosin staining. RESULTS AND CONCLUSION:Left ventricular ejection fraction and left ventricular end-systolic pressure were significantly higher in the combination group than in the transplantation group as wel as higher in the transplantation group than the model group (P<0.05);compared with the transplantation group, left ventricular end-diastolic pressure was significantly decreased in the combination group, but significantly increased in the model group (P<0.05);the number of cardiomyocytes in the infarction regions was significantly higher in the combination group than the other groups. Additional y, expression of heat shock protein 70 in the infarction regions was significantly increased in the combination group (P<0.05). To conclude, rosuvastatin combined with umbilical cord blood mesenchymal stem cel transplantation can significantly improve rat cardiac function after myocardial infarction.

BACKGROUND:Umbilical cord blood mesenchymal stem cel transplantation can reduce myocardial apoptosis and myocardial fibrosis, thereby improving the cardiac function. OBJECTIVE:To observe the effect of umbilical cord blood mesenchymal stem cel transplantation on myocardial apoptosis in rats. METHODS:(1) In vitro test:Normal myocardial cel s cultured for 72 hours were enrol ed as control group. Myocardial cel s injured by adriamycin alone were enrol ed as injury group. Adriamycin-injured myocardial cel s were co-cultured with umbilical cord blood mesenchymal stem cel s as co-culture group. After 48 hours of culture, TUNEL assay was used to assess myocardial apoptosis in rats. (2) In vivo test:Forty-five Sprague-Dawley rats were randomized into normal control, adriamycin, and umbilical cord blood mesenchymal stem cel transplantation (cel transplantation) groups. Two weeks after cel transplantation, PowerLab was used to test cardiac function, and western blot employed to detect Bax and Caspase-3 expression levels. Meanwhile, pathological changes of the myocardial tissues were observed in the three groups. RESULTS AND CONCLUSION:(1) In vitro test:The apoptotic rate of myocardial cel s was significantly increased in the injury group compared with the control group, but decreased significantly in the co-culture group compared with the injury group (P<0.05). (2) In vivo test:In the cel transplantation group, the cardiac function indexes, expression levels of Bax and Caspase-3 and myocardial infarction size were significantly improved compared with the adriamycin group (P<0.05), but stil not recovered to the normal levels (P<0.05). These results show that umbilical cord blood mesenchymal stem cel s acting on the rat myocardial cel s can inhibit adriamycin-induced myocardial apoptosis, and the relevant mechanism is probably related to down-regulation of Bax and caspase-3 expression.

Objective To Summary the first 40 cases underwent robotic atrial septal defect (ASD) closure or atrial septal defect closure combined bicuspid valve plasty (TVP) using da Vinci S surgical System on beating heart. Methods 40 cases of atrial septal defect or combined sever tricuspid valve regurgitation were repaired using da Vinic S surgical system on beating heart from March 2009 to December 2010 in cardiovascular department of PLA general hospital. The average age was (38 ± 13) yeas old. 23 cases were female and 17 cases were male. All patients were ostium atrial septal defect with or without pulmonary hypertension. The atrial defect diameter was 1.5 -3.5 cm, and the mean diameter was(2. 8 ±1.3)cm. 9 patients had sever tricuspid valve regurgitation. Without sternotomy, the extracorporeal circulation was established through groin artery,groin vein and internal jugular vein cannulation with the guidance of transeophageal echocardiography. 3 ports of 8 mm and 1 working port of 2 cm were made in the right chest wall. After da Vinci S syetem was set up, with the assistant of bed-side surgeon, the surgeon completed the atrial septal defect closure or combined tricuspid valve plasty in the surgeon console with three dimensions visualization. During the operation, without cardioplegia administrated and aortic occlusion, the procedure was completed through right atriotomy. The pleural space was insufflated with carbon dioxide to avoid the air embolism. The direct suturing was used in 22 cases and pericardial patch were used in 18 cases. 9 patients accepted concurrent De Vega tricuspid valve plasty. The transesophageal echocardiography were used to evaluate the result of atrial defect closure or tricuspid valve repair. The operation time, robotic using time and cardiopulmonary time were compared with totally robotic atrial defect repair in arrested heart. Results All cases were accomplished successfully without complication. There was no residual shunt and air embolism. The operation time, robotic using time and cardiopulmonary time were less than the arrested group. Conclusion Robotic atrial septal defect closure or combined tricuspid valve repair on beating heart can avoid aortic ocllusion and can be utilized effectively and safely.

Objective To investigate the effects of fracture displacement and operation method on perioperative blood loss of femoral neck fractures. Methods From December 2013 to October 2014, 130 cases (58 males and 72 females, aged from 18 to 91 years, with an average age of 71.1±12.9 years) with femoral neck fractures were retrospectively analyzed for the periopera?tive blood loss. The degree of displacement was described according to Garden's grades. According to the degree of fracture dis?placement, the patients were divided into two groups:GardenⅠ-Ⅱgroup and Garden Ⅲ-Ⅳgroup, and patients' preoperative hidden blood loss was compared between two groups. According to the degree of fracture displacement and the method of opera?tion, the patients were divided into four groups:Group 1 indicates the group in which patients received cannulated screws fixation for Garden gradeⅠ-Ⅱ;Group 2 in which patients received hemiarthroplasty for Garden gradeⅠ-Ⅱ;Group 3 in which patients received cannulated screws fixation for Garden grade Ⅲ-Ⅳ; Group 4 in which patients received hemiarthroplasty for Garden grade Ⅲ-Ⅳ; and variation in the following four parameters was analyzed: the dominant blood loss, postoperative hidden blood loss, total hidden blood loss, total blood loss in the four different groups. Results For the Garden gradeⅢ-Ⅳfemoral neck frac?ture group, the preoperative hidden blood loss was significantly higher than that of the Garden gradeⅠ-Ⅱfemoral neck fracture group (t=2.267, P=0.001). The dominant blood loss volume, postoperative hidden blood loss volume, total hidden blood loss vol?ume and total blood loss volume of hemiarthroplasty groups (402.1 ± 36.8 ml, 641.3 ± 53.2 ml, 880.7 ± 61.7 ml, 1 246.1 ± 76.7 ml) were higher than those of the cannulated screws fixation group (45.8±34.9 ml, 301.9±50.6 ml, 436.6±58.6 ml, 478.5±72.9 ml). Conclusion The perioperative hidden blood loss is mainly related with the degree of fracture displacement, the greater the de?gree of fracture displacement, the more the preoperative hidden blood loss. While the dominant blood loss volume, postoperative hidden blood loss volume, total hidden blood loss volume and total blood loss volume are mainly related to the method of operation, the blood loss in which patients received hemiarthroplasty should be increased significantly.
