1.Study on enrichment and purification of astragalosides Ⅳ from Radix astragali with macroporous resins

Objective:To study the enrichment and purification method of astragaloside Ⅳ from Radix astragali with macroporous resin,so as to lay a foundation for the large scale production of astragaloside Ⅳ.Methods: A HPLC-MSD method was established to determine the content of astragaloside Ⅳin Radix astragali.The adsorption ability and capability of different macroporous resins were investigated.The optimal alcohol concentration and capability of alcohol for elution were also confirmed through the determination of astragaloside Ⅳ in the eluted solution.Results: HPLC-MSD achieved satisfactory results in determination of astragaloside Ⅳ.D101 was proven to be the most potent macroporous resin;its adsorption capability of astragaloside Ⅳ was 0.42 mg/g when the elution solution was 5 folds of 70% alcohol.Conclusion: D101 macroporous resin can be used to effectively enrich and purify astragaloside Ⅳ,laying a foundation for analyzing the constituents of total astragalosides.
2.Expression of ALEX1 in breast cancer and its clinical significance

AIM: To detect the expression of ALEX 1 in the breast cancer tissues in order to verify whether ALEX1 has correlation with clinical pathological features in breast cancer .METHODS:Real-time PCR and immmunohis-tochemistry were applied to detect the expression of ALEX 1 at mRNA and protein levels in the breast tissues .The statistical analysis were performed for determining the correlation with the level of ALEX 1 and the clinical pathological features in breast cancer .RESULTS:The protein levels of ALEX1 in the breast cancer tissues were lower than that in the non-breast cancer tissues (P<0.01).The expression of ALEX1 had correlations with pathological grade , clinical stage, molecular type (P<0.05) but had no correlation with the patients ’ age, tumor size and tumor types in breast cancer .Furthermore, the result of real-time PCR showed that mRNA expression of ALEX 1 was also significantly reduced in the breast cancer tis-sues (P<0.01).CONCLUSION:The expression of ALEX1 in the breast cancer tissues is lower than that in non-breast cancer tissues .The pathological grade and clinical stage in breast cancer are negatively correlated with the expression of ALEX1.
3.Diversity of fungi derived from the Cricetulus migratorius from the Xinjiang region of China

To monitor and analyze Cricetulus migratorius fungal diversity ,60 adult Cricetulus migratorius brought from Xinjiang region of China were dissected after being euthanized and the specimens were collected .Fungal diversity research was carried out by TaqMan MGB probe real-time fluorescence quantitative PCR ,the ribosome cloning sequencing ,and fungal cul-ture identification techniques .The 60 fungal isolates were characterized from Cricetulus migratorius ,including Candida albi-cans ,Trichosporonasahii ,Aspergillus fumigatus ,Aspergillusniger ,Aspergillussydowii ,Aspergillus japonicus ,Asper-gillus ustus ,Aspergillus versicolor ,Penicillium chrysogenum ,Paecilomyces variotii ,Penicillium aurantiogriseum ,Neuros-pora sitophila ,Neurospora intermedia ,and Cladosporium cladosporioides .Many of them associated with grey hamster as zoonotic pathogens .The results showed that the most dominant fungal group was Aspergillus ,and Penicillium followed by it . These fungi were susceptible to nystatin ,clotrimazole and voriconazole .It’s indicated that application of TaqMan MGB probe real-time fluorescence quantitative PCR ,the ribosome cloning sequencing ,and fungal culture identification techniques could ef-fectively analyze Cricetulus migratorius .The results could provide a scientific basis for Cricetulus migratorius microbiological monitoring and quality standard establishment in China .Overall ,the findings of the present study constitute ,to the authors’ knowledge ,the first extensive report on the diversity of fungal flora associated with Cricetulus migratorius .

Polycystic ovary syndrome (PCOS) is one of the most popular diseases in obstetrics and gynecology research at internal and abroad at present, traditional Chinese medicine(TCM)in the clinical treatment of the disease have the advantage. Clinical epidemiological study of descriptive research method this research adopts investigation, observation of TCM syndromes and improper diet through 401 cases in Jiangsu Province confirmed PCOS patients, to explore the relationship between TCM syndrome type distribution and improper diet factors, and to provide the clinical basis for further etiology of this disease research. TCM syndrome type distribution of the disease is kidney deficiency, phlegm stagnation syndrome, qi stagnation and blood stasis syndrome, syndrome of dampness heat of liver channel and is composed of 4 basic syndromes and formed complex syndrome, and the composite and syndrome type (60.85%); combined with the analysis of traditional Chinese medicine dialectical, Pure empirical syndrome this disease (46.88%), followed by the actual card (45.39%), pure deficiency is rare. Improper diet factors associated with the disease, in which improper diet with different TCM syndrome type distribution significantly related. Stagnation of phlegm dampness syndrome is the main syndrome of the disease type, improper diet factors and every syndrome PCOS type distribution is as follows: the partial eclipse fatness greasy with basic syndromes of phlegm dampness stagnation of kidney deficiency syndrome, the nephrasthenia syndrome is less; eating spicy stimulation by basic syndromes of stagnation of Qi and blood stasis; eating cold people the basic certificate type of qi stagnation and blood stasis; The diet of patients are more prone to stagnation of phlegm dampness syndrome.
