1.Phacoemulsification and implantation of artificial lens for traumatic cataract

Objective To investigate the effect of surgical management of phacoemulsification and implantation of artificial lens for traumatic cataract.Methods The surgery was performed on 19 patients.Results Post-operatively, the visual acuities were 0.5 or better in 57.9% and 1.0 or better in 15.8% in one week; 0.5 or better in 78.9% and 1.0 or better in 21.0% in one month.Conclusion It can have better effect to perform phacoemulsification and implantation of artificial lens in different conditions with surgery skill and better equipment.
2.Efficacy of acting κ opioid receptor for prevention of high altitude pulmonary edema in rats

Objective To evaluate the efficacy of acting κ opioid receptor for prevention of high altitude pulmonary edema (HAPE) in rats.Methods Forty male Sprague-Dawley rats,aged 8 weeks,weighing 250-300 g,were randomly divided into 5 groups (n =8 each) using a random number table:control group (group C),hypobaric hypoxia group (group H),normal saline + hypobaric hypoxia group (group NH),U50488H (a selective kappa-opioid receptor agonist) + hypobaric hypoxia group (group UH),and nor-binaltorphimine (norBNI,a selective kappa-opioid receptor antagonist) + U50488H + hypobaric hypoxia group (group NUH).The rats were put into the hyperbaric chamber and exposed to hypobaric hypoxia (atmospheric pressure 355 mmHg,partial pressure of oxygen 74 mmHg) for 2 days to induce HAPE.At 3 days before HAPE,normal saline 0.5 ml,U50488H 1.25 mg/kg,and nor-BNI 2.0 mg/kg were injected intraperitoneally once a day in NH,UH,and NUH groups,respectively,and in addition U50488H 1.25 mg/kg was injected intraperitoneally 10 min later in NUH group.After 2 h exposure to hypobaric hypoxia,mean pulmonary artery pressure (mPAP) was detected,and arterial blood samples were collected for determination of serum malondialdehyde (MDA) and erythropoietin (EPO) levels.The rats were then sacrificed and lungs were removed for microscopic examination and for determination of the levels of nitric oxide (NO),inducible nitric oxide synthase (iNOS),MDA,superoxide dismutase (SOD),endothelin-1 (ET-1),thromboxane B2 (TXB2),and 6-keto-prostaglandin F1α (6-keto-PGF1α) in lung tissues.Lung water content and TXB2/6-keto-PGF1α ratio was calculated.Results Compared with group C,mPAP,lung water content,ET-1,MDA,TXB2 and 6-keto-PGF1α levels,TXB2/6-ketoPGF1α ratio,and serum MDA and EPO levels were significantly increased,and iNOS,NO and SOD levels were decreased in the other four groups (P < 0.05).Compared with group H,mPAP,lung water content,ET-1,MDA,TXB2 and 6-keto-PGF1α levels,TXB2/6-ketoPGF1α ratio and serum MDA and EPO levels were significantly decreased,and iNOS,NO and SOD levels were increased in UH group (P < 0.05),and no significant changes were found in the indexes mentioned above in NH and NUH groups (P > 0.05).The pathological changes of lung tissues were significantly attenuated in group UH as compared with H group.Conclusion Acting κ opioid receptor can produce prevention for HAPE in rats,and inhibition of lipid peroxidation and correction of the imbalance between vasoconstrictive factors and vasodilative factors may be involved in the mechanism.
3.Expressions and clinicopathological significance of MCM5 and E2F-1 in patients with gastric carcinoma

Objective:To explore the relationship between the expressions of MCM5 and E2F-1 with the genesis,clinicopathological parameters of gastric carcinoma.Methods:Immunohistochemistry SP method was used to detect the expressions of MCM5 and E2F-1 in gastric car- cinoma tissues of 57 cases and in normal gastric mucosa of 20 cases.The correlation between expressions of MCM5 and E2F-1 and clinicopathological parameters of gastric carcinoma was analysed. Results:The positive expression rate of MCM5 in cancer tissues was 71.9%(41/57),while in normal mucosa it was 0(0/20).The difference between the two groups was statistically significant (P0.05).The correlation between MCM5 and E2F-1 expressions in gastric carcinoma was highly significant positive(r=0.635,P=0.000).Conclusion: Overexpression of MCM5 and E2F-1 corelates with occurence and development of gastric carcinoma.MCM5 and E2F-1 may work in the early phase in carcinogenesis of gastric carcinoma.
4.Short-and medium-term efficacy of artificial cervical disc replacement versus fusion for cervical spondylosis

BACKGROUND:Recently, non-fusion technology representing as artificial cervical disc replacement continues to improve. On the basis of reconstruction of disc structure and function of involved segments, cervical spine structure of surgery area segment is significantly close to dynamic and static load stress distribution required by natural physiological systems. It effects are apparent in protecting intervertebral facet joints of degenerated segment and structure and function of the cervical spine of adjacent segments and in maintaining cervical dynamic stability, which presented obvious methodological strengths compared with segmental fusion technology.
OBJECTIVE:To evaluate the clinical outcomes of anterior cervical discectomy and fusion and Bryan artificial cervical disc replacement in the treatment of single-level cervical spondylotic myelopathy or radiculopathy.
METHODS:A total of 43 middle and old age patients with single-level cervical spondylotic myelopathy or radiculopathy, who were treated from March 2010 to March 2012, were enrol ed in this study. They were randomly assigned to anterior cervical discectomy and fusion group (fusion group) and Bryan artificial cervical disc replacement group. Range-of-motion of cervical overal and adjacent intervertebral area near the intervertebral space was observed with radiography. During fol ow-up, postoperative recovery of neurological function was evaluated using Japanese Orthopaedic Association scale, visual analog scale and neck disability index.
RESULTS AND CONCLUSION:None patients experienced complications of neurovascular injury during and after the surgery. Range-of-motion of postoperative overal cervical vertebra and adjacent joint was improved in the Bryan artificial cervical disc replacement group compared with the fusion group. Neurological function was apparently improved after surgery in each group. At 3 months after surgery, scores of Japanese Orthopaedic Association, visual analog scale and neck disability index were significantly improved in the Bryan artificial cervical disc replacement group compared with the fusion group (P<0.05). During final fol ow-up, there were significant differences in visual analog scale scores between the two groups. Japanese Orthopaedic Association scale score and neck disability index score were similar between the two groups. During fol ow-up, no prosthesis sinking, displacement or heterotopic ossification were detected. These data indicated that artificial cervical disc replacement could effectively keep the range of motion of cervical segments and protect disc degeneration of adjacent segment. Mid-term fol ow up obtained similar improvement of neurological function of fusion surgery. The moderate-term and short-term efficacies of non-fusion technology were better than fusion technology in the treatment of single-level cervical spondylopathy.
6.Strengthening a dental gypsum model by infiltration of cyanoacrylate.

OBJECTIVETo explore a simple but novel method of strengthening gypsum material by cyanoacrylate infiltration. To evaluate the influence of cyanoacrylate on the mechanical properties of dental gypsum models.
METHODSGypsum specimens were polished to the dimension of 35 mmx4 mmx4 mm. Butyl-cyanoacrylate was diluted with chloroform at different concentrations, namely 20% and 30% cyanoacrylate. Gypsum specimens were infiltrated by diluting one component of cyanoacrylate at different concentrations for 8 h and then dried for analysis. The changes in elastic modulus, fracture toughness, compressive strength, biaxial strength, brinell hardness were measured. The data were analyzed using software OriginPro 8.
RESULTSThe viscosity measurements indicated that diluted cyanoacrylate were Newtonian fluids and the viscosity increased slightly within the 48 hours of preparation but still similar as water at room temperature, which could be used to infiltrating gypsum. The gypsum infiltrated with cyanoacrylate exhibited good physicochemical properties. The biaxial strength, fracture toughness, compressive strength and brinell hardness of the gypsum were improved by 39%, 30%, 63% and 18%, respectively.
CONCLUSIONCyanoacrylate can significantly improve the strength of gypsum model which indicates the potential clinical application.

7.Expression of matrix metalloproteinases-13 in the damage process of rat articular chondrocyte induced by fluoride and aluminium

Objective To observe the influence of fluoride and aluminum on the expression of matrix metalloproteinase-13(MMP-13) in rat articular chondrocytes. Methods Original generation chondrocytes of rats was cultured and divided into fluoride group, aluminum group, fluoride plus aluminum group and control group. NaF and A1C13 at concentrations of 1 mmol/L and 2 mmol/L were administered to intoxicate the cells for 24, 48, 72 h respectively. Cells were extracted to undergo reverse transcription the polymerase chain reaction(RT-PCR) at different times to observe mRNA expression of MMP-13, and protein expression was detected by Western-blot. Results In 24 h, the content of MMP-13 mRNA in fluoride group(0.830±0.043), aluminum group(1.279±0.060) and fluoride plus aluminum group(0.983±0.028) was higher than that in the control group(0.707±0.026, P<0.05), and relative expression of MMP-13 mRNA in aluminum group was the highest. In 48 h, the content of MMP-13 mRNA in fluoride group (0.964±0.180), aluminum group (1.333±0.105) and fluoride plus aluminum group (0.915±0.137) was higher than that in the control group(0.660±0.055, P<0.05), and the relative expression in aluminum group was the highest. In 72 h, the content of MMP-13 mRNA in fluoride group(0.866±0.115), aluminum group(0.846±0.089) and fluoride plus aluminum group(0.967±0.196) had no statistical significance(P>0.05) compared with the control group(0.809±0.179). In 24 h, the content of MMP-13 protein in fluoride group(1.050±0.084), aluminum group(1.010±0.113) and fluoride plus aluminum group(0.977±0.202) had no statistical significance(P>0.05) compared with the control group(0.860±0.038). In 48 h, the content of MMP-13 protein in fluoride group(0.671±0.020), aluminum group(1.134±0.094) and fluoride plus aluminum group (0.923±0.087) was higher than that in the control group (0.647±0.025, P<0.05), but no significant difference being observed between groups (P>0.05). In 72 h, the content of MMP-13 protein in fluoride group(0.672±0.022), aluminum group(1.088±0.072) and fluoride plus aluminum group(0.772±0.030) was higher than that in the control group(0.577±0.026, P<0.05). It was the highest in the aluminum group, the intra-group difference had statistical significance(P<0.05). Conclusions Fluoride and aluminum damage chondrocytes to some extent, toxicity of aluminum itself is greater than fluoride and fluoride plus aluminum. Abnormal expression of MMP-13 can be observed in the chondrocyte damage process induced by fluoride and aluminum.
8.Clinical Study ofYiqi Huatan Granules Combined with Carbocisteine to Reduce Acute Exacerbation of Chronic Obstructive Pulmonary Disease

Objective To observe the efficacy and safety ofYiqi Huatan Granules combined with carbocisteine to reduce the acute exacerbation of chronic obstructive pulmonary disease (COPD). Methods Totally 108 patients in the stationary phase of COPD with qi deficiency or merging phlegm blockage/blood stasis syndrome were randomly divided into two groups. Patients in the control group were treated with carbocisteine and inhaled bronchodilators on-demand, while patients in the treatment group were treated withYiqi Huatan Granules combined with carbocisteine and bronchodilators for 24 weeks. Times and severity of acute exacerbations of COPD, TCM syndrome scores, lung function, and 6-minute walking distances of the two groups during the treatment period were observed.Results The average times of acute exacerbations of COPD in the treatment group (0.33 ± 0.55) were fewer than the control group (0.62 ± 0.71);the average times of severe acute exacerbations of COPD in the treatment group (0.12 ± 0.33) were fewer than the control group (0.28 ± 0.45), with statistical significance (P<0.05). Comparison between the TCM syndrome scores before and after treatment, TCM total syndrome scores, breathless/dyspnea score, breath shortness and fatigue score, and 6-minute walking distance in the treatment group were better than the control group (P<0.05), with better safety.Conclusion The treatment ofYiqi Huatan Granules combined with carbocisteine for COPD patients can effectively decrease times and severity of acute exacerbations of COPD, and improve TCM syndrome scores and 6-minute walking distance.
9.Clinical Study on Qingjin Huazhuo Prescription Combined with Western Medicine Therapy for Treatment of Acute Exacerbation of Chronic Obstructive Pulmonary Disease

Objective To observe the efficacy and acute inflammatory reactionof Qingjin Huazhuo Prescription combined with Western medicine therapy for treatment of acute exacerbation of chronic obstructive pulmonary disease (AECOPD). Methods Totally 60 hospitalized patients with AECOPD (Grade Ⅱ) and acute exacerbation course≤1 week were enrolled in this study. The patients were randomly divided into two groups, with 30 cases in each group. The control group was treated with conventional Western medicine. The observation group was treated with conventional Western medicine and oral administration of Qingjin Huazhuo Prescription for 10 days, once a day. TCM syndrome and dyspnea grading (mMRC score), white blood cell (WBC), C-reactive protein (CRP), procalcitonin (PCT) and partial pressure of oxygen (PaO2), partial pressure of carbon dioxide (PaCO2), were measured before and after the treatment. Results The total scores of TCM syndromes and cough, expectoration, wheezing, fever, and cyanosis in the two groups decreased significantly (P<0.05). The improving degrees of cough and expectoration were better in the observation group than in the control group (P<0.05). The scores of mMRC in the two groups were better than before treatment (P<0.05). The levels of WBC, CRP and PaCO2 in the two groups were lower than those before treatment (P<0.05). The decreased degrees of CRP and PaCO2 in the observation group were better than those in the control group (P<0.05, P<0.01). Conclusion Qingjin Huazhuo Prescription combined with Western medicine therapy can relieve clinical symptoms, reduce the acute inflammatory reaction of AECOPD patients and improve respiration.
10.The effect of apoptosis induced by IL-21 in SUDHL-4 cell line and its mechanism.

OBJECTIVETo investigate the apoptosis effect of diffuse large B-cell lymphoma cell line (DLBCL) SUDHL-4 induced by IL-21 and its related mechanism.
METHODSSUDHL-4 cells were treated with IL-21 at different concentration (1000 ng/ml, 100 ng/ml, 10 ng/ml, 1 ng/ml) for 24 h, 48 h, 72 h, respectively. The inhibitory rate of cell proliferation was detected by CCK-8 assay. The cell growth curves were drawn and half inhibitory concentration (IC(50)) values were calculated. The cell apoptosis were detected by flow cytometry (FCM), the expression of the caspase-9, caspase-3, cleaved caspase-3, Bcl-2, Bcl-XL, Bid, Bax and c-myc protein in SUDHL-4 cells treated with IL-21 by western blot, the mRNA expression of Bcl-2, Bcl-XL, Bid, Bax, c-myc by Survivin gene with RT-PCR.
RESULTSIL-21 markedly inhibited SUDHL-4 cell growth in a time- and dose-dependent manner. The 48 hIC(50) was 140.9ng/ml; The FCM showed that the apoptosis proportion of SUDHL-4 cells treated with 100 ng/ml of IL-21 apoptosis (AnnexinV-FITC(+) positive cells) gradually increased (48 h: 19.7 ± 2.3%). The protein expression of caspase-9, caspase-3, Bcl-2 and Bcl-XL decreased in a time-dependent manner. The Bax and c-myc protein markedly increased, but the Bid protein level did not change. IL-21 up regulated c-myc and Bax gene expression, however down regulated Bcl-2 and BCL-XL gene expression, but the gene expression of Bid and Survivin hadn't been changed significantly.
CONCLUSIONSIL-21 can inhibit proliferation and induce apoptosis of SUDHL-4 cell. The mechanism may involve in endogenous mitochondrial pathway mediated by the c-myc and the Bcl-2 genes.
