Journal of Chongqing Medical University 2003;0(05):-
Objective:To investigate effects of dendritic cells(DCs) pulsed with autologous hepatoma cell lysates on the immune function of the postoperative patients with hepatocellular carcinoma(HCC).Methods:DCs isolated from peripheral blood mononuclear cell of HCC patients were cultured and proliferated in vitro by using rhGM-CSF and rhIL-4,and then were pulsed with autologous hepatoma cell lysates.30 postoperative patients with HCC were randomly divided into 2 groups.15 cases were treated with dendritic cell vaccine.15 cases received only chemotherapy and were established as control group.Immune function、clinical effects in two groups of patients were compared and observed. Results: The levels of DC3~+、DC4~+/CD8~+ and NK cell in the DC vaccine group significantly increased after vaccination, while those of the control group had no significant changes. The levels of IgG、IgM、IgA in the DC vaccine group increased after vaccination, but didn't reach statistical significance. The concentration of IL-10 in the DC vaccine group significantly decreased after vaccination(P
2.Research progress of type Ⅲ interferons
Chinese Journal of Biologicals 2025;38(05):617-623
Interferons(IFNs) are a family of glycoproteins with antiviral, antitumor, and immunomodulatory activities, classi-fied into type Ⅰ, Ⅱ, and Ⅲ based on structural and receptor specificity. Among these, type Ⅲ IFNs(IFNλs), including IFNλ1(IL-29),IFNλ2(IL-28a),andIFNλ3(IL-28b),exhibittissue-specificreceptordistribution,predominantlyexpressedinepithelialcells and select immune cells, thereby demonstrating reduced systemic side effects compared to typeⅠIFNs. Mechanistically,IFNλs exert their antiviral effects through JAK/STAT signaling pathway inducing the expression of interferon-stimulatedgenes(ISGs), and inhibit tumor cell proliferation, promote apoptosis and regulate immune responses. Recent advances inrecombinant IFNλs therapeutic development include polyethylene glycol(PEG) modified IFNλ1, a longacting formulationcurrently in phaseⅢclinical trials for hepatitis C, hepatitis D, and COVID-19. In this paper, the structures and receptors, signaling pathway and regulation, biological functions and research progress of IFNλs are summarized in order to provideinsights for the development of IFNλs related products.

Chinese Journal of Laboratory Medicine 2003;0(07):-
The new, rapid, accurate and convenient tests for tuberculosis diagnosis and drug resistance are urgently needed for effective tuberculosis control. In this review, two novel, inexpensive diagnosis methods of active TB were introduced. They can detect tuberculosis disease and drug resistances directly form sputum specimens and might have greater applicability in developing countries.Several concerns about the research and development of innovative diagnosis technology in our country also are described.
5.Differentiation of marrow stromal cells into neurons: current progress
Chinese Pharmacological Bulletin 2003;0(07):-
Bone marrow stromal cells(MSCs) have been shown to be extremely versatile.Recently,it is demonstrated that MSCs differentiate into neurons. Moreover, under the appropriate conditions,MSCs differentiate into special phynotype of neurons, such as 5-hydroxytryptamine-sensitive neurons, and dopaminergic neurons. The mechanism remains to be elucidated, and an overexpression of neuronal specific genes(such as Sox,Pax,Notch gene) may play an important role, especially RB/PBO gene. However,RB triggers cholinergic differentiation.Extrinsic factor such as concentration of cAMP plays certain role in differentiation.MSCs have also been shown to have certain therapeutic effect on neuronal degeneration disease. These cells have great potential because it is easy to isolate, culture, amplify, and no ethics neither immunologic rejection problem was concerned.
6.Optimal immunohistochemical diagnostic index for supraclavicular lymph node metastasis in lung cancer
International Journal of Laboratory Medicine 2015;(1):5-7
Objective To explore the optimal immunohistochemical (IHC)diagnostic index for supraclavicular lymph node me-tastasis in lung cancer. Methods In the premise of conforming to the evidence-based medicine, the biopsy of lymph node in 135 pa-tients with supraclavicular lymph node metastasis was performed and 7 antibodies were selected to be detected.135 cases were di-vided into two classes:class A (110 cases)and B (25 cases). The class A included 14 cases of primary lesion inside the lung by the histological analysis,43 cases of primary lesion inside the lung by the cytological detection and 53.cases of primary lesion inside the lung by the radiographic analysis. The class B included 25 cases of lesion outside the lung by the histological analysis. The detecting antibodies included CK7,CK20,EMA,CEA,TTF-1,SPB and vimentin. Results The sensitivity and specificity of single antibody for deducing the primary lesion inside the lung were 90.0% and 56.0% for CK7,98.0% and 40.0% for CK20,90.9% and 4.0%for EMA,80.9% and 36.0% for CEA,62.7% and 100.0% for TTF-1,65.6% and 100.0% for SPB,and 60.9% and 60.0% for vimentin. The quadruple antibodies (CK7,CK20,TTF-1 and SPB)showed the highest sensitivity(85.0%)and highest specificity (100.0%),which made the relatively high detection rate (80.0%)of lung cancer in clinic. Conclusion Analyzing and researching the IMC results is of great significance to guide the surgical pathological practice based on the evidence-based medicine.

8.Evaluation of randomized controlled trials in the studies of clinical nutrition
Chinese Journal of Clinical Nutrition 2009;17(6):321-323
Objective To evaluate the quality of the randomized controlled trials (RCTs) published in two key Chineze journals on clinical nutrition. Methods The articles published in CHINESE JOURNAL OF CLIN-ICAL NUTRlTION and PARENTERAL & ENTERAL NUTRITION from 2000 to 2008 were reviewed and the RCTs were identified according to criteria of the handbook of Cochrane Collaboration. The Jadad scale was used to evalu-ate the quality of these RCTs.Results Totally 238 RCT articles were published in these two journals in this peri-od.The Jadad score of all RCT articles was 1.65±0.82.Twenty-eight articles (11.76%) were of high quality and only 5 articles (2.10%) were identified 5 points. There were some problems in the RCTs design, conduction and analyses included unclear randomization methods, poor comparison, lack of inclusion and exclusion criteria.less blinding employment, unclear withdrawals and dropouts, and improper sample size. Conclusion The design and quality control of Chinese clinical nutrition RCTs still have some problems and require further improvement.
9.Role of COX-2-independent targets of celecoxib and related compounds on cancer prevention and treatment
International Journal of Surgery 2009;36(8):541-544
We reviewed the role of COX-2 independent targets of celecoxib and related compounds in cancer prevention and treatment. Direct non-COX-2 targets of celecoxib might be protein-dependent kinase 1 (PDK-1),Ca+ ATPase and carbonic anhydrase (CA), and so on; all of which are inhibited by celecoxib and are the main components in various pathways. Inhibition of these proteins leads to the induction of apoptnsis, the inhibition of cell cycle progression, angiogenesis, and metastasis, which are the main mechanisms by which celecoxib exerts its anticarcinogenic activity.
10.Effect of 2-methoxyestradiol on apoptosis of bone marrow cells of patients with multiple myeloma
China Oncology 2001;0(03):-
Purpose:To explore the in vitro effects of apoptosis induction of 2-methoxyestradiol(2ME2),an estrogen derivative on primary myeloma cells and hematopoietic precursors from multiple myeloma patients. Methods:The primary myeloma cells and hematopoietic precursors from 14 multiple myeloma patients were examined determine apoptosis cells in situ by terminal-deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL)assay. Results:After co-cultured with different concentrations (1,4,16?mol/L) 2 ME2 for 48h respectively, apoptotic ratios of myeloma cells were 10.5? 1.2%,21.2?2.3%,36.8?3.6% respectively ,which shows a concentration- dependence. In comparision with the control group, the difference is of statistical significance(P0.05). Conclusions:2ME2 could electively induce apoptosis of primary myeloma cells from myeloma patients and has little apoptotic effect on granulocytic, erythrocytic and lymphocytic cells in the bone marrow.
