As a novel Golgi protein,GOLPH2 expression is associated with maintaining the structural integrity of the Golgi apparatus. As an oncoprotein,GOLPH2 expression is associated with malignant biological behavior of cancer cells.Important roles of GOLPH2 in the tumorigenesis and metastasis make it become a new target for tumor diagnosis and treatment.

To observe the changes in hemorheology, endotoxin and TNF ? in blood after hemoperfusion(HP) with adsorbent immobilized polymyxin B (PMB) on sepsis in rats. Wistar rats were randomly divided into three groups:LPS+HP+PMB,LPS+HP and LPS. All the rats received intravenously injection of lipopolysaccharide (LPS, Escherichia coil O111:B4,1mg/kg). Plasma of the rats in the group of LPS+HP+PMB was passed through a column containing sepharose coated activated charcoal immobilized polymyxin B at the 4th hour after LPS injection. The treated plasma was transfused bak after being mixed with blood cells. In LPS+HP group, the column did not contain immobilized polymyxin B. The animals of LPS group received LPS only. Quantitative endotoxin determination in blood was done with limulus amebocyte lysate test,TNF ? of the plasma assayed with ELISA, and hemorheology parameters were also observed after hemoperfusion. In LPS+HP+PMB group, the concentration of plasma was significantly decreased after hemoperfusion, but it was still significantly higher than the baseline value, and there was a decrease of blood cell ratio after hemoperfusion. The results suggest that specific adsorbent could remove endotoxin in the circulation and improve hemorheology.

Objective To study radiation protection of low dose CT scanning in orbit. Methods 126 cases of orbital disease were scanned by original standard parameter (130 kV,100 mA) and two different low dose parameters (130 kV,50 mA and 30 mA) respectively. All the images were reconstructed by different Kernels. The image quality and difference of the CTDIw (weighted CT dose index) of each group were compared. Results CTDIw was 30% of that of the conventional scanning and least when 130 kV 30 mA,but the difference of the image quality compared with that by conventional scanning was highly statistical significance (P0.05). Conclusion Low dose CT scanning (50mA) in orbit not only can assure the image quality but also can decrease the radiation dose.

Objective To study the characteristics of mesenchymal stem cells(MSCs)derived from chronic myelogenous leukemia(CML)patients' bone marrow and examine their hematopoiesis support in vitro. Methods MSCs from CML were obtained and cultured.Immunophenotype and in vitro differentiation capacities were investigated. Moreover, the Ph chromosome and bcr/abl gene of CML derived MSCs were detected. The expression of cytokines was detected by RT-PCR,and hema- topoiesis support of MSCs in vitro was detected by long-term bone marrow culture and the methylcel- lulose progenitor assay. Results CML derived MSCs showed a typical fibroblast like morphology. They were positive for CD29,CD44 and CD105,while negative for CD14,CD31,CD45,CD34 and HLA-DR.Under suitable conditions,CML derived MSCs could differentiate into osteoblasts and adi- pocytes.Further,CML derived MSCs showed a normal choromosome, and did not express bcr/abl gene.At last, they expressed hematopoiesis cytokines and possessed ability of hematopoiesis support in vitro. Conclusion There are MSCs with multidirectional differentiation potential in CML bone mar- row and they possess the ability of hematopoiesis support.

Objective To investigate the correlation between the formation of pigmented biliary calculus and biliary H.pylori infection. Methods Bile from 35 patients with pigmented biliary calculus and 10 healthy controls was cultured for aerobic, anaerobic and H.pylori.The expression of H.pylori- DNA in bile, bile duct mucosa and pigmented calculus were determined by PCR.The expression of H. pylori associated protein in bile duct mucosa was determined by Western-blot and Warthin-Starry staining. Results H.pylori culture was negative in all bile samples. In 35 patients with biliary pigmen- ted calculus,H.pylori was detected by PCR in the center of calculus, bile and bile duct mucosa of 14.29%,31.43% and 56.67% patients, respectively.Among H.pylori-DNA positive bile samples,7 contained anti-CagA antibodies, and 6 contained Vac A.in addition to Vacuolating cytotoxin(35000), glycoprotein(30000),Urase Band Urase A.Bacteria resembling H.pylori by Warthin-Starry stainning were found in 7 of 30(23.33%)bile duct mueosal samples from patients with biliary pigmented calculus. H.pylori-DNA and its associated protein were not detected in all bile and bile duct mucosae samples from the healthy controls. Conclusions The evidence of H.pylori-DNA and its associated protein in biliary system might indicate the role of H.pylori in the formation of biliary pigmented calculus.

To construct human antibody repertory with phage display technology. Total RNA was extracted from peripheral blood lymphocytes (PBL) of clinical glioma patients and subjected to reverse transcription PCR. Variable region genes(V ? and V H genes) were amplified from the cDNA using nested polymerase chain reaction. V ? gene library was first constructed and attached to the Linker which could specially connect V ? gene and V H gene, then V H gene was cloned into V ? gene library to build ScFv repertoire. Positive colonies were selected randomly and sequenced. The results implied that amplified human V ? gene and V H gene could be derived from PBL of clinical glioma patients. ScFv phage antibody gene repertoire in about 2?10 6 capacity was successfully constructed. The sequencing indicated that the cloned genes encoded variable regions of heavy chains of human antibody and were highly homologous with human kappa heavy chain group III of Kabat's database. Then human antibody repertoire was successfully constructed with phage display technology. The present study can be used as a foundation for succeeding screening of specific antibody against human glioma.
