1.High mobility group box 1 and its role in cervical cancer

Extracellular high mobility group box 1 (HMGB1) can cause persistent inflammation via related receptors and signal pathways such as receptor for advanced glycation endproduct,Toll-like receptors and chemokines 12,which results in tumorigenesis.In patients with cervical cancer,the expression of HMGB1 increases with the rising of malignant gTade.HMGB1 participates in the genesis and development,invasion and metastasis of cancer through multiple processes.The expression of HMGB1 is inhibited by RNA interference,thus affects the invasion and migration of cervical cancer cell,which provides a new concept for the development of antineoplastic agents.
2.Expression and correlation of Hedgehog signaling pathway and LKB1 gene in breast cancer

Hedgehog signaling pathway is excessive activated in breast cancer.LKBl is currently accepted as a tumor-suppressor gene,which can inhibit the proliferation of breast cancer.The overexpression of LKB1 can regulate the expression of CyclinDl series gene which is a target gene of Hedgehog signaling pathway.Meanwhile,the study found that PKA gene play an important role in Hedgehog signaling pathway,its activation is related with the cAMP state,and LKB1 genes can influence the cAMP state.Therefore,LKB1 gene and the Hedgehog signaling pathway may exist some inevitable connection.
3.Effects of Gravity on The Pressure of Blood Flow in a Tapered Vessel:Based on a 3D FSI Mathematical Model With Posture Change

Gravity is the most fundamental biomechanical stimulus for posture change.Pressure of blood flow is one of primary indicators to evaluate cardiovascular performance.Up to now, the underlying mechanism of effects of posture change on cardiovascular system is still unclear.A 3D FSI mathematical model with posture change was presented.By applying the body force terms to the fluid equation and the vessel wall equation, the model could be used to study posture change and the effects of gravity on the pressure of blood flow quantitively.Under different inlet-outlet pressure difference(IODP) and different postures such as horizontal, upright and upside-down one, the effects of gravity were simulated.In horizontal position, the pressure distributions of blood flow transformed from 2D(two-dimensional) axis-symmetry without gravity to 3D asymmetry with gravity under small IODP.With IODP increasing, gravity had less effects on pressure distribution and extreme value.As IODP reached 10 665.6 Pa(80 mmHg) and 2 666.4 Pa(20 mmHg) respectively, this effect was observed to be constant.Similar results were obtained from 3D fluid-only model.In either upright or upside-down position, 2D axis-symmetric pressure distribution was observed with and without gravity, yet the position, in which extreme pressure appeared, was different in upright position from that in upside-down one.Finally, the effect intensity of gravity in upright or upside-down position was more than twice as much as that in horizontal one.The results indicate that introducing body force term into the fluid and solid equations to present a novel model, which was based on hemodynamics, will provide a new way to study posture change.Effects of gravity on pressure distribution and extreme value changed with different postures and IODP.If IODP is small, ignoring effect of gravity and postures so as to simplify the hemodynamics model to 2D axis-symmetric one, the conclusion should be drawn with caution.
4.Effect of maternal hypothermic cardiopulmonary bypass on fetal homodynamic and carbohydrate metabolism

Objective To evaluate effects of maternal hypothermic cardiopulmonary bypass on fetal homodynamic and carbohydrate metabolism. Methods Twenty pregnant sheep were divided into four groups randomly: control group(n=5),normothermic group (35-36℃)(n=5), mild hypothermic group(32-34℃)(n=5) and moderate hypothermic group (28-31℃)(n=5).Thoracotomy was performed without CPB in the control group. Routine CPB was established with different temperature in other three groups. The temperature of normothermic group was kept normal; the left two groups were cooled down to the set point of temperature and then rewarmed back to normal level. Fetal and maternal temperatures, heart rate,mean blood pressure(BP), pulse index (PI) of fetal umbilical artery (UA) and internal carotid artery (CA) were evaluated at cooling and rewarming stages. Biochemical indicators including blood glucose and lactic acid were also measured at the same time. Results There are no differences in mesn BP of ewas and fetal lambs between the different groups (P>0.05). CA PI value of mild hypothermic group and moderate hypothermic group were significantly higher than those of control group and normothermic group (P<0.05). There was no difference of UA PI in the four groups, but PI increased following the prolonged duration of CPB. There was no difference change of blood glucose in the four group of fetus, which was significantly lower than the ewe groups. An upward trend of fetal blood lactic acid with time was observed in three CPB groups. The whole level of fetal blood lactic acid was much higher than that of maternal blood of lactic acid. Conclusion Cooling of maternal bypsss decreases fetal heart rate significantly,and fetal heart rate recovered to base line following rewarming phase. There was no signicant effect of CPB on fetal mean BP. However, CPB impacted on the blood flow of fetal brain and umbilical artey. Hypothermia CPB can increase fetal blood glucose and blood lactic acid dramatically.
5.Proliferation capacity of rabbit adipose tissues-derived mesenchymal stem cells at early stage in vitro

BACKGROUND:Surface marker molecules and differential ability of human adipose tissues-derived mesenchymal stem cells can change with the prolonged culture time.OBJECTIVE:To analyze the morphological characteristics and colony forming capacity of early cultured rabbit adipose tissues-derived mesenchymal stem cells in vitro.DESIGN,TIME AND SETTING:The cytological in vitro study was performed at the Institute of Ophthalmology,and Central Laboratory,Eye and ENT Hospital,Fudan University from January to March 2008.MATERIALS:Nine female New Zealand rabbits aged three months were obtained from the Shanghai Yingenyang Rabbit Room.METHODS:Adipose tissue was harvested from the nape fat pad of the New Zealand rabbits following anesthesia.Primary cultured cells were established using type I collagenase and cell cultures were maintained with DMEM containing 0.1 volume fraction of fetal bovine serum.Cells were passaged when 80% was confluent.The second,third and fourth passages of cells were utilized for the study.MAIN OUTCOME MEASURES:The morphology of adipose tissues-derived mesenchymal stem cells was observed using inverted microscope.Cell phenotype was detected utilizing flow cytometry.The cloning capacity was determined.RESULTS:Adipose tissues-derived mesenchymal stem cells were fibroblast-like and proliferated rapidly in vitro at the second,third and fourth passage.Flow cytometry showed that adipose tissues-derived mesenchymal stem cells expressed CD29 and PCNA.The colony-forming rates were respectively(8.0?0.6)%,(6.7?0.4)% and(4.6?0.5)%(F=12.18,P
6.Survey and analysis about the science research integrity of community health organizations in Yangpu District of Shanghai

Objective To survey the scientific research integrity situation of community health care organizations in Yangpu district in Shanghai,meanwhile,provide a reference for scientific research integrity management in any community health care organization.Methods Using the convenience sampling method,we sampled 200 health staff from 4 community health service centers.They were surveyed with uniform questionnaires.Results No dramatically relationship existed in health staff's knowledge about scientific research integrity breach and their age.However,education background,their academic titles,how many times they have learned relative information,publicity about science research integrity would impact their awareness on research integrity.Conclusions There is dramatically discrepancy on the scientific research integrity among these community health staff.
7.Clinical feature and immunological analysis of a cluster of medical workers with severe acute respiratory syndrome in Guangzhou

Objective To discuss epidemiological, clinical and laboratory characteristics,chest radiography, serum immunology and conditions in convalescence in patients with severe acute respiratory syndrome(SARS). Methods We retrospectively analyzed clinical data of 22 medical workers with SARS and observed the change of Immunoglobulin M and Immunoglobulin G in their serial sera immunology. Results The patients ranged from 23 to 53 years old (mean age 32.5?7.0). The incubation period ranged from 2 to 7 days(mean period 4.1?1.49 ). Infectious patterns suggested through air droplet or close contact transmission. The most common symptoms included fever(100%), chilly(68.2%), followed by nonproductive cough (72.7%) and dyspnea (63.6%) associated with infiltrates on chest radiography (77.3%). Normal in white blood count or Leukopenia (90.9%) , Lymphopenia (45.5%) and elevated alanine aminotransferase levels (54.5%) were common. By testing the serum IgM and IgG antibody of patients, we found a positive result in 2 weeks after the onset of SARS. Though IgM antibody show negative 3 months later, IgG antibody remained a high level. Conclusions SARS appears to be highly infectious.Fever and subsequent respiratory syndrome is the most common symptom and sign. Our research has disclosed that a positive serum IgM antibody means a infection in the near future, while a positive serum IgG antibody indicates a infection of past and probably with immunity.
8.Bactericidal mechanism of electrolyzed oxidizing water against Pseudomonas aeruginosa

Objective To investigate the bactericidal mechanism of electrolyzed oxidizing water (EOW) against Pseudomona aeruginosa (P.aeruginosa).Methods Bactericidal mechanism of EOW against P.aeruginosa was studied through intracellular protein leakage,nucleic acid,and cell membrane calcium ion permeability,2 % glutaraldehyde was used as positive control group,and normal saline (NS) was used as negative control group.Results The killing rates of EOW and 2% glutaraldehyde to P.aeruginosa were both>99.99% with 30-second contact time,and 100.00% with 60-second contact time.After 60-second contact with EOW,NS,and 2% glutaraldehyde,the protein leakage of P.aeruginosa detected by bicinchoninic acid (BCA) were (96.00 ± 7.42),(94.15 ± 7.49),and (216.97 ± 10.35)μg/mL,respectively,difference was significant(F =613.20,P<0.01),2% glutaraldehyde group was higher than EOW group and NS group;protein leakage did not change with the increase of contact time(all P>0.05).Electrophoretogram of random amplified polymorphic DNA showed high intensity dense band between 500-1000 Kb in EOW group and NS group,while 2% glutaraldehyde group was without amplified bands.The fluorescence intensity of calcium ion of EOW group and 2% glutaraldehyde group were both lower than that of NS group.Conclusion Bactericidal mechanism of EOW may be due to the damage of membrane permeability of P.aeruginosa,which causes Ca2+ leakage,but fails to cause protein leakage,the damage to nucleic acid is not obvious,DNA may not be a bactericidal target of EOW.
9.Approach to the patients with Gitelman syndrome

10.Expression of p73 Protein in Giant Cell Tumor of Bone

Objective: Our aims were to study the expression of p73 protein in giant cell tumor of bone, and to investigate its function in the carcinogenesis and development of giant cell tumor of bone. Methods: The expression of p73 was detected by using S-P immunohistochemical method in 40 cases of paraffin-embedded sections of giant cell tumor of bone. Results: The positive rate of p73 in 40 patients with giant cell tumor of bone was 30.0%,and 20.0% in osteochondroma.The difference between them was not significant(P<0.05). There was significant difference between the expression of p73 in different stages of giant cell tumor of bone(P>0.05). Conclusion: The expression of p73 in giant cell tumor of bone increased prominently in this experiment. It suggests that p73 may play an important role in the carcinogenesis and development of giant cell tumor of bone, and p73 may also have a certain value for the diagnosis of giant cell tumor of bone.
