1.Relationship of expression of clusterin with expressions of bcl-2 and p53 in prostate epithelia

Objective To investigate the expression of an inhibitor gene of apoptosis, clusterin, in prostate cancer and its relationship with the genesis and progression of prostate cancer and with the expressions of bcl 2 and p53. Methods The expressions of clusterin, bcl 2, and p53 in 10 cases of normal prostatic tissues, 15 cases of benign prostatic hyperplasia (BPH), and 49 cases of prostate cancer were examined by immunohistochemical staining. Results The rates of strong positive or weak positive of clusterin in normal prostatic tissues, BPH, and prostate cancer were 10% (1/10), 66.6% (10/15), and 91.8% (45/49), respectively. 3.Surgical Treatment of Skin Cancer in Facies in the Elderly

Objective To study the surgical repair mode of skin cancer in facies in aged patients. Methods The modes of surgical repair in 216 cases of aged patients with skin cancer in facies were analyzed. Results The special characteristics of physiology and pathology and the facies anatomy in the elderly determined the complexity of surgical repair. The surgical mode of excision and suture was the safest and the most efficacious. Skin flap transfer could mostly satisfy the function requirements of facies for the wound that was not able to be sutured after excision. Free skingrafting was applicable to the case that the deep tissue was not invaded by the tumor and the general condition of the patients was not much good. Conclusion The order of surgical repair modes for skin cancer in the facies in the elderly is excision and suture, skin flap transfer, and free skingrafting. Local skin flap must be the primary source as the selection of the skin flap is concerned. The suitable axial pattern skin flap or musculo-cutaneous flap can be selected according to the different section of the facies while there is no suitable local tissue.
4.Construction of a phage display library of human single-chain Fv antibodies from peripheral blood lymphocytes of people with positive serum antibody against hepatitis B virus core protein

Objective To construcr a phage display library of human single-chain Fv antibodies from peripheral blood lymphocytes of people with positive serum antibody against hepatitis B virus core protein. Methods The heavy chain and light chain variable region gene of human immunoglobulin were amplified individually by RT-PCR from peripheral blood Lymphocytes mRNA of patients with positive serum hepatitis B virus core protein antibody and randomly combined through a DNA linker encoding the peptide (Gly 4Ser) 3 to construct single-chain variable fragments (ScFv)gene. In the presence of helper phage M13KO7, the ScFv were displayed on the surface of recombinant phages. Results Repertoire antibody library was obtained with 10 6 clonies resistant to ampicillin and kanamycin. Conclusions A typical phage display library of human single-chain Fv antibodies has been constructed Using RT-polymerase chain reaction (RT-PCR) and phage display technique, human single-chain Fv antibodies can be screened from this library.
5.Clinical Application of Acellular Dermic Graft in Plastic Surgery

Objective To investigate the clinical effect of acellular allogenic dermis combined with split-thickness autogenous skin graft for coverage of wound in extremity joint. Methods After contraction scar, giant pigmented nevus and capillary hemangioma being removed, acellular allogenic dermis with split-thickness autogenous skin graft was used to repair 25 cases of wounds of elbow, ankle, knee and wrist joints. The area of wounds was from 7cm?11cm to 12cm?19cm. Out of 25 wounds, there were 4 in wrist, 8 in elbow, 7 in ankle and 6 in knee joints. The survival rate of graft skin and improvement degree of joint function were observed. Results All grafts survived and had the smooth surface without obvious pigmentation and wound contraction. The function of joint was significantly improved after operation. Conclusion Tissue-engineered dermis with autogenous epithelium is an ideal graft for coverage of various wounds in extremity joint.

7.Treatment of acromioclavicular dislocation (Tossy Ⅲ ) with AO clavicular hook plate

Objective To study the clinical effect and necessity of removing internal fixation after treatment of acromioclavicular dislocation (Tossy Ⅲ ) with AO clavicular hook plate. Methods From October 2001 to October 2004, 31 patients with acromioclavicular dislocation (Tossy Ⅲ ) were treated with AO clavicular hook plate and were followed up. All of their broken acromioclavicular joints were sutured and the clavicular insertions of deltoid and trapezius muscles were repaired. The coracoclavicular ligaments were sutured for only 9 patients. Their shoulder functions before and after taking out of the internal fixation were assessed by Constant and Murley evaluation and compared. Results 24 patients were followed up from 4 to 40 months after operations (averaging 22 months). There were no loosening or breakage of internal fixations. The plates were removed 12 to 27 months postoperatively (averaging 18 months) in only 9 patients. No redislocation occurred after taking out of the plates.
