1.Expression of ?_3 -adrenoceptor and its function in human detrusor cells

Objective To study the expression of ? 3 adrenoceptor (? 3 AR) and its function in human detrusor cells. Methods Cellular functional studies,radioligand binding experiments and reverse transcription polymerase chain reaction were used to determine the expression and its influence on detrusor contractions. Results mRNA of 3 subtypes of ? ARs was detected. Non selective ? AR agonists and ? 3 AR decreased the contractions of detrusor cells induced by KCL, but both agonist and antagonists of ? 1 and ? 2 AR could not influence the contraction;BRL37344 concentration dependently displaced dihydroalprenolol binding to the membrane of detrusor cells. Conclusions The mRNA of subtypes of ? ARs is distributed in human detrusor cells,but ? 3 ARs is the main receptor mediating detrusor relaxation.
2.Dual-dissociation method to record whole-cell current of wistar rat neurons

BACKGROUND: The main difficulties of whole-cell patch clamP are cell culture and whole-call current recording. OBJECTIVE: To reduce the difficulty of patch-clamp experiment by combining acute cell isolation with current separation technique. METHODS: A total of 40 wistar rats aged 4-7 days, irrespective of genders, were selected. The pallium of wistar rats were cut into slices with 400-600 μm, rested for 1 hour in artificial cerebrospinal fluid with gas mixture, simultaneously put in oxygen. The brain tissues were placed into artificial cerebrospinal fluid containing 16 u/mL(type X) and 2 u/mL(type XIV) proteinase, incubated for 60 minutes, and cleared the digestive enzyme. Under the whole-cell voltage clamp mode, the potential was hold at -80mV, depolarizing pulse stimulation from -60 mV to 60 mV, 10 mV step and 160 ms width. The total Tran membranes current was recorded, and then 70 mmol/LCsCI, 70mmol/L CsF was 11 μmol/L Na~+ channel blockers tetrodotoxin, 30 mmol/L tetraethylammonium, 1 mmol/L 4-AP was successively added into extracellular fluid. The inward sodium current, transient outward potassium current and delayed rectifier potassium current was recorded, and then the result was analyzed using clampfit. MAIN OUTCOME MEASURES:①Cell morphological observation; ②Whole-ceil current recording;③Inward sodium currentrecording; ④Outward potassium current recording.RESULTS AND CONCLUSION: The cell had clearly three-dimensional structure, smooth surface and whole neurodendrite or neuraxon. The cell viability could maintained for 8-10 hours at 25 ℃ temperature. The added 11 tetrodotoxin in extracellular fluid could block sodium Current. The outward potassium current could be blocked by tetraethylammonium with 30 mmol/L and 4-AP with 1 mmol/L. The calls harvested by modified rapid dissociation have good functions. Using current separation method, only specific blockers are needed, without extracellular fluid or electrode solution replacing, which can record the inward sodium current, transient outward potassium current, as well as delayed rectifier potassium current. This method can obviously improve the work efficiency than traditional one.
3.Age-dependent alterations in ?-adrenoceptor function in human detrusor and possible mechanism

0.05).The responsiveness of detrusor muscle cells in elderly group to isoprenaline,BRL37344 and forskolin was significantly lower than that in control group ( P 0.05 ).The Bmax of ?-adrenoceptor was 26.9 fmol/mg and 32.4 fmol/mg in elderly and control groups,respectively,with significant difference between the 2 groups (P0.05). Conclusions There is an age-related decline in ?-adrenergic responsiveness,which might be one of the causative factors of the reduced bladder compliance of the elderly.The decrease of cAMP level caused by reduced receptor density and adenylyl cyclase activity might be the molecular mechanisms underlying the changes of ?-adrenergic responsiveness.
4.Analysis on the differences of medical education between America and China from observing the teaching of Utah University

United States is the world's Medical Center, and American medical education is also a typical representative of the western medical education. This study takes the Medical School of Utah University and Tongji Medical College of Huazhong University of Science and Technology as an example to analyze and compare the similarities and differences between Chinese and American medi-cal schools in the aspects of subject setting , curriculum planning , teaching method and auxiliary teaching means. The result shows that both are basically the same in the setting of the course of clini-cal medicine, adopt integration module and PBL teaching, but there are fewer subjects of basic course at the school of medicine of Utah university, and online teaching and studying are more commonly used as an auxiliary teaching means.
5.Effect of NIP regime in treatment of non-small cell lung cancer

This research is to investigate the effect of NIP regime in the treatment of non small cell lung cancer (NSCLC) and its side effects.Thirty two patients with NSCLC (stageⅢ 28 cases, include Ⅲa 6 cases and Ⅲb 22 cases, stageⅣ 4 cases) received chemotherapy regime NIP.Vinorelbine(NVB)25 mg/m 2 V.D,first day and eighth day ;Ifosfomide(IFO) 1.6 g mg/m 2 V.D, from first day to third day; Cisplatin(DDP)25 mg/m 2 V.D,from first day to third day.Four weeks was a cycle, every patients was given 2 cycles. RR was 53.13%(CR 1/32,PR 16/32,SD 4/32,PD 9/32). Main side effect was mylosuppression.NIP regime is effective and safe for patients with advanced NSCLC.
6.Alternation in ?-adrenergic responsive cAMP of human detrusor and its regulation in the aged

Objective To study the influence of age on ?-adrenergic responsive cyclic adenosine monophosphate (cAMP) level of human bladder detrusor and its modulation mechanism. Methods The cAMP levels in detrusor muscle of aging group (15 cases) and young group (7 cases) were determined by radioimmunoassay at the presence of isoprenaline (ISO), BRL 37344 and forskolin, in addition, the modulation of cAMP content of aging group by pertussis toxin(PTX), cholera toxin(CTX), vinpocetine(VPT) and KT-5720 were also studied. Results After incubation with ISO, BRL 37344 and forskolin, the cAMP level of aging group was (15.0?1.6),(12.2?1.8)pmol/mg and (4.5?4.0) pmol/mg protein. The cAMP level of aging group was not influenced significantly by CTX; the ?-adrenergic responsive cAMP level was enhanced significantly by PTX and KT-5720, but the increment of cAMP content induced by forskolin was not changed; VPT significantly increased the cAMP levels induced by all the three agents. Conclusions There is age-related decline in the ?-adrenergic responsive cAMP level of detrusor muscle of the aging; reduced activity of adenylyl cyclase and enhanced activity of inhibitory G protein Gi may be the biochemical basis; inhibitors of calcium/calmodulin stimulated phosphodiesterase and protein kinase A improves the ?-adrenergic responsiveness of the senile, therefore might be used to treat the urinary storage dysfunction during aging.
7.Determination of diosgenin in Rhizoma Paridis by HPLC and TLC

Object To determine the contents of diosgenin in Rhizoma Paridis by HPLC and TLC. Methods ODS column was used in HPLC with mobile phase: acetonitrile water(90∶10), detection wavelength: 203 nm, column temperature: 35 ℃; silica gel plate was used in TLC with developer: cyclohexane ethyl acetate (4∶1), scan wavelength: 610 nm, reference wavelength: 420 nm. Results The linear ranges of HPLC and TLC were 1 2 - 7 2 ?g (r=0 999 8), 0 2 - 1 0 ?g (r=0 995 7). The average recovery and RSD were 102 2%, 3 39% (n=6) and 100 9%, 2 68% (n=6) respectively. Conclusion Both of two methods can be used for the quality control of diosgenin in Rhizoma Paridis.
8.Protective effects of hydrogen sulfide on acute spinal cord injury in rats

Objective To investigate the effects of hydrogen sulfide on autophagy and the apoptosis after acute spinal cord injury in rats. Methods Thirty-six adult male SD rats (250-300 g) were randomly divided into three groups (n=12 for each group):sham operation group (Sham group), spinal cord injury group (Model group) and hydrogen sulfide pre-treatment group (H2S group). Allen’s method was used to establish the rat model of spinal cord injury. Rats of sham operation group re?ceived only laminectomy. Rats of H2S group received sodium hydrosulphide injection intraperitoneally (50μmol/kg) 1h after spinal cord injury, and Model group was given the same amount of saline solution. Rats in the three groups were sacrificed 24 h after spinal cord injury, then the spinal cord was removed. The expressions of LC3, p70S6K and Cleaved caspase-3 were detected by Western blot assay. The expression of LC3 was also detected by immunofluorescence. The cell apoptosis was as?sessed by TUNEL stain. Results Compared with Sham group, the expression levels of LC3Ⅱ/LC3Ⅰand Cleaved caspase-3 were increased in Model group, but the expression of p70S6K decreased and cell apoptosis increased in Model group (P<0.01). Compared with Model group, the expression levels of LC3Ⅱ/LC3Ⅰand Cleaved caspase-3 were decreased significant?ly, while the expression of p70S6K increased and cell apoptosis decreased significantly in H2S group (P < 0.01). Conclu?sion Hydrogen sulfide can inhibit autophagy and reduce cell apoptosis after acute spinal cord injury in rats.

10.The immunohistochemical study of the macrophages and natural killer cells in hepatocellular carcinoma tissues

Objective:To investigate the number and distribution of macrophages (Mφ) and natural killer(NK) cells in hepatocellular carcinoma (HCC), paraneoplastic,cirrhosis and normal liver tissues and their relationship with the prognosis of HCC patients. Methods:Surgical specimens from 60 cases of HCC, 62 cases of cirrhosis and 23 cases of normal liver tissues were investigated by immunohistochemical staining of CD68 and CD57 with a streptavidin-horseradish peroxidase detective system.The correlation of the number of Mφ and NK cells in different tissues with the clinical tumor parameters was also studied. Results:①The order of the number of Mφ from the highest to the lowest was:paraneoplastic,cirrhosis, normal, HCC(P<0.05);and the number of NK cells from the highest to the lowest was:HCC, paraneoplastic, normal, cirrhosis(P<0.05).②The number of Mφ decreased successively with the decrease of the HCC differentiation(P<0.05);There was no relationship between the number of NK cells in HCC and histological grade. ③There was no relationship between the number of Mφ in HCC and clinical TNM stage ;The number of NK cells in HCC had degressive tendency with the clinical TNM stage(P<0.05). ④The number of Mφ and NK cells in HCC in the cases with metastasis in 15 months was significantly lower than that without metastasis(P<0.05, 0.01).⑤There was a linear positive correlation between the number of Mφ and NK cells in HCC (r=0.344, P<0.05). Conclusion:The number of Mφ and NK cells in HCC in the cases with metastasis is significantly lower than that without metastasis;The number of Mφ has a close correlation with the HCC differentiation, and positive correlation with the number of NK cells;The number of Mφ and NK cells might be important markers to estimate the immune status and useful factors to predict the prognosis of HCC patients.
