Objective To study the effects of hydrogen dioxide (oxygen free radical donator) and vitamin C (oxygen free radical scavenger) on the electric current of large conductance calcium -activated potassium channels (BKCa channels) in isolated outer hair cells in aging guinea pigs .Methods Acute enzyme was used to isolat outer hair cells of aging guinea pigs ,in which of BKCa channel's electric current was observed and recorded by whole-cell recording mode of patch -clamp .After recording the stable and normal electric current of BKCa channels ,added H2 O2 dilution (0 .2 mmol/L) 40 μl in the 2 ml chambers within freshly isolated outer hair cells so that the concen‐tration of H2 O2 in the balneum would be 4 μmol /L .The groups(n=5) received individually vitamin C solution (5 mg/ml) 0 ,10 ,20 ,40μl in the 2 ml chambers within freshly isolated outer hair cells so that the concentration of vi‐tamin C in the balneum would be 0 ,25 ,50 ,100 μg /ml ,observing and recording the effects of different concentration of vitamin C to electric current of BKCa channels .Results ①In the w hole-cell mode of patch -clamp ,the rapid activation and non-deactivation electric current with a string of large amplitude was recorded ,above -40~ -30 mV activation voltage .The electric current increased with the increasing membrane potential .The amplitude in‐creased continuously and performed characteristics of outward rectification .When the concentration of IbTX was 100 nmol /L ,the activity of the channel was completely blocked and confirmed BKCa channel's electric current .②Medication within three minutes ,when VT was +50 mV ,the BKCa channels'the maximum peak current densities of 4 μmol /L H2 O2 group rose from 22 .09 ± 0 .27 PA /PF to 43 .53 ± 1 .09 PA/PF ,amplification was 97 .06% .In H2 O2 4 μmol /L + vitamin C with different concentrations as 25 ,50 ,100 μg/ml groups ,the BKCa channels'elec‐tric current performed about concentration-dependent inhibition ,and electric current's amplitude and peak current density decreased with the increasing concentration of vitamin C ,the I-V curves were reduced .However ,this still could not be recovered to the normal levels .Conclusion The oxygen free radical /BKCa exists in the process .The vitamin C as oxygen free radical scavenger can reverse the process to a large extent .

Objective To observe and assess the effect about bundles of cure Rule of three three in the treatment for residual stones after Flexible Ureteroscope.Methods 120 patients with a single stone after urinary lithotripsy were divided into two groups according to the different indwelling time.Two cases of the control group were rejected because of severe hematuria.The control group (n=58) was subjected to the regular education (drinking water 3 000 ml per day with moderate exercise);The experiment group (n=60) was subjected to the Rule of three three (drinking 3 cups of water in thc morning、midday and night,with contralateral position 30 minutes,hot towel applying and gently tapping the kidney region 30 minutes).Results The calculi discharge rate and the net rate of experiment group [86.67％ (52/60),55.00％ (33/60)] were higher than the control group [53.45％(32/58),18.97％(11/58)] 14 days after surgery.The difference was statistically significant (P ＜ 0.01).Further analysis showed that the discharge rate of residual stones in lower renal calyx in experiment group [86.67％(26/30)] was higher than the control group [32.14％(9/28)].The difference was statistically significant (P ＜ 0.01).Conclusions The bundles of cure called rule of three three can improve the discharge rate of residual calculi after flexible ureteroscope and increase the discharge rate of residual stones in lower renal calyx significantly.

Objective To explore the influences of different inductive methods on cartilage repair by tissue engineered cartilage with the seed cell of mesenchymal stem cells.Methods Rabbit mesenchymal stem cells were divided into dexamethasone-inducing group and TGF-?_1-dexamethasone co-inducing group.The cartilage defects were repaired by autologous tissue engineered cartilage constructs.The defects of control group were filled with scaffold without cells.Specimens were harvested 6 and 12 weeks postoperatively and assessed by histological grading and in situ detection of apoptosis.Results The repair tissue formed perpendicular column structure resembling that of normal cartilage in TGF-?_1-dexamethasone co-inducing group.The histological score 12 weeks postoperatively was higher in co-inducing group(20.26?1.35) than those in dexamethasone-inducing group(14.52?1.46) and control group(4.12?1.13).But the formation of tidemark was not observed in the repair tissue.Cells of repair cartilage at the bone-cartilage interface showed apoptosis.The ratios of apoptosis in dexamethasone-inducing group were(21.4?4.5) six weeks postoperatively and(7.3?2.2) twelve weeks postoperatively.The ratios of apoptosis in TGF-?_1-dexamethasone co-inducing group were(19.8?4.7) six weeks postoperatively and(6.9?2.0) twelve weeks postoperatively.The ratios of apoptosis at 6 th week were higher than those at 12 th week postoperatively with statistical significance(P

Objective To study the effect of total flavonoids of Pollen Typhae on the autophagy of macro-phages and its effect on Akt/mTOR signaling pathway. Methods To observe the effect of serum containing drugs on the proliferation of SD rats. The serum containing 20%of the drug-containing serum and the macrophages were selected by the method of MMT. Interleukin(IL-10),interferon-γ(IFN-γ)and autophagy-related genes Akt and mTOR mRNA and protein were detected. Results There was no significant difference between the OD value of TYTF serum in the concentration range of 10% ~ 40% and so as the OD value of normal control(P > 0.05). The expression of autophagosomes in each dose group of TYTF was significantly increased under TEM. The autophagic expression was not typical in the normal control group. The expression of Beclin1 protein in TYTF group was signifi-cantly higher than that in normal control group(P<0.05),while the expression of p-Akt and p-mTOR protein was significantly lower than that of normal control group(P<0.05). Akt and mTOR protein expression compared with the normal control group was not significantly different (P > 0.05). The expression of Beclin1 mRNA in TYTF group was significantly higher than that in normal control group (P < 0.05),while the expression of Akt and mTOR mRNA was significantly lower than that of normal control group (P < 0.05). The expression of IL-10 in TYTF group was significantly higher than that in normal control group (P < 0.05). The expression of IFN-γ was significantly lower than that of normal control group(P<0.05). Conclusions The total flavonoid serum has anti-atherosclerotic effect. The mechanism may be related to inhibition of the activation of Akt/mTOR signaling path-way,including macrophage autophagy activity induction ,plaque macrophage infiltration reduction and inflamma-tory response factors secretion inhibition ,inflammatory response activities inhibition. Thereby it can promote atherosclerosis plaque stability of the purpose of vulnerability.

BACKGROUND:Human leukocyte antigen (HLA), the major histocompatibility complex of human, plays an important function in the transplant rejection. Decreasing the expression of HLA wil prolong the survival time of transplants.
OBJECTIVE:To design smal interfering RNA (siRNA) of HLA-A2 and to detect the effect of siRNA-HLA-A2 on the expression of HLA-A2.
METHODS:Four kinds of siRNA-HLA-A2 domains were designed, and recombinant lentivirus expression vector were formed. The 293T cells, highly expressing HLA-A2, were infected in vitro. Then the knockout efficacy of four domains was detected to select the highly efficient siRNA-HLA-A2 target sequences. The human embryo lung fibroblasts were cultured in vitro and infected with the lentivirus carrying the target sequence. The infecting efficiency of LV-siRNA-HLA-A2 was observed under the fluorescence microscope and the silence function of this siRNA in human embryo lung fibroblasts was detected by western blot analysis.
RESULTS AND CONCLUSION:According to the mRNA sequence of HLA-A2 in Genbank, three siRNAs were designed and synthesized. In vitro, the over expression of HLA-A2 in 293K cells was successful y silenced. The HLA-A2 expression in human embryo lung fibroblasts was also efficiently silenced after the human embryo lung fibroblasts were infected by the highly efficient siRNA of HLA-A2. The efficacy was up to 80%.

Objective To study the low dose hyper-radiosensitivity in human lung cancer cell line A549,and its possible mechanisms.Methods Exponentially growing A549 cells were irradiated with 60Co γ-rays at doses of 0-2 Gy.Together with flow cytometry for precise cell sorting,cell survival fraction was measured by mean of conventional colony-formation assay.ATM1981 Ser-P protein expression was examined by Western blot.Apoptosis was identified by Hoechst 33258 fluorescent staining,and Annexin V-FITC and propidium iodide staining flow cytometry.Cell cycle distribution was observed by flow cytometry.Results There was an excessive cell killing per unit dose when the doses were below about 0.3 Gy,and the cells exhibited more resistant response at the doses between 0.3 and 0.5 Gy,the cell survival fraction was decreased as the doses over 0.5 Gy.The expression of ATM1981Ser-P protein was first observed at 0.2 Gy,followed by an increase over 0.2 Gy,and reached the peak at 0.5 Gy(compared with 0.2 Gy group,t=7.96,P＜0.05),with no further increase as the doses at 1.0 and 2.0 Gy(t=0.69,0.55,P＞0.05).24 hours after irradiation,part cells presented the characteristic morpholos4cal change of apoptosis,and the apoptosis curve was coincident with the dose-survival curve.Compared with the control group,the cell cycle had no change post-irradiation to 0.1 and 0.2 Gy.G2/M phase arrest was manifested at 6 and 12 hours post-irradiation to 0.3,0.4 and 0.5 Gy(t=2.87,2.88,4.92 and 3.70,3.12,8.11,P＜0.05),and the ratio of G2/M phase was decreased at 24 hours post-irradiation(t=3.87,4.77,3.01,P＜0.05).Conclusions A549 cells displays the phenomenon of hyper-radiosensitivity(HRS)/induced radioresistance(IRR).The model of cell death induced by low dose irradiation is mainly apoptosis.The activity of ATM and cell cycle change might play an important role in HRS/IRR.

AIM:To investigate the differences among Lenstar 900, A-scan ultrasound and keratometer in measurement of axial length ( AL ) , anterior chamber depth ( ACD ) and corneal curvature ( K1 , K2 , Km ) , and evaluate the consistency of the instruments, with the purpose providing references for the clinical application of Lenstar 900.
METHODS: In this study we picked up 36 patients ( 50 eyes ) underwent cataract surgery, and lens nucleus hardness were under level IV. Before the operation, AL, ACD and K1 , K2 , Km were measured by Lenstar 900, A-scan ultrasound and keratometer respectively. The differences between the results were compared by the paired t-test. The correlation of the results was analyzed by Pearson correlation analysis, and the consistency was measured by Bland-Ahamn method.
RESULTS: The mean AL and ACD values measured by Lenstar 900 and A-scan ultrasound had no significantly statistic differences (P>0. 05). The K1, K2, Km measured by Lenstar 900 and keratometer were not significantly statistical different (P>0. 05). The results measured by these three instruments had close linearity correlation ( r>0.9, P<0. 01). The consistency of the results was well in Bland-Ahamn analysis.
CONCLUSION:The preoperatively biometric result of Lenstar 900, A - scan ultrasound and keratometer in patients with cataract are all reliable, and they can be substituted by each other. However, Lenstar 900 can not only measure AL, ACD and corneal curvature at the same time, but also cornal thickness, lens thickness, white to white, pupil size, optical axis eccentricity, retinal thickness and so on. It has a number of advantages such as non-touching, convenient and efficient, and can be recommended to use widely.
