1.Genetic analysis of clinical varicella-zoster virus isolates collected in China

Objective To study the genetic characteristrics of vaccine and varicella-zoster virus(VZV)strains isolated from patients with chickenpox or zoster by molecular analysis.Methods SNP based VZV genetic characteristrics were analyzed in 19 VZV isolates using the restriction fragment length polymorphisms analysis of DNA fragments of the open reading frames 6,38,62 and sequence alignment of the open reading frames 1,31,51,62.Results All vaccine strains were revealed Alu Ⅰ~-Pst Ⅰ~-Sma Ⅰ~+ BssH Ⅱ~+ Nae Ⅰ~+,96% clinical isolates were revealed Alu Ⅰ~+ Pst Ⅰ ~+Sma Ⅰ BssHⅡ Nae Ⅰ~-,2% clinical isolates were revealed Alu Ⅰ~-Pst Ⅰ~-Sma Ⅰ~+ BssH Ⅱ~+,2%clinical isolates were revealed Alu Ⅰ~+ Pst Ⅰ~-Sma Ⅰ~-BssHⅡ Nae Ⅰ~-by restriction fragment length polymorphisms analysis and sequence alignment revealed the mutations also presented in this four vaccine strains.Conclusion Use the restriction fragment length polymorphisms analysis of DNA fragments of the open reading,frames 6 and 62 could be distinguished VZV wild-type strains and vaccine strain in clinical isolates in China.In order to find the adverse effect caused by vaccine from certain company's,analysis on the SNPs in ORFs 1,31,51 and 62 is needed.
2.High Density Culture of Embryoid Bodies Enhanced Cardiac Differentiation of Murine Embryonic Stem Cells

Objective To investigate the role of different culture densities of embryoid bodies (EBs) in cardiac dif-ferentiation of mouse embryonic stem cells (ESCs). Methods The mouse ESCs were cultured in hanging drops for 3 days, followed by another 2 days for suspension culture to form EBs. Suspended EBs of different densities (60 or 120 EBs/60 mm tissue culture dish) were transferred onto tissue culture plates. The cardiac specific troponin T (TnT) was detected by immu-nocytochemistry. The percentage of beating EBs was calculated at different time points. The mRNA expression of cardiac spe-cific transcriptional factors Nkx2.5, GATA4 and cardiac specific proteinsβ-MHC and ANF were detected by RT-PCR. The protein expressions of TnT and p38 were detected by Western-blot assay. Results Spontaneously beating EBs were posi-tively stained with TnT. There were significantly higher percentage of beating EBs, higher gene expression levels of Nkx 2.5, GATA4,β-MHC and ANF and higher protein expression of TnT in high density culture group than those of low density cul-ture group (P<0.05). Furthermore, there was significantly higher activity of p38 pathway in high density culture group than that of low density culture group. And the percentages of beating EBs and TnT protein expression were decreased by p 38 pathway inhibitor SB203580. Conclusion The culture density of EBs is important in regulating the cardiac differentiation of ESCs. The high cell-density density culture of EBs enhances the cardiac differentiation of ESCs, which might be mediated by p38 signaling pathway.

Based on quantum mechanics, rotational spectroscopy is a branch of fundamental science to study the rotational spectra of molecules, free radicals and ions, and has great applications on radio astronomy and many analytical fields such as atmospheric remote sensing. In this paper we reviewed the basic theory of rotational spectroscopy, two different types of Fourier transform microwave spectrometers, some typical spectroscopic analysis, and future prospect of the microwave spectroscopic techniques as well.

6.Human papillomavirus as an independent predictor in oral squamous cell cancer.

AIMThere is an increasing evidence for the role of high risk human papillomavirus (HPV) in the pathogenesis of oral squamous cell carcinoma (OSCC). The purpose of this study is to evaluate the relevance of HPV infection to the survival and prognosis of OSCC.
METHODOLOGYFifty-two patients with OSCC were followed from 4 to 88 months with a median of 50.7 months. HPV DNA was identified in formalin-fixed, paraffin-embedded tumor specimens by nested PCR with MY09/MY11 and GP5+/GP6+ primer pairs and the HPV genotype was determined by direct DNA sequencing. Association between the HPV status and risk factors for cancer as well as tumor-host characteristics were analyzed. Survival curves were calculated by the Kaplan-Meier method and analyzed using the log-rank test.
RESULTSHPV was found in 40.4% of the tumors with HPV16 accounting for 63.5%, HPV18 for 30.8%, HPV6 for 3.9% and HPV11 for 1.8%. No infection with more than one HPV genotype was detected. HPV infection was significantly associated with poor histological grade, TNM stage I-II, alcohol usage and no smoking status. Multivariate analysis showed that HPV had an independent prognostic effect on the overall survival after adjusting other confounding factors such as histological grade, TNM stage and tobacco usage. The presence of HPV was significantly correlated with a better survival in patients with OSCC.
CONCLUSIONHPV infection can act as an independent predictor for the survival and prognosis of OSCC.

7.The application of deltopectoral flaps with delayed expansion in repairing large scars on face.

OBJECTIVETo evaluate the effect of use of deltopectoral flaps with delayed expansion in repairing large scar on face.
METHODSThirteen patients with large scars on face received above-mentioned surgery. The cutaneous branches of thoracoacromial artery and transverse cervical artery of patients were divided when the expander was implanted. As a result, the blood supply of expanded deltopectoral flaps principally depended on the second and third intercostal branches of internal thoracic artery, and a delaying effect was achieved. The distribution of vascular net in the flap and the state of blood supply of the flap were observed to evaluate the effect of delaying.
RESULTSThe vascular net of expanded flap increased and thickened. No blood supply disorder occurred at the distal end of the transferred flap. Patients were followed up for 4 to 18 months. The outline of the burn side of face was in accord with the opposite side, which was not fat and clumsy. The complexion of the flap was similar to that of normal skin of the face, and the flap was soft in texture.
CONCLUSIONSThe application of delaying can better ensure the blood supply of transferred deltopectoral flap. The result of applying delayed expansion of deltopectoral flaps in repairing large scars on face was good.

8.Effects of acupuncture on granulocyte apoptosis and expressions of apoptosis-associated genes in the ovary of perimenopausal rats.

OBJECTIVETo explore the mechanism of acupuncture for treatment of perimenopausal syndrome.
METHODSThe rats of perimenopausal syndrome were randomly divided into 3 groups, including an acupuncture group treated with acupuncture, a medication group with Gengnian'an, and a perimenopausal control group, with young rats used for a control group. Granulocyte apoptosis and expressions of Bcl-2 and Fas proteins in the ovary of the rat were detected.
RESULTSGranulocyte apoptosis increased significantly (P < 0.01), expression of Bl-2 proteins decreased significantly (P < 0.01) and expression of Fas proteins increased significantly (P < 0.01) in the ovary of perimenopausal rats as compared with the young rats; after acupuncture treatment, granulocyte apoptosis decreased significantly (P < 0.05), expression of Bel-2 proteins increased significantly (P < 0.05) and expression of Fas proteins decreased significantly (P < 0.01); after treatment of Gengnian'an, granulocyte apoptosis did not significantly change (P > 0.05), expression of Bcl-2 prteins increased significantly (P < 0.05) and expression of Fas proteins decreased significantly (P < 0.01).
CONCLUSIONAcupuncture can inhibit granulocyte apoptosis, up-regulate expression of Bcl-2 proteins and down-regulate expression of Fas proteins in the ovary of the perimenopausal rat.

9.Spindle cell lipoma/pleomorphic lipoma of soft tissue: a clinicopathologic analysis of 8 cases

Purpose To investigate the clinicopathological features, the diagnosis and differential diagnosis of spindle cell lipoma/pleomorphic lipoma (SCL/PL). Methods Eight cases of SCL/PL were studied by HE staining and immunohistochemistry. The literature was reviewed. Results Microscopically, the neoplasm consisted of spindle cells, mature adipocytes and thick rope-like collagen bundles in variable proportions with mucinous degeneration. The floret-like multinucleated giant cells were easy to be seen in the pleomorphic lipoma. Immunohistochemical analysis revealed that the tumor cells were positive for CD34 and vimentin. The Ki-67 was approximately 2%. The patients were followed up for 3 ~ 74 months, and no recurrence or metastasis occurred. Conclusion The SCL/PL is a special subtype of benign lipoma, surgery is considered the first choice, and outcome is favorable. The diagnosis and differential diagnosis of SCL/PL are important.
10.Effects of Flos Puerariae-Semen Hoveniae Compatibility on Liver Function and Pathologic Morphology in Rats with Alcoholic Liv-er Disease

OBJECTIVE To explore the de-alcoholic and hepatoprotective function of Flos puerariae-Semen hoveniae (2∶1) compatibility and determine the optimal dosage regimen.METHODS Chronic alcoholic liver injury animal models were es-tablished by gavages of 56°Red Star Erguotou in rats.Experimental animals were given different doses(3,6,12 g/kg)of Flos puerariae-Semen hoveniae (2∶1) extracts within different medication courses (4,8,12 weeks).The liver index,ALT, AST,TP,ALB and ALP in serum were detected.The liver pathological morphologies were also observed.RESULTS Chron-ic alcoholic liver injury models were successfully replicated.Flos puerariae-Semenhoveniae(2∶1)compatibility group of each dose displayed no therapeutic effect within four weeks.The level of ALT in Flos puerariae-Semen hoveniae (2∶1) compatibil-ity group of each dose significantly decreased(P<0.05~0.01)after eight weeks administration.After 12 weeks,Flos puer-ariae-Semenhoveniae(2∶1)compatibility groups showed significantly decreased AST and increased ALB(P<0.05).Low and high dose groups displayed reduced ALT (P<0.05) while the low dose group showed increased TP (P<0.01).The changes in liver pathological morphology were fairly consistent with relevant indicators of liver function.CONCLUSION Flos puerariae-Semen hoveniae (2∶1) compatibility group of each dose shows promising de-alcoholic and hepatoprotective effects after eight weeks of administration.The relevant indicators of liver function improve to varying degrees as the therapy time prolongs.Among them,low dose group performs better than medium and high dose groups.
