1.Influencing Factors in Acidity Test in the Quality Standard of Lactasin

Objective:To investigate the influencing factors in acidity test in the quality standard of lactasin. Methods:The acid-ity was tested under the conditions of milk from different sources, various inoculation amount and preservation conditions. Results:The acidity was not stable under the conditions of milk with different brands and batches. The current quality standard could not truly reflect the influence of preservation conditions on the sample quality. Conclusion:The unreliable acidity test results are caused by the quality problem of milk culture media and the current unreasonable standard method.
2.Ion-sensitive nanoemulsion-in situ gel system for ophthalmic delivery of flurbiprofen axetil.

The aim of the study is to prepare flurbiprofen axetil nanoemulsion-in situ gel system (FBA/NE-ISG) and observe its ocular pharmacokinetics, rheological behavior, TEM images, irritation and cornea retention. Production of nanoemulsion was based on high-speed shear and homogenization process, and then mixed with gellan gum to prepare FBA/NE-ISG. Rheological study showed that FBA/NE-ISG possesses strong gelation capacity and its viscosity and elastic modulus increases by 2 Pa*s and 5 Pa respectively when mixed with artificial tear at the ratio of 40 : 7. TEM images suggested no significant changes in particle morphology of the pre and post gelation. Good ocular compatibility of FBA/NE-ISG was testified by the irritation test based on histological examination. In vivo fluorescence imaging system was applied to investigate the characteristics of cornea retention, and the results indicated that the nanoemulsion-in situ gel (NE-ISG) prolonged the cornea retention time significantly since K(NE-ISG) (0.008 5 min(-1) was much lower compared with flurbiprofen sodium eye drops (FB-Na, 0.03% w/v) of which the K(Eye drops) was 0.105 2 min(-1), indicated that the cornea retention time of NE-ISG was prolonged significantly. Pharmacokinetics of FBA/NE-ISG in rabbit aqueous humor was studied by cornea puncture, the MRT (12.3 h) and AUC(0-12h) (126.8 microg x min x mL(-1)) of FBA/NE-ISG was 2.7 and 2.9 times higher than that of the flurbiprofen sodium eye drops respectively, which meant that the ocular bioavailability was improved greatly by the novel preparation. Therefore, FBA/NE-ISG can enhance the ocular bioavailability by prolonging drug corneal retention significantly. What's more, encapsulated by emulsion droplets prodrug flurbiprofen (FBA) instead of flurbiprofen (FB) can reduce the ocular irritation.

4.Ocular pharmacokinetics of dexamethasone cubosomes in rabbit aqueous humor

Aim: To prepare novel cubosome system for effective ocular drug delivery with dexamethasone(DEX) as model drug, and investigate its pharmacokinetic profile in rabbit aqueous humor. Methods: DEX cubosomes was prepared by the method of high-pressure homogenization, and its particle size was determined by the laser particle sizer, and the microstructure observed by cryo-TEM. In addition, Draize method was used to evaluate the ocular irritation of DEX cubosomes. Finally, aqueous humor microdialysis was utilized to evaluate its pharmacokinetics in rabbits. Results: Average diameter of DEX cubosomes was about 200 nm, and the cubic structure of the particles was evident under the cryo-TEM. It was indicated by Draize scores that this dosage form exhibited excellent ocular tolerance. Results of pharmacokinetic profiles in aqueous humor showed that AUC_(0→240) and c_(max) of the rabbit group administered with DEX cubosomes were significantly higher than those of the control group( DEX sodium phosphate eye drops), with AUC_(0→240) of the formulation Fl( 10% oil content) and F2(20% oil content) is being about 1. 8 and 2. 9 times higher than those of the control group, respectively( P <0. 05). Conclusion: The novel ocular drug delivery system of DEX cubosomes was capable of increasing significantly the drug concentration in aqueous humor, and improving the ocular bioavailability.
5.Transpedicular impacted grafting after bone loss calculation in the treatment of thoracolumbar compression fracture:a biomechanical study

6.Usefulness of CT in Diagnosis of Acute Focal Bacterial Nephritis

ObjectiveTo evaluate the value of CT in diagnosis and discrimination of acute focal bacterial nephritis(AFBN). Methods 6 cases of AFBN were reported which were all performed with ultrasound and CT scan.4 of 6 cases were observed by delay contrast enhanced CT examination. ResultsCT scan showed wedge-shaped masses with thickened Gerota fascia.The delayed contrast enhanced CT revealed more clear shapes and verges of the lesions than immediate contrast enhanced CT. ConclusionCT scan is of significant value in diagnosis and discrimination of AFBN with overall review of documents and statistic analysis.
7.Advantages and Disadvantages of Bilingual Teaching in Medical Biochemistry

8.Efficacy of celecoxib in the treatment of peri-operative pain of orthopaedic patients

[Objective]To observe the effect and the safety of the COX-2 inhibitor Celecoxib as the analgesia in orthopaedic post-operative patients.[Method]Sixty-four operative patients were selected in all between 2004~2005 in hospital,and divided into two groups randomly.Celecoxib was used as the postoperative analgesia in the experiment group and the "Patient-controlled analgesia" in the control group.The administration of Celecoxib: 8-12 hours before operation generally,just before the abrosia,and 6 hours after operation when patients can take food.drug withdrawal was depended on the complex of the operation and the severety of pain in 3 to 5 days.Dosage:400 mg first time,if a complicated operation,larger dose would be given.VAS score,adverse reaction and the satisfaction of patients were observed.[Result]The effect of Celecoxib is similar as the PCA,with less adverse reaction and more satisfaction.[Conclusion]Celecoxib has a satisfactory effect and safety as a postoperative analgesia,and is suitable to be a peri-operative analgesia of orthopaedic patients.
9.Effects of polyporus polysaccharide on activity and mRNA expression of inducible nitric oxide synthase in peritoneal macrophages of mice

OBJECTIVE: To study the mechanisms of the antitumor and immunoregulation functions of polyporus polysaccharide (PPS). METHODS: The production of nitric oxide (NO), the activity and mRNA expression of inducible nitric oxide synthase (iNOS) in peritoneal macrophages of mice administered with different dose of PPS were observed by Griess reaction, fluorimetry assay and RT-PCR, respectively. RESULTS: PPS could elevate the iNOS activity with dose-dependence and stimulate the iNOS mRNA expression of peritoneal macrophages in mice. CONCLUSION: The regulation of PPS on the production of NO in peritoneal macrophages of mice may occur at transcriptional level of iNOS. This indicates that the mechanism of PPS's antitumor and immunoregulation functions may be related to increasing NO output of macrophages through stimulating iNOS's denovo synthesis.
10.Effects of lentinan on nitric oxide production and intracellular glutathione in mouse peritoneal macrophages

Object To investigate the mechanisms of action of lentinan (LTN) activated mouse peritoneal macrophages to produce nitric oxide (NO). Methods The effects of LTN on NO output and intracellular glutathione (GSH) in mouse peritoneal macrophages and the correlation of them were studied. Results (1) LTN can significantly increase production of NO in mouse peritoneal macrophages, decrease level of intracellular GSH followed increase of NO production. (2) The above effect can be blocked efficiently by the NO production inhibitors. (3) NO production can be inhibited by GSH lowering drugs. Conclusion LTN increase NO production with depletion of intracellular GSH in the activated mouse peritoneal macrophages. It suggested that intracellular GSH plays an important role in regulation of NO production and protection of host cells from cytotoxic attack induced by NO.
