2.Stathmin: a potential tumor biomarker
Tumor 2010;(1):73-76
Stathmin, a ubiquitously expressed cytosolic protein(Mr=19×10~3), is also called oncogene protein 18 (Op18). Stathmin is involved in the assembly of microtubule (MT) and spindle by binding the tubulin protein. It plays a key role in cell proliferation, differentiation, regeneration, and migration and has regulatory effects on the signal transduction. Recently, it is reported that stathmin is overexpressed in a variety of human malignancies. It induces tumor cell migration and invasion by regulating MT depolymeri-zation. Its post-translational modification influences the interaction with p53 protein and is involved in the initiation and progression of malignant tumor. Stathmin alone or in combination with chemotherapeutics has been used for tumor therapy. The internal association of stathmin with cancer etiology is still unknown. So, further studies are needed to determine the role of stathmin as potential tumor biomarker and a drug target in tumor therapy.

Journal of Integrative Medicine 2007;5(5):555-8
OBJECTIVE: To investigate the changes of neuropeptides content in a rat model of spleen-deficiency syndrome. METHODS: Rats were randomly divided into three groups: normal group, untreated group and treatment group. There were ten rats in each group. Spleen-deficiency syndrome was induced by intragastrically administration of rhubarb concentrate in rats of the untreated and treatment group. The rats in the treatment group were intragastrically administered with Sijunzi decoction, a traditional Chinese compound herbal medicine for strengthening spleen and supporting qi. Normal saline was intragastrically administered to the rats in the normal and untreated groups. The somatostatin (SS) and cholecystokinin-8 (CCK-8) content in hypothalamus and colons was detected by the method of radioimmunoassay and the D-xylose content in urine was also detected. RESULTS: The CCK-8 content in hypothalamus of the untreated group was higher than that of the normal group (P<0.05). SS and CCK-8 content in colons of the untreated group was higher than that of the normal group (P<0.05). The CCK-8 content in colons of the untreated group was lower than that of the treatment group (P<0.05). The excretion rate of D-xylose in the untreated group was lower than that of the normal group (P<0.05). CONCLUSION: The disorder of neuroendocrine regulation in rats with spleen-deficiency syndrome may be chiefly responsible for "spleen-deficency and dysfunction of spleen" in traditional Chinese medicine.

Medical Journal of Chinese People's Liberation Army 2001;0(12):-
To construct a cDNA subtractive library of genes transactivated by hepatitis C virus core protein with suppression subtractive hybridization technique. mRNA was isolated from HepG2 cells transfected with pcDNA3 1(-)-core and pcDNA3 1(-) empty vector, respectively, then cDNA was synthesized. After restriction enzyme RsaI digestion, small sized cDNA were obtained. Then tester cDNA was divided into two groups and ligated to the specific adaptor 1 and adaptor 2, respectively. After tester cDNA was hybridized with driver cDNA twice and underwent two times of nested PCR, and then it was subcloned into T/A plasmid vectors to set up the subtractive library. Amplification of the library was carried out with E.coli strain JM109. The cDNA were sequenced and analyzed in GenBank with Blast search after PCR. The subtractive library of genes transactivated by HCV core protein was constructed successfully. The amplified library contained 233 positive clones. Colony PCR product showed that 213 clones contained 100~ 1 000 bp inserts. Sequence analysis was performed in 63 clones. Six of the sequences were unknown genes before. The full length sequences were obtained with bioinformatics method, which had been accepted by GenBank. It suggested that six novel cDNA sequences might be target genes transactivated by HCV core protein.
