Objective:To study the effect of different cell culture densities and sera on growth of mice bone marrow stromal cells in vitro.Methods:Bone marrow stromal cells were cultured in DMEM/F12 containing different cell densities and sera.The rate of adhered cells,cell viability by trypan blue test,proliferation of bone marrow stromal cells by MTT and cell morphological structure with light microscope were observed.Results:The cultured cells proliferated well in DMEM/F12 containing 10%~20% sera and the densities of 1~4?106/ml.It prolifrated better in horse serum than fetal bovine serum(P

3.Effect of angelica polysaccharide on bone marrow hematopoietic stem cells and intercellular adhesion molecule 1 on the surface of stromal cells in mice

BACKGROUND:Angelica polysaccharide has the function of enriching blood and promoting blood circulation, but how to improve the hematopoietic microenvironment and promote hematopoiesis has become one of the hot topics. OBJECTIVE: To investigate the effect of angelica polysaccharide on Sca-1+, CD34+ hematopoietic stem cells and intercellular adhesion molecule 1 level in bone marrow stromal cells in mice. METHODS:Mouse models of hemorrhagic anemia were made and then randomly divided them into control group, low-concentration angelica polysaccharide group and high-concentration angelica polysaccharide group. Angelica polysaccharide groups were intraperitoneally injected with the corresponding concentration of angelica polysaccharide (4, 6 mg/kg) for continuous 6 days, while the control group was injected with equal amount of normal saline. General conditions of the model mice were observed. Peripheral blood red cells were counted by automatic blood cell analyzer. Bone marrow hematopoietic stem cels were counted by magnetic bead sorting technique and flow cytometry. Proliferation of bone marrow stromal cells was detected by MTT method. Expression of intercellular adhesion molecule-1 in bone marrow stromal cells was detected by flow cytometry. RESULTS AND CONCLUSION:Angelica polysaccharide had no obvious effect on the body mass of hemorrhagic anemia mice (P > 0.05). The number of peripheral blood cells, the percentage of bone marrow CD34+ and Sca-1+ cells, the number of bone marrow stromal cells and the level of intercellular adhesion molecule 1 were ranked as follows: high-concentration group > low-concentration group > control group (P < 0.05). To conclude, angelica polysaccharide could promote the proliferation of bone marrow stromal cells and increase the expression of intercellular adhesion molecule 1, thereby promoting the proliferation of hematopoietic stem cells and regulating their functional activities.
4.Value of color Doppler ultrasound-guided percutaneous transhepatic cholangiol drainage

Objective To evaluate the clinical value of color Doppler ultrasound-guided percutaneous transhepatic cholangiol drainage(PTCD) for treating obstructive jaundice. Methods 248 cases of PTCD were performed in 224 patients under the guidance of color Doppler ultrasound. 118 PTCD cases were carried out passing through the inferior segmental biliary duet of Left laterallobe (Left drainage mode). Right lobe drainage mode (right drainage mode)was conducted in 130 patients. Left plus right lobe drainage mode in 16 patients. Results The total success rate was 99.59 %(247/248), and the success rate of the firs time PTCD was 92.33 %(229/248). The success rate of left drainage mode was significantly higher than righl lobe drainage (96.61% vs 88.46 %, P <0.05). Conclusion The technique of color Doppler ultrasound-guided PTCD is an efficient method in obstructive jaundice therapy, with the advantages of safety, convenience, minimal invasion, and higher success rate.
5.Comparison of intrarectal filling with water and idling in rubber sheath with water in endoluminal ultrasonography for preoperative staging of rectal tumors

Objective To compare the value of intrarectal filling with water and filling in rubber sheath with water in endoluminal uhrasonography for preoperative staging of rectal tumors. Methods 215 cases of rectal tumors were divided into two groups. 120 cases using the method of filling in rubber sheath with water and 95 cases using intrarectal filling with water were diagnosed with uhrasonography. The results were compared with corresponding pathologic results for the judgment of their accuracy. Results The accuracy for the diagnosis of infiltration depth was 60.0 % in the first group and was 73.7 % in the second. The difference was statistically significant (P <0.05). The accuracy for diagnosis of metastases of perirectal lymph nodes was 82.5 % in the first group and was 80.0 % in the second. The difference has no statistical significance (P >0.50). Conclusion Intrarectai filling with water is better than filling in rubber sheath with water in the diagnosis of infiltration depth of rectal tumors, while there is no difference in the diagnosis of metastasis of perirectal lymph nodes.
6.Analysis of ultrasonic characteristics of primary breast lymphoma

Objective To analyze the ultrasonic imaging characteristics of primary breast lymphoma and their significance for diagnosis.Methods Data of ultrasonograph from 21 cases of primary breast lymphoma were reviewed, and all cases had been certificated by pathology.Results All of 21 cases with primary breast lymphoma were solid mass.The contours of 16 cases were regular, and the boundaries of 9 cases were clear.20 cases were hypoechoic masses, besides 3 cases with lower echo like liquefactive degeneration.The internal echoes of 19 cases were disorderly, part of them with hyperechoic trabs.The rear echoes of 17 cases were neither enhanced nor fading.The surrounding tissues of 19 cases were not changed.13 cases had more than 3 blood vessels on color Doppler imaging.Conclusion There are some ultrasound characteristics in primary breast lymphoma, which would be helpful in the diagnosis of primary breast lymphoma.
7.Impact factors investigation in oral ultrasonic contrast agent on diagnosis of ulcerative type gastric neoplasms on T staging

Objective To explore the factors affecting diagnosis accuracy on T stage of oral ultrasonic contrast agent examination on ulcerative gastric neoplasms.Methods Data from 82 patients were analyzed who were pathologically diagnosed as ulcerative gastric neoplasms ultrasounds data of oral ultrasonic contrast agent before surgery were compared to postoperative pathology,analysis had been done on influence of the lesion site,size of the T staging on ulcerative gastric neoplasms.Results The diagnosis accuracy rate of T stage on lesser curvature of stomach and gastric antrum were 91.3 % (21/23) and 85.7 % (24/28),compared with the pathological results were in good concordancy (Kappa =0.763,0.68,P < 0.05).The accuracy rate of T stage on cardiac lesions was 68.0 % (17/25),compared with the pathological results consistency in general (Kappa =0.446,P < 0.05).Further research on the effects of T stage accuracy would be necessary.The accuracy rate ofT staging on tumor size ≤ 5.0 cm group and > 5.0 cm were 92.3 % (36/39) and 72.1% (31/43),the difference was significant in two groups (x2 =5.591,P < 0.05).Conclusion Oral ultrasonic contrast agent application is more accurate on lesser curvature of stomach,gastric antrum and lesions size ≤ 5.0 cm.

9.Comparative study between cardiac catheterization intervention therapy and transthoracic small incision surgery for closure of congenital atrial septal defect by domestic occluder with echocardiographic monitoring

Objective:To evaluate the safety of cardiac catheterization intervention therapy and transthoracic small incision surgery in the occlusion bydomestic occluder under echocardiography guiding in patients with atrial septal defect (ASD).Methods:A total of 1 080 patients with ASD in the occlusion by domestic occluder were analyzed retrospectively,and the interventional treatment were performed in 734 cases through cardiac catheterization intervention therapy and 346 cases through transthoracic small incision surgery.The patients undergone cardiac catheterization intervention therapy were guided under the digital substraction angiography (DSA) and were monitored by transthoracic echocardiography (TTE) in the whole interventional process,and the efficacy was evaluated with TTE.The occlusion of transthoracic small incision surgery was guided under the transesophageal echocardiography (TEE),which was used to monitor the position of occluder and evaluate the efficacy immediately.Results:Two kinds of intervention in the occlusion by domestic occluder had achieved satisfactory results in patients with ASD.There was no statistically difference in the longest size of ASD between the 2 intervention methods,while there were statistically differences in the ratio between ASD longest diameter and atrial septal length,and the size of the occlusion,and the disparity between the size of the occluder and ASD longest diameter (D value),respectively (all P<0.05).When the size of arithmetic mean of the ASD was <30 mm,the success rate of the 2 methods was both 100%.When the size of arithmetic mean of the ASD was ≥ 30 mm,the success rate was 100% in the transthoracic small incision surgery and 50% in the cardiac catheterization intervention therapy.Conclusion:Domestic occluder is safe.Compared with the imported one,its cost is lower.When the size of the defects is same,the occlusion is smaller in the transthoracic small incision surgery compared with that in the cardiac catheterization intervention therapy.When the size of arithmetic mean of the ASD is ≥ 30 mm,the success rate of the transthoracic small incision surgery is higher compared with the cardiac catheterization intervention therapy.When the cardiac catheterization intervention therapy fails,the transthoracic small incision surgery may be a better choice.
10.Effect of pain intervention on limb function exercises in patients with peripheric fractures of knee joint

Objective To explore the effect of pain intervention on limb function exercises in patients with peripheric fractures of knee joint.Methods 60 patients with peripheric fractures of knee joint were randomized in equal number into the observation group and control group.Both groups took functional exercises for affected limbs.Besides,the former and latter groups were administered with celecoxib at a dosage of 200mg twice a day from pre-operation to discharge and after operation,respectively.The two groups were compared in terms of pain degree at different time points as well as the functional recovery of affected limbs.Results The observation group was lower in pain scores than the control group at hours 6,8,12,24,36 and 48.The active and passive motions of the affected limbs in the observation group were significantly better than those in the control at days 1,2,3,4 and 5(all P<0.001).Conclusion Pain intervention with celecoxib before operation may help patients to take functional exercises as early as possible,promoting the rehabilitation of functions of affected limbs.
