Objective To explore the effect of repetitive transcranial magnetic stimulation (rTMS) on behaviors and hippocampal glucocorticoid receptor (GR) protein expression in chronic stress depression model rats and the possible antidepressant mechanism of rTMS. Method Seventy-five male Sprague-Dawley rats were randomly divided into the blank control group (n=15) and the stress-induced group (n=60). Singly housing and chronic unpredictable mild stress (CUMS) were used to induce the depression model in stress-induced group. Forty-five CUMS rats were selected and ran?domly divided into rTMS group (receiving 10 Hz rTMS intervention for 3 weeks), sham group (receiving pseudo rTMS treatments for 3 weeks) and depression group (with no further treatment). Body weight measurements and performance in the sucrose consumption and forced swimming test (FST) were evaluated before modeling, after modeling and after inter?vention. The GR protein and GR mRNA expression level in the hippocampus were examined after intervention. Results Compared with control group, the body weight growth rate and the sugar water preference were significantly lower in stress-induced group (P<0.01), and the immobility time of FST was significantly longer (P<0.01). After the 3-week rTMS intervention, the body weight growth rate and the sugar water preference in rTMS group, which were insignificantly differ?ent from control group (P>0.05), were higher than those in sham group and depression group (P<0.01). The immobility times of FST in rTMS group and control group were shorter than sham group and depression group (P<0.01). Compared with rTMS group and control group, GR and GR mRNA expression levels in the hippocampus were significantly reduced in sham group and depression group (P<0.01). Conclusion rTMS can improve depression behavior of CUMS rats, which may be associated with upregulation of GR expression in the hippocampus.

OBJECTIVE:To provide reference for promoting the further development of pharmaceutical service in chain phar-macies and the regulation of related departments. METHODS:Field investigation and questionnaire survey were used to investigate the basic situation of pharmaceutical care in 5 chain pharmacies in Chengdu city and analyze the results. RESULTS:In terms of core service,it showed that 2 pharmacies had no suspension of pharmacists license and the pharmacist was not on-the-job;there was significant difference in the medication guidance frequency of pharmacists in those pharmacies(P<0.05);more than 50% sur-veyed consumers received medication guidance when buying drugs in pharmacy B,C and E. In terms of perceived service,3 phar-macies existed illegal drug advertising and 3 existed illegal promotion;drug partitions of all pharmacies were good;service archive material showed best in pharmacy C and worst in pharmacy D;in service advisory zone setting, setting of independent advisory zone was worse. In terms of extended service,5 pharmacies had different preferential ways for memberships except for the prefer-ence in membership day;the unbundling service in those pharmacies was worse, and no pharmacies had no less than 10 unbun-dling varieties. CONCLUSIONS:Pharmaceutical service in chain pharmacies in Chengdu city is barely satisfactory. Pharmacy should strengthen the assessment and management of pharmacists and improve pharmaceutical service, and regulatory authorities strengthen the supervision of advertising and promotion of drugs. What’s more,it is suggested to establish cloud supervision plat-form with the internet information technology, carry out remote service and cloud service and strengthen brand marketing service on the internet to improve the pharmaceutical service levels in chain pharmacies.

Objective To investigate the signalling events follow the activation of RAGE in podocytes. Methods AGE and CML generated dichloroflurescin-sensitive intracellular ROS were measured by confocal microscopy. The activation of MAP kinases family were studied using Western blotting. MCP-1 mRNA expression was detected by semi-quantitative RT-PCR. Results Basal ROS was located in nucleus of starvation podocytes. AGE and CML rapidly generated intracellular ROS in podocytes. NAC pre-treated podocytes suppressed basal and inducible ROS generation and antibody for RAGE suppressed the inducible ROS. Blockage of ROS induced by NAC suppressed the expression of CML and H_2O_2-induced MCP-1. Phosphorylated extracellular signal-regulated kinase (ERK) was found in CML incubated podocytes at 10 min and was prevented by NAC or AFC. PD98058 Pre-treated podocytes partially inhibited the expression of CML-induced MCP-1 mRNA. No evidence were showed that p38 MAPK, SAPK/JNK, PI3K and PKC were involved in the signal transduction. Conclusion Activation of RAGE induces MCP-1 expression in podocytes via ROS-ERK signalling pathway.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2005;25(4):442-4, 447
To study the effect of tumor necrosis factor-related apoptosis inducing ligand (TRAIL) on PC-3M cell line, PC-3M cell line was incubated with gradient concentrations of TRAIL for 4--24 h. Annixin-V fluorescence staining and TUNEL method were employed to detect the apoptosis of PC-3M cells. The morphology of apoptotic PC-3M cells was observed by electron microscopy. The relationship between TRAIL concentrations and the percentage of apoptotic cells was evaluated by flow cytometry. The proliferation inhibitory ratio was calculated by using MTT colorimetry. Our results showed that apoptosis of PC-3M cells could be induced by treatment with TRAIL for at most 4 h. The results of flow cytometry and MTT colorimetry demonstrated a time- and concentration-dependent relationship between cell apoptosis rate and TRAIL concentration. It is concluded that apoptosis of PC-3M cells can be induced by TRAIL. Because of the selective killing effect of TRAIL on tumor cells, it may become a potential alternative for the treatment of advanced prostate cancer.
