1.Role of receptor-interaction proteins in phenethylisothiocyanate-induced apoptosis in human leukemia cells in vitro

Objective To explore the role of receptor-interaction proteins(RIP) in phenethylisothiocyanate(PEITC)-induced apoptosis in human leukemia cells.Methods U937 cells were exposed to PEITC at various concentrations of 0,2,4,6 and 8 ?mol/L for 3 and 6 h,or at 8 ?mol/L for different time intervals of 1,3,6,9,12 and 24 h.Cells were stained with Annexin V/PI,and apoptosis was determined by using flow cytometry.Total protein extracts were prepared and subjected to Western blot assay using antibodies against poly(ADP-ribose)polymerase(PARP),Caspase-3,Caspase-8,and RIP.?-actin was used to ensure equivalent loading.Results After exposure to PEITC at concentrations of 2,4,6 and 8 ?mol/L for 3 h,U937 cells showed an increase in cell apoptosis(9% ,25% ,44% and 56% respectively).While when the time expanded to 6 h,the amount of apoptotic cells was increased to 16% ,28% ,51% and 82% respectively.A dose-dependent manner was seen when U937 cells were exposure to PEITC at different concentrations.When U937 cells were treated by PEITC at concentration of 8 ?mol/L for 1,3,6,9,12,and 24 h,the apoptotic cells covered 12% ,57% ,79% ,86% ,90% ,and 91% of all cells respectively,indicating a time-dependent manner.Western blot assay showed that PEITC caused the activation/cleavage of Caspase-3 and-8,and improved the PARP cleavage,and also decreased the expression of RIP.PEITC-induced apoptosis was proceeded by degradation of RIP.Conclusion Our results indicate that PEITC induces the apoptosis in human leukemia cells through a process that involves death receptor pathway,and RIP may play an important role in the apoptosis.

The ancient books on traditional Chinese medicine in Jiangsu Province were recorded and analyzed, fol-lowed by an introduction of their holdings in Jiangsu Province, and suggestions were put forward for their systemiza-tion, such as enforcing the role of special libraries of traditional Chinese medicine, promoting the general survey and report of ancient books on traditional Chinese medicine, and compiling《Jiangsu union catalogue of ancient books on traditional Chinese medicine》.
3.Evaluation of the agreement of preoperative ultrasonography, CT, intraoperative ultrasonography and intraoperative palpation in the diagnosis of liver tumors

Objective To assess the agreement of preoperative ultrasonography (US), CT, intraoperative ultrasonography(IOUS) and intraoperative palpation in the diagnosis of liver tumors. Methods The results of preoperative US,CT, IOUS and intraoperative palpation in the diagnosis of liver tumors in 70 patients were reviewed,and the agreement of them were evaluated. Results There was a moderate level of agreement between US and the pathology and follow-up in the diagnosis of liver tumors (Kappa = 0.63). There was a high level of agreement among CT, IOUS and the pathology and follow-up(CT vs the pathology and follow-up, Kappa = 0.72;IOUS vs pathology and follow-up, Kappa = 0. 89;CT vs IOUS, Kappa =0.53), whereas the level of agreement between intraoperative palpation and the pathology and follow-up (Kappa =0.38), US and intraoperative palpation (Kappa = 0.23), US and IOUS (Kappa = 0.23) was low. Conclusions In the assessment of liver tumors,CT and IOUS have a high agreement. Although the agreement of US and intraoperative palpation were low, they were still indispensable. Four methods were integrated, will contribute to the diagnosis and treatment of liver tumors.
4.CXCR4/CXCL12 axis and relapsed leukemia

The binding complex of chemokine receptor CXCR4 and its specific ligand CXCL12 triggers the downstream signaling pathway to form biological axis of CXCR4/CXCL12. The axis plays an important role in homing, repopulation of hematopoietic stem cells, as well as maintenance of normal hematopoiesis and homeostasis of hematopoietic microenvironment. Meanwhile, CXCR4/CXCL12 axis is also closely related with relapsed leukemia, since it may promote survival, proliferation, metastasis and drug resistance in leukemic cells.The expression level of CXCR4 could be treated as an important marker for predicting prognosis in patients wizh leukemia, therefore, the combination of CXCR4 inhibitors with routine chemotherapy may represent a powerful approach for the treatment of leukemia.
5.Intrauterine devices, subcutaneous contraceptive implants and tubal contraception devices made of various types of materials

BACKGROUND:Aclear understanding on the characteristics of contraceptive materials, techniques and effects on reproductive health is of great significance to reduce unwanted pregnancies and the abortion rate. OBJECTIVE: To summarize the progress, current situation and usage of the contraceptive materials, and to comprehensively analyze the characteristics, effects and adverse effects of intrauterine devices, subcutaneous contraceptive implants and tubal contraception materials, so as to provide the basis for women of childbearing age to choose safe and reliable contraceptive materials. METHODS:The related articles were retrieved from China National Knowledge Infrastructure and PubMed database during 1985 to 2015 by taking “intrauterine device, subcutaneous contraceptive implant, tubal contraceptive material, reproduction, health, contraceptive method” as the Chinese key words and taking “intrauterine contraceptive device, subcutaneous contraceptive implants, oviduct contraceptive, materials, reproductive health, influences” as the English key words. Finaly 44 articles were included in result analysis. RESULTS AND CONCLUSION:Intrauterine devices have a rapid development and wide audience, which are reliable, and mature to have a higher awareness and usage rate among females who have bred children. Subcutaneous contraceptive implants are characterized as strong affinity, constant release, good release effect, simple and convenient operation. Oviduct contraceptive suppository materials have strong biological nature, memory and tolerability, as wel as possess non-invasive, reusable, safe, long-term advantages. To seek safe, reliable contraceptive materials and methods with no adverse reactions is of great significance to ensure women’s reproductive health.

Objective To evaluate the effects of SHENG JI YU HONG herbs loading on modified gelatin sponges on the wound healing of mechanical trauma.Methods The round size of 1.8cm diameter of full thickness back skin was cut mechanically and covered with petrolatum gauzes,modified gelatin sponges,SHENG JI YU HONG plasters and SHENG JI YU HONG herbs loading on modified gelatin sponges respectively.On day3,day7 and day14 after model mading,the wound surface healing rate,the amount of hydroxyproline,hemoglobin,the number of capillaries and the cells with positive rhVEGF165 by immunochemical method inside granulation of the wound were detected individually.Results On day3,the wound surface healing rate and the number of capillaries and the cells with positive rhVEGF165 of SHENG JI YU HONG herbs loading on modified gelatin sponges were significant enhanced than those in the other groups.On day14,all the experimental indicators of SHENG JI YU HONG herbs loading on modified gelatin sponges were better than those in the other groups with P
8.Effect of reducing the radiation pneumonitis by abdominal breathing exercises in patients with chest malignant tumors

Objective To evaluate the prophylactic effect of abdominal breathing exercises on radiation pneumonitis in patients with chest malignant tumors.Methods 100 patients with chest malignant tumors were given radiotherapy and divided with random digit table into the exercise group and the control group with 50 patients in each group.The incidence rate of radiation pneumonitis in both groups was compared.The prophylactic effect of abdominal breathing exercises on radiation pneumonitis was evaluated.Results The incidence of radiation pneumonitis in abdominal breathing exercise group was significantly reduced [34% (17/50) vs.56% (28/50),Z=2.397,P<0.05],the incidences of grade Ⅰ [22% (11/50) vs.30% (15/50)] and grade Ⅱ [12% (6/50) vs.18% (9/50)] radiation pneumonitis decreased,and no grade Ⅲ and Ⅳ radiation pneumonitis occurred.Conclusions The incidence of radiation pneumonitis in patients with chest malignant tumors treated with radiation therapy may be decreased by abdominal breathing exercises,which may be widely applied in patients treated with radiation therapy.
9.Preparation, crystallization and preliminary X-ray diffraction analysis of PH1948, predicted RNA methyltransferase from Pyrococcus horikoshii.

RNA methyltransferase is responsible for transferring methyl and resulting in methylation on the bases or ribose ring of RNA, which existed widely but mostly remains an open question. A recombinant protein PH1948 predicting RNA methyltransferase from Pyrococcus horikoshii OT3 has been crystallized. The crystals of selenomethionyl PH1948 belong to space group C2, with unit-cell parameters a=207.0 A, b=43.1 A, c=118.2 A, b=92.1 degrees , and diffract X-rays to 2.2 A resolution. The V(M) value was determined to be 2.8 A3/Da, indicating the presence of four protein molecules in the asymmetric unit.

10.Alleviative effects of green tea polyphenols on cyclosporine A-induced inhibition of vasorelaxation

BACKGROUND: Vasorelaxation plays an important role In the occurrence of cyclosporine A (CsA)-induced nephrotoxlcity.OBJECTIVE: To observe the alleviative effects of green tea polyphenols (GTP) on CsA-induced inhibition of vasorelaxation and the underlying mechanisms.METHODS: Sprague-Dawley rats were randomly and evenly divided into four groups: CsA, control, CsA + GTP, and GTP. After 5 weeks of drug treatment, blood urea nitrogen (BUN) and creatinine (Cre) levels were determined. Then the thoracic aorta rings were mounted on a bath system, and acetylcholine was used to induce vasorelaxation. The effects of L-NAME and indomethacin and the denuded vasorelaxation were evaluated.RESULTS AND CONCLUSION: The BUN and Cre levels in the CsA group were significantly higher than those in the control group (P < 0.05). The maximal response (Emax%) for acetylcholine-induced vasorelaxation in the CsA group was significantly lower than that in the control and GTP groups. After pretreatment with L-NAME, vasorelaxation was significantly lower in the CsA,CsA+GTP and GTP groups than in the control group. After pretreatment with indomethacin, vasorelaxation was significantly higher in the control, CsA +GTP, and GTP groups than in the CsA group. The level of nitric oxide metabolites in the vascular tissue in the CsA group was significantly lower compared with other groups. The results demonstrated that CsA can decrease nitric oxide levels in vascular tissues and induce abnormal endothelium-dependent vasorelaxation, which is mediated by nitric oxide pathway.
