Journal of Acupuncture and Tuina Science 2004;2(3):24-25
Selected mainly were acupoints in the adjacent areas and along the meridians, assisted by ashi points. Fengmen (BL 12), Geshu (BL 17) and Xuehai (SP 10) were selected for migratory arthralgia;Shenshu (BL 23) and Guanyuan (CV 4) for cold-aggravated arthralgia; Pishu (BL 20), Zusanli (ST 36)and Yinlingquan (SP 9) for damp arthralgia; Dazhui (GV 14) and Quchi (LI 11) for heat-induced arthralgia. Acupuncture in combination with far infra-red radiation was performed.76 patients with rheumatoid arthritis were treated and 74 cases with herb therapy as control. The result showed that there was statistically significant difference in curative effect between the 2 groups, χ 2=10.18, P< 0.01, indicating that the curative effect was significantly better in the treatment group than in the control group. Statistics showed that there was no significant difference in curative effect among the 4 syndromic types, namely migratory arthralgia, cold-aggravated arthralgia, damp-induced arthralgia and heat-induced arthralgia. But there was a statistically significant difference in curative effect between the above 4 syndromic types and deficiency-type arthralgia, indicating that the curative effect in deficiency-type arthralgia was worst.

3.MiR-26b regulates invasion and migration of lung cancer cells through targeting hENTI depending on RhoA/ROCK-I pathway
Journal of Central South University(Medical Sciences) 2017;42(7):755-761
Objective:To investigate the effect of miR-26b on the invasion and migration of lung cancer cell and to explore its mechanism. Methods:qPCR was used to detect the expression of miR-26b in lung cancer.Luciferase reporter gene was used to detect interaction between miR-26b and hENT1.Transwell assay was used to detect invasion ability after treatment of miR-26b mimics.Scratch assay was used to detect migration ability after treatment of miR-26b mimics. The expressions of hENT1,ROCK-I and RhoA were detected by Western blot. The changes of cytoskeleton after miR-26b mimics treatment with phalloidin were observed. The effect of miR-26b mimics on the tumor size and volume of lung cancer was determined by subcutaneous tumor formation in nude mice.Results:MiR-26b expression was significantly reduced in lung cancer. With the progress of lung cancer, the expression of miR-26b was reduced. With the progress in differentiation of lung cancer, the expression of miR-26b was decreased.Decrease of miR-26b was associated with lung cancer lymph node metastasis.HENT1 was the direct target of miR-26b;miR-26b regulated the invasion and migration ability of human lung carcinoma A549 cells.MiR-26b regulated the expression of hENT1,ROCK-1 and RhoA.After the treatment with miR-26b mimics, the F-actin staining was significantly reduced, whereas the formation of wrinkles and the formation of pseudopodia were significantly reduced.Subcutaneous tumor formation in nude mice showed that miR-26b mimics treatment significantly reduced the tumor size and mass.Conclusion:MiR-26b plays a role in tumor suppression in lung cancer.miR-26b can regulate the invasion and migration ability of lung carcinoma A549 cells by targeting hENT1 depending on the RhoA/ROCK-1 pathway.
4.Effects of Creatine Supplements on Contractile Properties of Skeletal Muscle in Elder Rats
Chinese Journal of Rehabilitation Theory and Practice 2013;19(6):527-531
Objective To investigate the effects of creatine supplement on contractile properties of soleus (SOL) and extensor digitorum longus (EDL) in elder rat. Methods 20 male Sprague-Dawley rats were randomly divided into control group and creatine supplement group with 10 rats in each group. After 6 weeks, the content of creatine and phosphocreatine were detected by high performance liquid chromatography, and contractile properties were examined in SOL and EDL in vitro at 30 ℃. Results The content of creatine and phosphocreatine both in SOL and EDL were higher in the creatine supplement group than in the control group (P<0.05). In SOL, creatine supplements increased the maximum twitch tension (Pt), maximum titanic tension(P0), decreased time to maximum twitch tension (TPT), but only time from maximum twitch to half relaxation (TPR50) significantly decreased, compared with the control group. In EDL, creatine supplements significantly increased Pt, P0 and decreased TPT (P<0.05). But there was no significant difference in TPR50 between 2 groups (P>0.05). Creatine supplements significantly increased the fatigue resistance of the SOL and there was no significant effect on EDL. The force potentiation which occurred in EDL during the initial 20~30 s of the fatigue test significantly increased (P<0.05). Conclusion Creatine supplement is effective on the contractile properties of SOL and EDL in elder rats, but the effect is dependent on the fiber types.

The general information, vital signs, blood gas analysis, serum CC16 and sRAGE levels, duration of mechanical ventilation, length of ICU stay and prognosis during hospitalization were collected. The receiver operating characteristic (ROC) curve was drawn and the area under ROC curve (AUC) was calculated to evaluate the clinical value of CC16 and sRAGE lonely or combination in the diagnosis and prognosis of ARDS.Results:The duration of mechanical ventilation in the ARDS group was significantly longer than that in the non-ARDS group (days: 15.44±3.04 vs. 12.61±3.73, P < 0.01), and the hospitalization mortality was higher (38.0% vs. 9.0%, P < 0.01), but there was no significant difference in gender, age, body mass index (BMI), acute physiology and chronic health evaluation Ⅱ (APACHEⅡ) score or the length of ICU stay between the two groups. There were 62 ARDS patients survived and 38 ARDS patients died during hospitalization. The APACHEⅡ score of the death group was significantly higher than that of the survival group (23.55±2.83 vs. 19.40±4.10, P < 0.01), but there was no significant difference in age, BMI, oxygenation index, mean arterial pressure, duration of mechanical ventilation or the length of ICU stay between the two groups. The serum levels of CC16 and sRAGE in the ARDS group were significantly higher than those in the non-ARDS group [CC16 (mg/L): 38.78±14.70 vs. 21.87±2.45, sRAGE (pg/L): 2 470.95±288.70 vs. 2 013.22±131.15, both P < 0.01]; and the serum levels of CC16 and sRAGE of ARDS patients in the death group were significantly higher than those in the survival group [CC16 (mg/L): 42.02±10.81 vs. 30.52±9.47, sRAGE (pg/L): 2 638.34±324.07 vs. 2 279.91±163.70, both P < 0.01]. ROC curve showed that the AUC of CC16 and sRAGE in the diagnosis of ARDS alone were 0.859 [95% confidence interval (95% CI) was 0.808-0.911] and 0.821 (95% CI was 0.762-0.879), and the best cut-off values were 25.76 mg/L and 2 203.00 pg/L, respectively; the AUC of combined detection of CC16 and sRAGE was 0.932 (95% CI was 0.900-0.965) with the sensitivity of 89.0% and the specificity of 87.6%. The AUC of CC16 and sRAGE in predicting the death of patients with ARDS during hospitalization were 0.747 (95% CI was 0.651-0.843) and 0.819 (95% CI was 0.737-0.902), and the best cut-off values were 32.95 mg/L and 2 554.50 pg/L, respectively; the AUC of combined detection of CC16 and sRAGE was 0.900 (95% CI was 0.828-0.972) with the sensitivity of 88.7% and the specificity of 84.5%. Conclusions:Serum CC16 and sRAGE have clinical value for the diagnosis and prognosis of ARDS. The combined detection of them is superior to individual detection for early prediction of ARDS and prognosis.

Chinese Journal of Pathophysiology 2016;32(9):1627-1634
AIM: To observe the anti-apoptosis effect of liraglutide on the islet through microRNA-375 (miR-375) for providing additional pharmacodynamic evidence for its clinical application.METHODS: For in vivo study, C57BL/KsJ-db/m mice aged 8 weeks served as normal control group.A total of 40 male genetically diabetic C57BL/KsJ-db/db mice at the same age were randomly divided into diabetic control group (the db/db mice were injected subcutaneous-ly with equivalent amount of saline) and liraglutide group (the db/db mice were injected subcutaneously with liraglutide at dose of 300 μg? kg-1? d-1 ).After 8 weeks of administration, body weight (BW) was measured and blood was collected for detection of fasting blood glucose (FBG), fasting blood insulin (FINS), triglyceride (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C).Before sacrifice, intraperitoneal glucose tolerance test (IPGTT) and insulin tolerance test (ITT) were conducted. The histopathological features in the islet tissue were examined with HE staining. The apoptosis in the islet tissue was detected by TUNEL staining. The protein levels of caspase-3, Bcl-2 and Bax were deter-mined by Western blot. The level of miR-375 in the islet tissue was detected by qPCR.For in vitro study, the MIN-6 cells were cultured and divided into control group (incubated with equivalent amount of solvent), miR-375 mimic group and miR-375 mimic +liraglutide group. The cell viability was examined by MTT assay. The protein levels of caspase-3, Bcl-2
and Bax were detected by Western blot.RESULTS: In the in vivo study, compared with control group, the levels of BW, FBG, FINS, TC, TG and LDL-C were decreased significantly in liraglutide group. The islet apoptosis was reduced by the administration of liraglutide. The expression of Bcl-2 was up-regulated significantly, while the protein levels of caspase-3 and Bax were down-regulated significantly in liraglutide group. The level of miR-375 was decreased significantly. In the in vitro study, the cell viability was decreased in miR-375 mimic group and increased in miR-375 mimic +liraglutide group. Moreover, the expression of Bcl-2 was decreased and the protein levels of caspase-3 and Bax were increased with the incu-bation of miR-375 mimic, while the expression of Bcl-2 was increased and the protein levels of caspase-3 and Bax were de-creased with the co-incubation of miR-375 mimic and liraglutide.CONCLUSION: Liraglutide attenuates islet apotosis, and the mechanism may be associated with its effects of reducing the elevated level of miR-375 in islet tissues.
