2.Recent developments in pathogenesis of Hirschsprung's disease

Hirschsprung's disease (HD) is a conginental disorder of enteric nervous system (ENS) and is associated with the abnormal migration of gut neural cerst cell (GNCC).Recent studies showed that the regulation of this migration is a complex gene network.By the development of epigenetic and stem cells,this network is becoming much more clear.Transplantation of the modified gut neural crest stem cells/gut neural crest precursors(GNCSC/GNCP) acquired from the HD bowl is becoming the potential therapy of HD.This paper summerizes the recent advances and is to promote better understanding the pathogenesis of HD.
4.Effects of Emodin on Migration and Invasion Ability and Expressions of E-cadherin and Slugof Human Hepatoma Cell Lines HepG2

Objective To investigate the effects of emodin on the migration and invasion ability and expressions of E-cadherin and Slug of human hepatoma cell lines HepG2; To discuss the possible mechanisms of anti-hepatocellular carcinoma.Methods HepG2 cells were cultured in vitro. The experimental group was treated with emodin at concentrations of 10μmol/L and 20μmol/L as. The negative control group was treated with the same volume of RPMI-1640 medium, while the positive control group was treated with 10μmol/L floxuridine. The cell matrix adhesion assay, wound healing and transwell chamber in vitro invasion assay were used to observe the effects of emodin on HepG2 cell adhesion rate and migration and invasion ability. Western blot analysis was used to observe the changes of expressions of E-cadherin and Slug.Results Compared with the negative control group, emodin inhibited significantly HepG2 cell adhesion rate and migration and invasion ability were in a dose-dependent manner (P<0.05,P<0.01); Western blot analysis showed that the protein expression of E-cadherin increased significantly, and the level of Slug decreased significantly in a dose-dependent manner (P<0.05,P<0.01).Conclusion Emodin can significantly inhibit migration and invasion of HepG2 cells, which mechanism may up-regulate expressions of E-cadherin and down-regulate Slug.

Psychometrical methodology was widely applied in the development of TCM syndrome scales,without considering the consistency between the methods and clinical TCM syndrome differentiation practice and the difference between the syndrome diagnosis and syndrome evaluationt.In this article,we described the theory foundation of the TCM syndrome diagnosis scales,distinguished the difference between the syndrome diagnosis scales and syndrome evaluation scales from several aspects,and expounded the key steps and technical point of TCM syndrome diagnosis scales development superfi cially.Ischemic Stroke TCM Syndrome Scales based on these represented a satisfying performance in clinical validation.
6.Degenerative lumbar scoliosis has no correlation with osteoporosis

BACKGROUND:Osteoporosis is characterized as bone microstructure damage of the whole body and systemic osteopenia. Some scholars believe that the anterior vertebral height of patients with osteoporosis cannot decrease, but shows a trend of increase. OBJECTIVE: To investigate the correlation between degenerative lumbar scoliosis and osteoporosis. METHODS:Thirty-seven patients with degenerative lumbar scoliosis were selected as observation group and another 37 inpatients with no lumbar scoliosis as control group. Modified Cobb method was used to detect the maximum angle between paralel lines to the endplate on the T12-L5 vertebrae, and the vertebrae with these two paralel lines were regarded as upper and lower vertebrae. Dual energy X-ray absorptiometry was used to measure the bone mineral density of L2-L4 segments, femoral neck, Ward's triangle and femoral trochanter. Linear regression analysis was used to analyze the correlation between osteoporosis and degenerative lumbar scoliosis. RESULTS AND CONCLUSION:There were significant differences in the T values in the L2-L4 segments, femoral neck, Ward's triangle and femoral trochanter between the observation and control groups (P < 0.05), and the bone density of the femoral parts was lower than that of the lumbar vertebrae (L2-L4). The incidence of osteoporosis in the patients with lumbar scoliosis was significantly higher than that in the patients with no lumbar scoliosis (P < 0.05). Osteoporosis was a risk factor for degenerative lumbar scoliosis, and the T values of the lumbar vertebra and proximal femur had no remarkable correlation with Cobb's angles. These findings indicate that osteoporosis is the risk factor for degenerative lumbar scoliosis, but it has no correlation with the severity of scoliosis.
7.Biochemical and magnetic resonance imaging features of cartilage in primary osteoarthritis

Objective To investigate the morphological and biochemical changes of primary knee joint osteoarthritis of guinea pigs and study its initial pathogenesis. To study the appearance of magnetic resonance imaging (MRI) in early stages of cartilage degeneration and to evaluate its values. Methods Thirty female Hartley guinea pigs were divided into postnatal 1, 3 and 6 months groups with 10 animals in each group. The degeneration of knee joint cartilage was observed. The proteoglycan (PG) and estradiol (ET) in serum were measured and the concentration of hydroxyproline (HOP), uronic acid (UA) and water ratio (WR) in articular cartilage were determined. Guinea pigs were scanned with magnetic resonance imaging ,using a 1.5T ASC-NT Gyroscan unit (Philips) magnet with 16 cm coil. Gradient echo sequences and spin echo sequences were performed in the sagittal planes at postnatal 1, 3, 6 months. Then signal intensity and thickness of cartilage were measured. The proteoglycan level was measured biochemically and histochemically. Results Degeneration of knee joint cartilage was seen in guinea pigs of 3 months old. The level of serum ET and cartilage WR were increased with age, while the value of serum PG and cartilage UA were decreased with age. 8.Clinical observation of patellar anaplasty and traditional knee arthroplasty in treatment of osteoarthritis

Objective To observe the clinical effect of patellar anaplasty and traditional knee arthroplasty in the treatment of osteoarthri-tis. Methods According to the operation method,50 patients with osteoarthritis were divided into patellar anaplasty group and knee arthro-plasty group,25 cases each. After surgery,all patients were followed up. Results The differences of the operation time,intraoperative blood loss,HSS score before and after operation,the WOMAC scores and radiographic evaluation between two groups were not statistically significant (P>0. 05). The complication rate of patellar anaplasty group was lower than those of knee arthroplasty group significantly,and the difference between two groups was statistically significant(P<0. 05). Conclusion Patellar anaplasty and traditional knee arthroplasty can improve the patient’s knee function effectively,and all the curative effect are reliable,but the complication rate of patellar anaplasty is lower,which is worthy of further promotion and research.
9.Arachidonate 5-Lipoxygenase-Activating Protein and Stroke

5-lipoxygenase-activating protein Gene (ALOX5AP) on chromosome 13q12-13 encodes 5-lipoxygenase-activating protein (FLAP). The latter plays a key role in regulating the synthesis of leukotriene. Studies have suggested that the subjects who carry this gene may increase stroke risk by two times. Therefore, ALOX5AP plays an important role in the pathogenesis of stroke.
10.Cluster Analysis of TCM Diagnostic Data in Different Stages of Ischemic Stroke

To discuss the distribution and evolvement of TCM syndromes in ischemic stoke,we investigated the TCM diagnostic data from 195 cases in the first three months.After the cluster analysis based on the data of the first 3 days,28 days,90 days after the attack,information of the first three days indicated that the variables could cluster into qi deficiency and blood stagnation,phlegmheat,qi and yin deficiency three groups;the information of the 28 days indicated that the variables could cluster into qi deficiency,phlegm-wet,blood stagnation,phlegm-heat four groups;the information of the 90 days indicated that the variables could cluster into qi deficiency and blood stagnation,qi deficiency,phlegm-heat and phlegm-wet four groups.The cluster analysis result shows us a dynamic process of the syndrome evolvement,TCM syndromes are variance in different stage,and become simplicity from complex after treatments.
