1.DISTRIBUTION AND RADIORECEPTORIMAGING OF ~131I-LABELED ANGIOSTATIN IN TUMOR-BEARING MICE

To provide an evidence for clinical application, the distribution and radioimmunoimaging of 131I-labeled angiostatin in tumor-bearing mice were studied. Angiostatin was labeled with 131 I by the conventional chloramines T (Ch-T) method and injected into C-57 mice bearing Lewis lung cancer. The biodistribution of 131 I-angiostatin and whole body ECT imaging were studied at various intervals after injection. The average percentage of ID/g was 12. 48, 18. 56 and 23. 17 for tumor and 7. 04 ,5. 47 and 1. 73 for liver at 48, 96, 144 hours postinjection, respectively. The T/NT ratios were 1. 77, 3. 39 and 13. 39 for liver at 48 ,96 and 144 hours postinjection, respectively. The tumor was showed clearly at 96 hours after injection . The quality of tumor imaging was relevant to the T/NT ratio . The results demonstrated that the 131I-angiostatin binds specifically to the receptor on endothelial cells in the tumor and may also possess the capability of locating lung cancer tissue.
2.Improvement effect and mechanism of paeoniflorin on glucose metabolism,inflammation and oxidative stress in rats with gestational diabetes mellitus

OBJECTIVE To explore the effect of paeoniflorin on glucose metabolism, inflammation and oxidative stress in rats with gestational diabetes mellitus (GDM) and its potential mechanism based on nuclear factor-erythroid 2-related factor 2 (Nrf2)/ heme oxygenase-1 (HO-1)/nicotinamide adenine dinucleotide phosphate:quinone oxidoreductase 1 (NQO1) signaling pathway. METHODS The female rats fed with high fat and high sugar diet and the male rats fed with an ordinary diet were caged, the successfully conceived rats were collected, and streptozotocin was injected intraperitoneally once to induce the GDM model. The successfully modeled rats were randomly divided into the model group, metformin hydrochloride group (200 mg/kg metformin by gavage), paeoniflorin low-, high-dose groups (45, 90 mg/kg paeoniflorin by gavage, respectively), paeoniflorin+ML385 group (90 mg/kg paeoniflorin by gavage and intraperitoneal injection of 30 mg/kg Nrf2 inhibitor ML385), with 12 rats in each group; in addition, another 12 conceived rats fed with an ordinary diet were selected as the control group. The rats in each drug group were given the corresponding drug/normal saline, once a day, for 2 consecutive weeks. Glucose metabolism indexes [fasting blood glucose (FBG), fasting insulin (FINS), insulin resistance index (HOMA-IR)], serum inflammatory factors [interleukin-6 (IL-6), tumor necrosis factor- α (TNF- α)] and renal tissue oxidative stress indexes [superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px)] were detected; the pathological changes of renal tissue were observed, and the protein expressions of Nrf2, HO-1 and NQO1 in renal tissue were detected. RESULTS Compared with the control group, the renal tissue lesions of the model group were obvious, including glomerular atrophy, edema degeneration of renal tubular epithelial cells and a large number of inflammatory cell infiltration; the levels of FBG and FINS, HOMA-IR, the levels of IL-6 and TNF-α in serum, and the level of MDA in renal tissue were significantly increased (P<0.05), while the levels of SOD and GSH-Px and the protein expressions of Nrf2, HO-1 and NQO1 in renal tissue were significantly decreased (P<0.05). Compared with the model group, the renal tissue lesions of rats in paeoniflorin low-dose and high-dose groups were reduced, the above quantitative indexes were significantly improved, and the improvement effect was better in high-dose group (P<0.05), while ML385 could significantly reverse the improvement effect of paeoniflorin on the above indexes (P<0.05). CONCLUSIONS Paeoniflorin can improve the abnormal glucose metabolism, inflammation and oxidative stress damage of renal tissue in GDM rats, which may be related to the activation of Nrf2/HO-1/NOQ1 signaling pathway.
3.The influence of LncRNA MEG3 in temozolomide resistance of glioma cells

Objective :
To explore the effect of LncRNA MEG3 in temozolomide (TMZ) resistance of glioma cells.
Methods :
TMZ⁃resistant glioma cells U87(U87/TMZ) were constructed,qRT⁃PCR was used to detect the LncRNA
MEG3 expression level, and MTT was used to detect the cell proliferation ability. The LncRNA MEG3 in U87/TMZ was overexpressed by liposome method (pcDNA⁃MEG3 group), the empty vector was transfected as the empty vector group ( pcDNA group), and the conventionally cultured U87/TMZ was used as the blank group ( Control group),then treated with 10 μg/ml TMZ. The plasmid transfection effect was analyzed by qRT⁃PCR. The expression of LncRNA MEG3⁃related protein was confirmed by Western blot. The cell cloning experiment detects the effect of LncRNA MEG3 on cell proliferation. The effect of LncRNA MEG3 on cell invasion was tested by Transwell. The effect of LncRNA MEG3 on cell apoptosis was detected by flow cytometry. And mouse transplant tumoranimal model was constructed for in vivo experiments.
Results :
Compared with U87, the expression level of Ln cRNA MEG3 in U87/TMZ was lower (P < 0. 01), and the cell viability of U87/TMZ was higher than that of U87 (P < 0. 01) . After overexpression of MEG3 combined with TMZ, compared with the Control group and the pcDNA group, the pcDNA⁃MEG3 group up⁃regulated the expression of MEG3 mRNA and p53 protein, and down⁃regulated the expression of MDM2 protein, and the proliferation and invasion ability of the pcDNA⁃MEG3 group decreased while the apoptosis ability increased (P < 0. 01) . The construction of mouse transplant tumor animal model showed that the tumor volume and mass of the pcDNA⁃MEG3 group were reduced compared with the pcDNA group (P < 0. 01) .
Conclusion
Overexpression of LncRNA MEG3 can attenuate the drug resistance of TMZ⁃resistant glioma cells.
4.Back-forward bending CT in simulated surgical position to evaluate the remaining real angle and flexibility of thoracolumbar kyphosis secondary to old osteoporotic vertebral compression fracture.

OBJECTIVE:
To introduce a scout view scanning technique of back-forward bending CT (BFB-CT) in simulated surgical position for evaluating the remaining real angle and flexibility of thoracolumbar kyphosis secondary to old osteoporotic vertebral compression fracture.
METHODS:
A total of 28 patients with thoracolumbar kyphosis secondary to old osteoporotic vertebral compression fracture who met the selection criteria between June 2018 and December 2021 were included in the study. There were 6 males and 22 females with an average age of 69.5 years (range, 56-92 years). The injured vertebra were located at T 10-L 2, including 11 cases of single thoracic fracture, 11 cases of single lumbar fracture, and 6 cases of multiple thoracolumbar fractures. The disease duration ranged from 3 weeks to 36 months, with a median of 5 months. All patients received examinations of BFB-CT and standing lateral full-spine X-ray (SLFSX). The thoracic kyphosis (TK), thoracolumbar kyphosis (TLK), local kyphosis of injured vertebra (LKIV), lumbar lordosis (LL), and the sagittal vertical axis (SVA) were measured. Referring to the calculation method of scoliosis flexibility, the kyphosis flexibility of thoracic, thoracolumbar, and injured vertebra were calculated respectively. The sagittal parameters measured by the two methods were compared, and the correlation of the parameters measured by the two methods was analyzed by Pearson correlation.
RESULTS:
Except LL ( P>0.05), TK, TLK, LKIV, and SVA measured by BFB-CT were significantly lower than those measured by SLFSX ( P<0.05). The flexibilities of thoracic, thoracolumbar, and injured vertebra were 34.1%±18.8%, 36.2%±13.8%, and 39.3%±18.6%, respectively. Correlation analysis showed that the sagittal parameters measured by the two methods were positively correlated ( P<0.001), and the correlation coefficients of TK, TLK, LKIV, and SVA were 0.900, 0.730, 0.700, and 0.680, respectively.
CONCLUSION
Thoracolumbar kyphosis secondary to old osteoporotic vertebral compression fracture shows an excellent flexibility and BFB-CT in simulated surgical position can obtain the remaining real angle which need to be corrected surgically.

5.Causes of ABO forward and reverse grouping discrepancy: 75 cases

【Objective】 To analyze the causes and solutions of ABO forward and reverse grouping discrepancy, and to ensure the safety of blood transfusion in clinical. 【Methods】 Samples from 75 patients with discrepancy between forward and reverse grouping in December 2019 and March 2022 were tested and analyzed in combination with saline method(forward and reverse grouping), direct antiglobulin test, antibody screening, antibody identification, increasing plasma volume, incubation enhancement test at 4℃, absorption-eluation test and molecular biological tests. 【Results】 Causes for abnormal reverse typing were as follows : weak antibody in 11 cases(14.67%), unexpected antibody in 19 cases(25.33%), inverted albumin and globulin ratios in 9 cases(12.00%), increased white blood cells in 3 cases(4.00%), inadequate anticoagulation in 3 cases(4.00%), bone marrow transplantation in 2 cases(2.67%), multiple myeloma in 4 cases(5.33%), acute gastrointestinal bleeding in 3 cases(4.00%). Causes for abnormal forward typing were as follows: self-coagulating in 1 case(1.33%), acute leukemia in 7cases(9.34%), post-transplantation in 1 case(1.33%). Causes for abnormal forward/reverse typing: acute leukemia in 1 case(1.33%), condensin syndrome in 1 case(1.33%), autoimmune hemolytic anemia in 6 cases(8.00%), subtype in 4 cases(5.33%). 6.Epidemiological characteristics of facial trauma in preschool children in Fuyang City of Anhui Province

Objective:
To investigate the current situation and related factors of facial trauma in school age children in urban areas of Fuyang, and to provide basis for child facial trauma prevention.
Methods:
By cluster stratified random sampling method, a questionnaire survey was conducted among parents of school age children aged 3-6 in four kindergartens in Fuyang City. A total of 1 062 valid questionnaires were included.
Results:
A total of 146 (13.7%) children with facial trauma were included. The ratio of boys to girls was 1.84∶1.In primary and secondary classes in kindergarten, the reporting rate of boys (24.5%,16.4%) was higher than that of girls (13.4%, 9.0%), and the difference was statistically significant ( χ 2=6.66, 4.25, P <0.05).The incidence of facial trauma was highest (19.4%) in junior kindergarten class children, which decreased with age. Falls injury(61.6%) was the leading cause of facial trauma. Most of facial trauma occurred in family (55.5%) and kindergarten (26.0%). Forehead (26.0%) and mandible (21.2%) were the main injured body sites. Skin laceration (81.5%) was the main type of injury. Facial trauma mostly occurred at 11:00-13:00(19.9%) and 17:00-19:00(27.4%), during June to August (39.7%). The results of univariate analysis showed that the facial trauma of preschool children was associated with gender, class, parentrated child temperament, parental education and family type( χ 2=14.51,18.84,39.66,6.56,12.31, P <0.05).
Conclusion
Facial trauma in preschool children in Fuyang is common, which varies in gender, class, temperament and family type.
7.Effects and mechanisms of APETx2 on visceral sensitivity in mice with post-infectious irritable bowel syndrome

Objective:
To investigate the regulatory effect and mechanism of specific antagonist of acid-sensitive ion channel 3 (APETx2) on visceral sensitivity in mice with post-infectious irritable bowel syndrome (PI-IBS) .
Methods:
The PI-IBS model was established by National Institutes of Health (NIH) mice infected with Trichinella spiralis.Gastrointestinal transport function was assessed by measuring the time to first black stool and the number of fecal pellets collected for 6 hours ; abdominal wall withdrawal reflex (AWR) was used to assess visceral sensitivity ; the expression of calcitonin gene-related peptide ( CGRP) in the colon tissue was detected by immunohistochemistry ; the expression of brain-derived neurotrophic factor (BDNF) and CGRP mRNA in the colon tissues was detected by quantitative real time polymerase chain reaction ( qRT-PCR) . The expression levels of acid sensing ion channel 3 (ASIC3) ,CGRP,and transient receptor potential vanilloid 1 (TRPV1) protein in brain tissue were detected by Western blot analysis.
Results:
Compared with the control group,the PI-IBS group significantly reduced the time of first black stool,the number of fecal particles and AWR score within 6 hours significantly increased,the protein expression of CGRP in colon tissue,BDNF and CGRP mRNA significantly increased,and the protein expression of CGRP,ASIC3 and TRPV1 in brain tissue significantly increased.Compared with the control group,the PI-IBS group significantly reduced the time to first black stool,the number of fecal particles and AWR score within 6 hours significantly increased,the expression of CGRP protein in colon tissue,the expression of BDNF and CGRP mRNA significantly increased,and the protein expression of CGRP,ASIC3 and TRPV1 in brain tissue significantly increased ; compared with the PI-IBS group,the first time of black stool clearance in the APETx2 group was significantly prolonged,the number of fecal particles and AWR score within 6 hours were significantly reduced,the expression of CGRP protein in colon tissue,the expression of BDNF and CGRP mRNA was significantly reduced,the protein expression of CGRP,ASIC3 and TRPV1 in brain tissue was significantly reduced,and the difference was statistically significant (P<0. 05) .
Conclusion
APETx2 can alleviate visceral sensitivity and regulate gastrointestinal motility in PI-IBS mice by downregulating the expression of BDNF,CGRP,ASIC3 and TRPV1.APETx2 may provide a new therapeutic option for the treatment of IBS.
8.Thyroglobulin and thyroglobulin antibody in differential diagnosis between differentiated thyroid carcinoma and nodular goiter

Objective To investigate the clinical significance of thyroglobulin(TG) and thyroglobulin antibody (TGAb) in differential diagnosis between nodular goiter and differentiated thyroid carcinoma(DTC).Methods From Jan.2006 to Jan.2007,180 cases of thyroid carcinoma were included in group A(male:45,average age:47 ; female:135,average age:43)and 150 cases of nodular goiter were included in group B (Male:55,average age 43 ; female:95,average age:41).All the cases came from the General Department of Chengde Central Hospital.Group C (the control group) included 120 cases (male:40,average age:38 ; female:80,average age:44).All patients underwent thyroids ultrasound,thyroid function detection and CT.Results The positive rate of TG and TGAb was 40.0% and 43.3% in group A,which were obviously higher than those in group B (7.33%,9.33%)and group C(4.17%,7.50%).The difference had statistical significance(P <0.05).However,there was no statistical difference between group B and group C (P > 0.05).Conclusion The measurement of TG and TGAb has clinical significance in differential diagnosis between DTC and nodular goiter.
9.Relationship between maternal emotional symptoms and emotional and behavioral problems of preschool offspring

Objective:
To explore the relationship between maternal emotional symptoms and emotional and behavioral problems (EBPs) of preschool offspring, and to provide reference for prevention and intervention of emotional and behavior problems in preschool children.
Methods:
A total of 4 100 preschool children aged 3-6 in Fuyang City were enrolled by stratified cluster sampling method in June 2021. The Strengths and Difficulties Questionnaire (SDQ) and Depression Anxiety and Stress Scale (DASS-21) were used to investigate preschool children s EBPs and maternal emotional symptoms, respectively.
Results:
The detection rates of preschool children s emotional symptoms, conduct problems, hyperactivity, peer interaction problems, prosocial behavior and difficulty scores were 15.7%, 17.4%, 20.0%, 32.3%, 15.1% and 15.8%, respectively. The detection rates of mother s depression, anxiety and stress symptoms were 7.9%, 12.7% and 4.8%, respectively. Multivariate Logistic regression analysis showed that, compared with children of mothers with no depression, anxiety and stress symptoms, offspring of mothers reported depression, anxiety and stress symptoms had significantly higher risks of emotional symptoms, conduct problems, hyperactivity, peer problems and total difficulties ( OR=1.76-6.35, P <0.01). Maternal emotional symptoms had the most significant effect on children s emotional symptoms, and there was no significant correlation between maternal stress symptoms and children s prosocial behavior ( OR=1.40, P >0.05). There was no significant gender difference in the relationship between maternal emotional symptoms and children s emotional and behavioral problems ( ROR=0.73-1.07, P >0.05). For only children whose mothers reported stress symptoms showed a higher risk of emotional and behavior problems than non singleton children ( ROR=2.03, P <0.05).
Conclusion
Maternal emotional symptoms are related to preschool children s emotional and behavioral problems. Early identification and intervention of mother s negative emotional symptoms, are beneficial to the prevention of preschool children s emotional and behavioral problems.
10.Controlled observation on catgut implantation at acupoint for treatment of prolapse of lumbar intervertebral disc.

OBJECTIVETo compare therapeutic effects of catgut implantation at acupoint and routine acupuncture on prolapse of lumbar intervertebral disc.
METHODSOne hundred and forty cases were randomly divided into a treatment group and a control group, 70 cases in each group. The treatment group were treated with catgut implantation at acupoint, once each week, 3 sessions constituting one course, and the control group with routine acupuncture, once every other day, 10 sessions constituting one course.
RESULTSThe effective rate was 95.6% after treatment of one course and 88.2% 3 months later in the treatment group, which were better than 84.6% and 72.3% in the control group, respectively (Both P < 0.05).
CONCLUSIONCatgut implantation at acupoint has a better therapeutic effect on prolapse of lumbar intervertebral disc, with lower cost.
