Objective To establish a method for the determination of alum residual amount in Rhizoma Pinelliae Preparata from different companies,and to provide a scientific evidence for quality evaluation of Rhizoma Pinelliae Preparata.Methods Na2EDTA titration method was applied for the quantitative analysis.Results There were obvious differences in residual quantity of alum in Rhizoma Pinelliae Preparata from different companies.Conclusion The method is simple,reproducible and accurate,it can be used for the determination of alum residual amount in Rhizoma Pinelliae Preparata.

3.USP11 mediates the proliferation and invasion of OSCC cells via regulation of IGF2BP3 expression

Objective:To explore the mechanism of ubiquitin-specific protease 11(USP11)affecting the proliferation and invasion of oral squamous cell carcinoma(OSCC)cells by regulating IGF2BP3 expression.Methods:USP11 expression in OSCC tissues and adja-cent tissues from OSCC patients(n=50)was detected by immunohistochemistry and western blot,and USP11 expression in normal hu-man oral keratinocyte(HOK)cell line and human OSCC cell lines SCC-25 and CAL-27 was detected by western blot.SCC-25 and CAL-27 cells were transfected with siRNA USP11(si-USP11)or siRNA negative control(si-NC).Western blot was performed to de-tect the silencing efficiency of USP11.CCK-8,wound healing assay and Transwell assay were carried out to evaluate the effects of USP11 silencing on cell proliferation,migration and invasion.Western blot was employed to detect IGF2BP3 expression after the knockdown of USP11.Nude mice were inoculated with SCC-25 cells to construct the transplanted tumor model,and the inhibitory effect of USP11 knock-down on SCC-25 cell tumorigenicity was investigated.Results:The USP11 protein level in carcinoma tissues of OSCC patients was significantly higher than in the adjacent tissues,USP11 protein expression was significantly higher in SCC-25 and CAL-27 cells than in HOK cells.The knockdown of USP11 markedly reduced the proliferation,migration and invasion of SCC-25 and CAL-27 cells,and down-regulated the expression of IGF2BP3 cells.Compared with the USP11 silencing group,the proliferation,migration and invasion of SCC-25 and CAL-27 cells were significantly increased in the simultaneous knockdown of USP11 and overexpression of IGF2BP3 cells.Compared with the USP11 overexpression group,the proliferation,migration and invasion of SCC-25 and CAL-27 cells were decreased in the simultaneous IGF2BP3 knockdown and USP11 overexpression cells.Tumorigenicity experiments in nude mice showed that the tumor volume and weight were significantly declined by USP11 knockdown.Conclusion:USP11 is highly expressed in OSCC tissues,which may promote the proliferation,migration and invasion of OSCC cells through up-regulation of IGF2BP3 expression.
4.lncRNA MAFG-AS1 regulates proliferation and apoptosis of oral squamous cell carcinoma HSC4 cells by targeting miR-3180-3p

Objective:To investigate effect of long non-coding RNA(lncRNA)MAFG antisense RNA1(MAFG-AS1)target-ing miR-3180-3p on proliferation and apoptosis of oral squamous cell carcinoma.Methods:MAFG-AS1 and miR-3180-3p expressions in oral squamous cell carcinoma tissues and corresponding normal mucosa were detected by real-time quantitative PCR.Oral squamous cell carcinoma HSC4 was used as research object to construct oral squamous cell carcinoma lines that interfered with MAFG-AS1,overexpressed miR-3180-3p or miR-3180-3p inhibition.Cell viability,clonal formation numbers,apoptosis rate,cleaved cleaved-cas-pase 3 and cleaved-caspase 9 expressions were detected by CCK-8,plane cloning experiment,flow cytometry and Western blot,re-spectively.Targeting relationship between MAFG-AS1 and miR-3180-3p was assessed with luciferase reporter experiment.Results:Compared with normal mucosa,MAFG-AS1 expression was remarkably increased(P<0.05),while miR-3180-3p expression was re-markably decreased(P<0.05)in oral squamous cell carcinoma tissues.Interference with MAFG-AS1 or miR-3180-3p overexpression could reduce HSC4 cell viability and clonal formation numbers(P<0.05),promote cell apoptosis(P<0.05),and up-regulate expres-sions of cleaved-caspase 3 and cleaved-caspase 9 proteins(P<0.05).miR-3180-3p was direct target of MAFG-AS1,and MAFG-AS1 negatively regulates miR-3180-3p expression.miR-3180-3p inhibition could increase HSC4 cell viability and colony formation number(P<0.05),inhibit cell apoptosis(P<0.05),and down-regulate cleaved-caspase 3 and cleaved-caspase 9 protein expressions(P<0.05),thereby weakening effect of interference with MAFG-AS1 on proliferation inhibition and apoptosis promotion on HSC4 cells(P<0.05).Conclusion:Interfering with MAFG-AS1 can inhibit proliferation and promote apoptosis of oral squamous cell carcinoma cells HSC4 by up-regulating miR-3180-3p expression.
5.Clinical value of serum adenosine deaminase combined with whole blood microRNA-197-3p in the diagnosis of human brucellosis

Objective:To study the clinical value of serum adenosine deaminase (ADA) combined with whole blood microRNA (miR)-197-3p in the diagnosis of human brucellosis.Methods:From January 2020 to September 2023, 120 patients with brucellosis were selected from the Department of Pulmonary and Critical Care Medicine, Affiliated Hospital of Jining Medical University (referred to as this hospital) and assigned to the brucellosis group, including 89 patients in the acute stage, 19 patients in the chronic stage, and 12 patients in the latent infection. In addition, 60 healthy volunteers from the animal husbandry industry were selected as the control group from the physical examination population in this hospital during the same period; sixty critically ill patients with community-acquired pneumonia were selected from the Department of Pulmonary and Critical Care Medicine of this hospital as the infection control group. Patients in the brucellosis group and infection control group were given standardized treatment. Fasting elbow venous blood samples were collected in the morning before and after treatment in the brucellosis group and infection control group, at enrollment in the control group, and serum ADA and whole blood miR-197-3p levels were tested. Multivariate logistic regression analysis was used to study the risk factors affecting the occurrence of brucellosis. Receiver operating characteristic (ROC) curve was used to evaluate the diagnostic efficacy of ADA, miR-197-3p, and their combination for brucellosis.Results:Before treatment, the levels of serum ADA (U/L: 12.35 ± 2.89, 18.33 ± 4.57, 29.75 ± 6.46, 22.20 ± 4.78, 14.15 ± 3.03) and whole blood miR-197-3p (1.09 ± 0.33, 2.25 ± 0.41, 2.68 ± 0.59, 2.43 ± 0.51, 1.18 ± 0.40) in the control, infection control, acute stage, chronic stage, and latent infection groups were compared, and the differences were statistically significant ( F = 4.38, 4.02, P = 0.014, 0.019). Compared with the control group, the serum ADA and whole blood miR-197-3p levels in the other 4 groups were higher ( P < 0.05). Compared with the infection control group, the serum ADA level in the acute stage and chronic stage groups was higher, and the serum ADA level in the latent infection group was lower ( P < 0.05); the level of whole blood miR-197-3p was higher in the acute stage group, but lower in the latent infection group ( P < 0.05). Compared with before treatment, the levels of serum ADA and whole blood miR-197-3p in the infection control, acute stage and chronic stage groups were lower after treatment ( P < 0.05). Multivariate logistic regression analysis showed that increased levels of serum ADA and whole blood miR-197-3p were risk factors for brucellosis [odds ratio ( OR) = 2.235, 3.404, 95% confidence intervals ( CI): 1.491 - 3.362, 1.623 - 7.605, P < 0.001]. ROC curve results showed that serum ADA and whole blood miR-197-3p had auxiliary significance in the diagnosis of brucellosis ( P < 0.001). The area under curve (AUC) of serum ADA combined with whole blood miR-197-3p was 0.933, which was superior to the diagnostic efficacy of serum ADA and whole blood miR-197-3p alone (AUC = 0.823, 0.840). Conclusion:The combination of serum ADA and whole blood miR-197-3p has high clinical value in the diagnosis of human brucellosis, and can provide an important reference for the clinical diagnosis and efficacy evaluation of brucellosis.
6.A comparative study of international and Chinese public health emergency management from the perspective of knowledge domains mapping.

BACKGROUND:
At the end of 2019, the outbreak of coronavirus disease 2019 (COVID-19) severely damaged and endangered people's lives. The public health emergency management system in China has played an essential role in handling the response to the outbreak, which has been appreciated by the World Health Organization and some countries. Hence, it is necessary to conduct an overall analysis of the development of the health emergency management system in China. This can provide a reference for scholars to aid in understanding the current situation and to reveal new research topics.
METHODS:
We collected 2247 international articles from the Web of Science database and 959 Chinese articles from the China National Knowledge Infrastructure database. Bibliometric and mapping knowledge domain analysis methods were used in this study for temporal distribution analysis, cooperation network analysis, and co-word network analysis.
RESULTS:
The first international article in this field was published in 1991, while the first Chinese article was published in 2005. The research institutions producing these studies mainly existed in universities and health organizations. Developed countries and European countries published the most articles overall, while eastern China published the most articles within China. There were 52 burst words for international articles published from 1999-2018 and 18 burst words for Chinese articles published from 2003-2018. International top-ranked articles according to the number of citations appeared in 2005, 2007, 2009, 2014, 2015, and 2016, while the corresponding Chinese articles appeared in 2003, 2004, 2009, and 2011.
CONCLUSIONS
There are differences in the regional and economic distribution of international and Chinese cooperation networks. International research is often related to timely issues mainly by focusing on emergency preparedness and monitoring of public health events, while China has focused on public health emergencies and their disposition. International research began on terrorism and bioterrorism, followed by disaster planning and emergency preparedness, epidemics, and infectious diseases. China considered severe acute respiratory syndrome as the starting research background and the legal system construction as the research starting point, which was followed by the mechanism, structure, system, and training abroad for public health emergency management.
