1.Expression and clinical significance of serum miR-139-5p in patients with ovarian cancer

Objective To explore the expressions and clinical values of serum microRNA-139-5p (miR-139-5p) in diagnosis and prognosis of patients with ovarian cancer.Methods From March 2009 to December 2011,a total of 34 patients with serous ovarian cancer and 34 female health examiners were collected in our hospital.The expressions of serum miR-139-5p before and after operation were detected in 34 patients with ovarian cancer and 34 healthy women by real-time quantitative polymerase chain reaction (RT-qPCR).The data was analyzed by t-test.The clinical values of serum miR-139-5p in diagnosis and prognosis of patients with ovarian cancer were analyzed.Results The expression of serum miR-139-5p in patients with ovarian cancer (9.45 ± 0.40) was significantly higher than that in healthy controls (3.56 ± 0.10),with a significant difference (t =14.340,P < 0.001).The expression of miR-139-5p at 1 month after operation of patients with ovarian cancer (5.53 ± 0.49) was lower than that before operation,with a significant difference (t =7.800,P <0.001).The expression of serum miR-139-5p was correlated with lymph node metastasis (P =0.038),but was not correlated with age (P =0.728),clinical stage (P =0.118),tumor size (P =0.732) and differentiation (P =0.071).Kaplan-Meier survival curve revealed that the median survival time of ovarian cancer patients with low serum miR-139-5p expression (44.9 months) was longer than that of patients with high serum miR-139-5p expression (25.9 months),with a significant difference (x2 =5.721,P =0.017).Conclusion The expression of miR-139-5p is increased significantly in the serum of patients with ovarian cancer,which can be used as a potential biomarker for diagnosis and prognosis of patients with ovarian cancer.
2.The effect of respiratory muscle training on the swallowing function of dysphasic stroke survivors

Objective To observe the effect of respiratory training on post-stroke dysphagia.Methods Forty-two stroke survivors with dysphagia were randomly divided into a control group and an observation group,each of 21.Both groups were given routine swallowing training,while the observation group was additionally provided with respiratory training once a day for 4 weeks.Before and after the treatment,the Kubota drinking water test (KDWT) was administered and a videofluoroscopic swallowing study (VFSS) was performed to assess swallowing function.Forced vital capacity (FVC),forced expiratory volume in one second (FEV1) and peak expiratory flow (PEF) were also measured to assess the subjects' pulmonary function.Results After the treatment,the average KDWT and VFSS scores were significantly better in the observation group than in the control group.The effectiveness rate of the observation group (85.71%) was significantly higher than that of the control group (61.90%).After the intervention,the average FVC [(3.57-±0.48)L],FEV1[(2.83±0.49) L/s] and PEF [(5.36±1.04) L/s] of the observation group were significantly better than those before the treatment and those of the control group after the treatment.Conclusion Swallowing training supported by respiratory training is superior to swallowing training alone for improving the swallowing (and pulmonary) function of post-stroke dysphagia patients.Such combined treatment is worthy of application in clinical practice.
3.The effect of respiratory muscle training combined with occupational therapy on the upper limb function of stroke survivors

Objective:To observe the effect of combining respiratory muscle training with occupational therapy in rehabilitating the upper limb function of stroke survivors.Methods:Fifty stroke survivors with upper limb dysfunction were randomly divided into an observation group and a control group, each of 25. Both groups were given routine rehabilitation treatment including proper positioning of the affected limb, physical therapy and motor function training. The observation group also received progressive resistance training of the inspiratory muscles and respiration control training combined with occupational therapy twice daily for 4 weeks. The trunk control test (TCT), Berg balance scale (BBS), Fugl-Meyer Upper Extremity Assessment (FMA-UE), Action Research Arm Test (ARAT), Modified Ashworth Scale (MAS) for the Hemiplegic Upper Limb and the Modified Barthel Index (MBI) were used to assess the core stability, balance, upper limb functioning, upper limb muscle tension and ability in the activities of daily living of all of the subjects.Results:Before the treatment there were no significant differences in any of the indexes between the two groups. Afterward the average TCT, BBS, FMA-UE, ARAT, MAS and MBI scores of both groups had improved significantly, but the improvements were all significantly greater in the observation group.Conclusions:Combining respiratory muscle training with occupational therapy can further improve the function of the upper limbs and daily living ability beyond what is observed with traditional rehabilitation therapy after a stroke.
4.Effects of Lentivirus-Mediated CIB1 Silencing on Biological Behavior of Breast Cancer Cells

Objective To investigate the effects of calcium and integrin-binding protein 1(CIB1)on the cell proliferation,invasion,apoptosis,and migration of triple-negative breast cancer cells and its possible mechanism.Methods MDA-MB-231 and MDA-MB-468 cells were divided into CIB1-knockdown(infected with CRISPR/Cas9 lentivirus)and negative-control groups.Cell Counting Kit-8(CCK-8)and 5-ethynyl-2′-deoxyuridine(EdU)assays were performed to detect cell proliferation.Cell apoptosis was determined through flow cytometry.Scratch and Transwell experiments were conducted to measure the migration and invasion abilities of cells.The mRNA and protein expression levels of β-catenin,adenomatous polyposis coli(APC),glycogen synthase kinase 3β(GSK-3β),and c-myc were detected via real-time quantitative polymerase chain reaction and Western blot.Results Compared with the negative-control group,the CIB1-knockdown group showed decreased cell proliferation,invasion,and migration(P<0.05)and increased cell apoptosis(P<0.05).The mRNA and protein expressions of β-catenin,APC,and c-myc decreased(P<0.05),and that of GSK-3β increased(P<0.05).Conclusion CIB1 knockdown can inhibit cell proliferation,invasion,and migration and promote the apoptosis of breast cancer cells.Its mechanism may be related to the inhibition of Wnt/β-catenin signaling pathway.
5. Neck lymphatic metastasis, surgical methods and prognosis in early tongue squamous cell carcinoma

Objective:
To investigate the different pattern of neck lymph node metastasis, the choice of surgical methods and prognosis in early tongue squamous cell carcinoma.
Methods:
A total of 157 patients with early oral tongue squamous cell carcinoma were included in this study. Statistical analysis was performed to identify the pattern of lymph node metastasis, to determine the best surgical procedure and to analyze the prognosis.

Objective :
To explore the effects of piperlongumine ( PL) on the proliferation and apoptosis of triplenegative breast cancer MDA⁃MB⁃231 cells in vitro and in vivo as well as its possible molecular mechanism.
Methods:
MDA⁃MB⁃231 cells were treated with different concentrations of PL. MTT assay was performed to detect the level of cell proliferation. The apoptotic level was detected by flow cytometry. Western blot was performed to detect protein expression levels of proliferating cell nuclear antigen(PCNA) , B ⁃cell lymphoma⁃2 (Bcl⁃2) , cyclin⁃dependent kinase inhibitor 1A(CDKN1A/p21) , phosphorylated signal transducer and activator of transcription 3(p⁃STAT3), signal transducer and activator of transcription 3 (STAT3) , hypoxia inducible factor⁃1 α ( HIF⁃1α ) and Survivin. MDA⁃MB⁃231 cells were used to model tumor bearing nude mice and then PL was injected by intraperitoneal ( i.p. ) . The tumor protein expression levels of PCNA , p ⁃STAT3 , STAT3 , HIF⁃1 α and Survivin were detected by Western blot.
Results :
PL inhibited cell proliferation and induced apoptosis in a dose⁃dependent manner. PL down⁃regulated the protein expression levels of PCNA , Bcl⁃2 , p ⁃STAT3 , HIF⁃1 α and Survivin , and up⁃regulated the protein expression level of p21 . Furthermore , PL inhibited tumor growth and down⁃regulated the protein expression levels of PCNA , p ⁃STAT3 , HIF⁃1 α and Survivin in nude mice.
Conclusion
PL inhibits the proliferation of MDA⁃MB⁃231 cells in vitro and in vivo , and the underlying mechanism can be related to the negative regulation of STAT3/HIF⁃1 α pathway.
7. Application of cerebral regional oxygen saturation monitoring in neonatal blood exchange transfusion

Objective:
To observe the changes in cerebral regional oxygen saturation (CrSO2) during neonatal blood exchange transfusion(BET) and its effect on the level of bilirubin in neonates with severe hyperbilirubinemia.
Methods:
From January 2017 to March 2018, 52 newborns with severe hyperbilirubinemia were hospitalized in the Department of Neonatology, Children′s Hospital of Fudan University.Every newborn was treated with BET.Near infrared spectroscopy was used to monitor CrSO2 in the process of BET.The monitoring lasted from 2 hours before the beginning of BET to 2 hours after the completion of BET.The CrSO2 were recorded every 2 minutes and total surem bilirubin (TSB) and transcutaneous bilirubin(TCB) was measured.During this period, it is accompanied by the monitoring of neonatal body temperature, heart rate, respiration and bolld oxygen saturation(SpO2). The differences in CrSO2 changes at different time points during BET were compared.At the same time, the correlations between CrSO2 and blood oxygen saturation, TSB and TCB levels were analyzed.The results of repeated measurement analysis of variance compared between the two groups were corrected by Bonfferoni.
Results:
Among the 52 children, there were 33 males (63.46%) and 19 females (36.54%). The gestational age, average birth weight and average head circumference of newborns were (38.6±2.1) weeks, (3 338±444) g and (33.6±3.2) cm, respectively.The Apgar score of newborn was (8.1±1.6) scores at 1 minute after birth.The level of TSB detected for the first time after admission was (457.9±97.8) μmol/L.The CrSO2 after BET (74.6%-76.0%) was significantly higher than that before BET (69.4%-69.0%), and the difference was statistically significant (

Objective:
To investigate the effect of activation of the stimulator of interferon genes(STING) pathway on macrophage polarization function and its role in T-cell response.
Methods:
Mouse macrophage RAW264.7 cells were used.STING signaling related proteins in RAW264.7 macrophage treated with STING agonist diABZI were analyzed by Western blot,including TANK-binding kinase-1(TBK1),interferon regulatory factor-3(IRF3),STING,p-TBK1,p-IRF3,p-STING.The polarization of macrophage RAW264.7 cells treated with diABZI was analyzed by flow cytometry.Co-culture of diABZI-treated RAW264.7 macrophage and T cells was applied to evaluate the change of T cell response.
Results:
STING signaling related proteins were upregulated in macrophage RAW264.7 cells treated with diABZI for 3 hours.The expression of CD86 was upregulated on the surface of macrophages after 12 hours of diABZI treatment,and the CD86/CD206 ratio was elevated,which presented the M1 polarization phenotype.When coculturing diABZI-treated macrophage RAW264.7 cells with T cells,the cytokine secretion ability of T cells including CD4+T and CD8+T cells was enhanced and the expression of CD107a in CD8+T cells was upregulated.
Conclusion
STING signaling induces M1 polarization of macrophages which enhance the function of T cells,especially CD8+T cell immune response.
