1.Study of survivin-siRNA combined with the chemotherapy drugs to enhance the breast cancer MCF-7 cells apoptosis and reverse drug resistance

Objective To investigate the effects of siRNA against survivin combined with the neoadjuvant chemotherapy (paclitaxel and epirubicin) on the apoptosis in breast cancer MCF-7 cell and reverse drug resistance.Methods Molecular cloning technique was applied to construct the expression vector of siRNA against survivin,and effectene transfection reagent was used to transfect MCF-7 cell.Survivin expression was detected by qPCR and Western blot methods.The effects of paclitaxel or epirubicin combined with or without siRNA against survivin,on the drug susceptibility of MCF-7 cell were detected by CCK-8 assay and that of including MCF-7 cell apoptosis were detected by flow cytometry.Results Survivin-siRNA-2 effectively inhibited the expression of survivin mRNA (0.30±0.03) and protein (0.37±0.09) (P < 0.05).Both paclitaxel [IR 24,48 h =(38.5±4.0) %,(41.7±6.3) %] and epirubicin [IR 24,48 h =(37.0±8.6) %,(40.6± 12.1) %] can suppress the proliferation of MCF-7 cell and induce its apoptosis,and siRNA against survivin combined with chemotherapy drugs enhanced both effects significantly[paclitaxel IR 24,48 h =(76.0±2.9) %,(85.1±4.0) %; epirubicin IR 24,48 h =(74.6±5.6) %,(82.5±4.8) %] (P < 0.05).The inhibition rate of MCF-7 was concentration and time dependent.Conclusions Blocking survivin gene expression by RNAi,and combining with the appropriate chemotherapy can significantly enhance the sensitivity of MCF-7 cell to drugs and cell apoptosis.This technology has important potential clinical value in the therapy of breast cancer.
2.A study on microvessel density, VEGF and bFGF in oromaxillofacial sarcoma

Objective:To explore the role of MVD, bFGF and VEGF in OMFS and the relationship between them. Methods: Immunochemistry streptavidin peroxidase method was used to detect the MVD and the bFGF, VEGF expressions in 57 cases of OMFS samples. Results: MVD, bFGF and VEGF expressions were different between OMFS and benign. The close correlations were found among the expressions of VEGF, bFGF and MVD. Conclusion: The expressions of VEGF and bFGF are associated with MVD which indicated that VEGF and bFGF may play important roles in angiogenesis.
3.Influencing factors of mandible osteoporosis

OBJECTIVE: With the prolongation of life span, more and more people care their health. In this paper we summarized the studies about correlative factors of mandible osteoporosis so as to investigate the cause of osteoporosis and how to inhibit or postpone the occurrence of mandible osteoporosis and improve people's quality of life.DATA SOURCES: We searched Qsinghua Tongfang Database for papers about mandible osteoporosis published between January 1991 and May 2005 with the key words of "osteoporosis, mandible bone, alveolar bone"in Chinese. Meanwhile, related English literatures were also searched in Medline between January 1975 and May 2005 with the same key words.STUDY SELECTION: The literatures included all the related papers of correlative factors of mandible osteoporosis. Inclusive criteria were: the study types were randomized controlled trials, drug stress test and clinical drug effect test; the samples were both human and animals. Exclusive criteria: Reviews and literatures without controls were excluded.DATA EXTRACTION: Totally 182 literatures related to the correlative factors of mandible osteoporosis were collected, and 36 were accorded with the inclusive criteria. The excluded were 146 papers of repeated drug effect study.DATA SYNTHESIS: A total of 36 papers including 5 321 human and 152 animals analyzed the data of correlative factors of mandible osteoporosis in age, sex, hormones, biting force, physical training and living habit,etc., respectively, which included 6 interfere-test papers of ovariactomized rats.CONCLUSION: It is concluded that mandible osteoporosis can result in a part of bone mine loss, which is related to age, sex, hormones, biting force,physical exercise and living habits, and so on.

Objective:To investigate the relationship between the expression of P16 protein and the clinico-pathology of squamous cell carcinoma(SCC) of buccal mucosa and its precursor lesions. Methods:The immunohistochemical stain against p16 was performed in 30 cases of SCC of buccal mucosa, 32 cases of buccal leukoplakia and buccal lichen planus and 10 cases of normal buccal mucosa. All data were analyzed quantitatively by imag analysis technique. Objective: To study the effect of nicotine on alkaline phosphatase (ALP) activity of human dental papilla mesenchymal cells in vitro. Methods: Human dental papilla mesenchymal cells were cultured in the presence of nicotine at various concentrations, ALP level of the cells was meseased at different time with an enzyme dynamical method and was expressed as A value at 410 um. Results: After 3day-exposure of the cells to nicotine(g/L) at 0, 0.05, 0.1, 0.2 and 0.4 the A values(per 10 5cells) were 0.64 ? 0.15, 0.61?0.03, 0.47 ? 0.05, 0.44 ? 0.06 and 0.39? 0.03; after 5-day-exposure those were 0.55 ? 0.12, 0.42?0.16, 0.29? 0.10, 0.19 ? 0. 4 and 0.10?0.02; after 7 day exposure those were 0.40? 0.12, 0.31 ? 0.10, 0.26? 0.06, 0.13? 0.02 and 0.09? 0.02, respectively. Conclusion: Nicotine inhibited the alkaline activity of human dental papilla mesenchymal cells in a dose and time dependant manner. Nicotine may play a role on the differentiation of the cells.
