Chinese Journal of Pathophysiology 2014;(12):2284-2288
[ ABSTRACT] Diabetes is characterized by an absolute or relative deficiency inβ-cell mass, which cannot be re-versed with existing therapeutic strategies. The restoration of the endogenous islet β-cells can stabilize the level of blood glucose. The isletβ-cells can be obtained from the directional differentiation of stem cells, but the process is complex and has the risk of teratomas generation.Cell direct reprogramming, one terminal differentiated cell can transdifferentiate into another kind of terminal differentiated cell, which is other than directional differentiation from stem cells.Direct reprogram-ming gives rise to the generation of isletβ-cells from one terminal differentiated cell, may be preferable for diabetes therapy because of its unique advantage.

Low back pain caused by intervertebral disc degeneration is a common clinical chronic disease. The regenerative ability of intervertebral disc tissue is extremely poor. Meanwhile, current treating methods can not fundamentally solve such problems. With the increasing awareness of the mechanism of disc degeneration and the rapid development of the fields of cellular and molecular biology, gene and materials engineering, using stem cells and tissue engineering technology to slow down or reverse the progress of disc degeneration may become possible. The author reviewed the application of stem cells for treating degenerative discs from present researching status and concepts for the future in the combination of researches reported both at home and abroad.

Objective To investigate the nursing intervention of pregnant women with antiphespholipid syndrome(APS). Methods In experimental group 40 cases with APS positive and control group 40 cases with APS ACA-negative for comparison. Results The abortion occurred in 6 cases(15. 0%) ,2 cases of stillbirth(5.0%) ,loss of pregnancy(abortion+stillbirth) 8 cases (20. 0%) in experimental group. The abortion occurred in 2 cases (5.0%), no stillbirth in control group. The loss of pregnancy in experimental group were significantly higher than the control group (X2=9. 54, P<0. 005 ). The rate of positive in ACA-IgG (40. 0%) after treatment lower than before treatment (52. 5 %) (X2 = 3.85, P<0. 05). The rate of positive in ACA IgM (37. 5%) after treatment lower than before treatment(40. 0)% (X2=3.81 ,P<0. 05 ). Conclusion The APS positive rate and the loss of pregnancy were closely related. The overall nursing intervention conld improve efficacy and reduce the complications.

AIM: To compare the therapeutic effects of transplantation of human umbilical cord mesenchymal stem cells (hUCMSC) through different ways on diabetic mice.METHODS: hUCMSCs were labeled with enhanced green fluorescent protein (EGFP) and luciferase (Luc) reporter gene, and then the cells were transplanted into the diabetic mice through pancreas or tail vein to monitor the migration of the hUCMSCs in vivo. The pathological changes of pancreas tissue sections were determined by HE staining.Weight and blood glucose of the mice were measured dynamically. To compare the therapeutic effects, serum insulin levels were analyzed and glucose tolerance test were also performed.RESULTS: In vivo bioluminescence imaging results showed that the hUCMSCs transplanted into pancreatic capsule was mainly located in the pancreas while the hUCMSCs transplanted through vein tail injection was mainly located in the lung.HE staining illustrated that islet cells presented distinctive boundary and no infiltration of inflammatory cells in pancreatic capsule transplantation group was observed, but a little inflammatory cell infiltration and fibrosis formation in tail vein injection group were seen.A significant decrease in blood glucose level and a significant increase in serum insulin level in pancreas transplantation group were showed as compared with vein tail injection group.CONCLUSION: Transplantation of hUCMSCs through different approaches demonstrates different effects. The transplantation of hUCMSCs into pancreatic capsule is more effective on hyperglycemia reversion, insulin secretion and improvement of beta-cell function than that through tail vein.

Journal of Third Military Medical University 1983;0(03):-
Objective To investigate the roles of activin A(ACT-A) and follistatin(FS) in renal interstitial fibrosis. Methods Renal interstitial fibroblasts were isolated from SD rat kidney and primarily cultured. The cells were divided into A group,F group and A+F group, and then each group was further divided into 5 subgroups according to their culture media being added with ACT-A(0,0.3,3,30,or 100 ng/ml),FS(0,0.3,3,30,or 100 ng/ml),and ACT-A(30 ng/ml) plus FS(0,0.3,3,30,or 100 ng/ml) respectively.Levels of type Ⅳ collagen(Ⅳ-C) and hydroxyproline(HYP) in the supernatants of cultured fibroblasts were measured by ELISA assay. Results Concentrations of Ⅳ-C and HYP in cultured rat renal interstitial fibroblasts were in a dose-dependent manner with ACT-A treatment.FS had no effect on these concentrations, but it inhibited the effects of ACT-A on Ⅳ-C and HYP in the supernatants of cultured fibroblasts in a dose-dependent manner. Conclusion ACT-A might be involved in occurrence and development of renal fibrosis by affecting renal interstitial fibroblasts.Exogenous FS could suppress the effects of ACT-A on the cultured renal fibroblasts.

Chinese Journal of Parasitology and Parasitic Diseases 1987;0(02):-
Indirect fluorescent antibody test (IFAT) using frozen sections of Brugia malayi adult worms as antigen was employed in the diagnosis and epidemiological investigation of human filariasis. Sera were collected from 704 cases with bancroftian or malayan microfilaremia. the positive rate was 92.8-99.1%. Of 150 healthy people from non-endemic areas, only one showed a positive reaction (false positive rate 0.7%) (Table 1). This technique proved to be highly sensitive and specific for the diagnosis of filariasis and the antigen is easy to prepare. It might be used in sero-epidemiological investigation for the assessment of filariasis control.
