1.Effects of ketamine on effector functions of lipopolysaccharide-stimulated human neutrophils in vitro

Objective To investigate the effects of ketamine on the effector functions (phagocytosis, respiratory burst and release of proteolytic enzymes) of lipopolysaccharide (LPS) stimulated human neutrophils in vitro. Methods Blood samples were collected from healthy volunteers. The study was divided into four groups: LPS group and three ketamine groups (K1, K2 and K3). The final concentration of ketamine in each group was 0, 3, 30 and 300 ?g/ml respectively. Phagocytosis was assessed in whole blood by NBT phagocytosis test (n=8) and respiratory burst by flow cytometry (with dihydrorhodamine 123 as fluorescent marker, n=5). The release of three proteolytic enzymes was measured with isolated polymorphonuclear neutrophils (PMNs) by turbidimetry (lysozyme) and chromatometry (elastase and ? glucurolidase) methods (n=9).Results Ketamine dose dependently inhibited phagocytosis, respiratory burst and proteolytic enzyme releasing of LPS stimulated human neutrophils in vitro. Higher concentrations of ketamine were required to suppress respiratory burst as compared with the concentrations needed to suppress phagocytosis and proteolytic enzyme releasing.Conclusions The inhibitory effects of ketamine on the effector functions of LPS stimulated human neutrophils may contribute to the attenuation of neutrophil mediated inflammatory injuries.
2.Observing the change of cerebral blood flow perfusion in acute vertebral artery stenosis in dogs by ~(99)Tc~m-ECD imaging

AIM: To observe the early changes of local cerebral blood supply in dogs during the acute vertebral artery stenosis by 99 TcmECD cerebral blood flow perfusion imaging(CBFPI).METHODS: Twenty-four dogs were randomized into groups of normal controls(A),moderate stenosis(B),severe stenosis(C) and arterial occlusion(D),with 6 in each group.Group A were free from any intervention,but Group B,C and D were undergone ligation of the right vertebral arteries by the extent of 50%~69%(Group B),70%~99%(Group C) or 100%(Group D).The imaging agents were injected intravenously 0.5-1 h after ligation and the single photon emission computed tomography(SPECT) was performed after 1 h.RESULTS: CBFPI examination of the early changes of cerebral blood supply during the acute vertebral artery stenosis by visualization showed that the sensitivity was 33.3%,83.3% and 100% in Group B,C and D,respectively,with an average sensitivity of 72.2%,while semi-quantitative assessment suggested that the sensitivity was 50%,100% and 100% in the three groups,respectively,with an average sensitivity of 83.3%.When examining the activity ratio for regions of interest(ROI) by semi-quantitative 99 Tcm-ECD CBFPI,the findings suggested that Group D and C decreased significantly(exception in cerebellum in Group C),but Group B had no evident changes,as compared with Group A.By comparison of the groups of B-C,B-D and C-D,differences were observed in the right temporal lobes,whereas the changes in left temporal lobes were seen in Group B-C or B-D.The changes were seen in occipital lobes when groups of B and D were compared,but with no differences in cerebellum.CONCLUSION: The degree of stenosis in vertebral arteries may involve varied regions,the temporal lobe is susceptive,followed by occipital lobe,while cerebellum is somewhat resistant from the involvement.
3.The HRCT staging and pulmonary function tests abnormal analysis of early coal workers pneumoconiosis *

Objective To explore the early coal pneumoconiosis patients with HRCT staging and pulmonary function abnormali-ties indicators ,provide an objective basis for clinical diagnosis and treatment early pneumoconiosis .Methods Strict control of the inclusion criteria :no smoking ,no pneumonia ,no tuberculosis ,no lung cancer and no lung surgery ,as a result ,total 33 early coal pneumoconiosis patients were chosen in the study .All of them were underwent paired CT obtained at full inspiratory position .High kv chest radiography ,and pulmonary function tests at the same day .Reference to the chest X-ray pneumoconiosis classification method as pneumoconiosis HRCT staging .Analyzed the correlation between early coal pneumoconiosis with HRCT staging and the parameters ,including Forced vital capacity (FVC) ,Forced expiratory volume in 1 second(FEV1) ,FEV1/FVC ,Maximum midexpi-ratory flow(MMEF) ,Peak expiratory flow(PEF) ,FEF75 ,FEF50 ,FEF25 ,Total lung capacity(TLC) ,Residual volume(RV) ,Carbon monoxide diffusion capacity(DLco) ,KCO and so on .Results HRCT could be more accurately display the shape ,size and distribu-tion of coal pneumoconiosis small nodules .There were significant correlation with MMEF ,PEF ,FEF75 ,FEF50 and FEF25 in different HRCT staging .Conclusion HRCT can early detection and validation early coal workers pneumoconiosis .Early coal worker′s pneu-moconiosis small airway flow rate decline caused by inhalation coal dust .
4.Research in preventive effect of auricular therapy in schizophrenia patients with constipation

Objective To evaluate the preventive effect of auricular therapy in schizophrenia pa-tients with constipation. Methods 200 psychotic inpatients with anti-psychotic medication were random-ly divided into the intervention group and the control group with 100 cases in each group. The intervention group received conventional therapy combined with auricular therapy while the control group adopted only conventional therapy. The incidence of constipation was observed in the two groups. Results The rate of constipation was 11.34% and 36.73% in the intervention group and the control group, respectively. Auricu-lar therapy showed various effect in constipation caused by different drugs, especially for constipation caused by clozapine, chlorpromazine and pentiapine. Conclusions Auricular therapy showed good effect in preventing constipation induced by antipsychotic drags with less pain and adverse effect, easy and safe to operate, besides, it can improve the medication compliance of patients and promote their rehabilitation.
5.MRI evaluation of acute spinal cord injury

Objective To assess the clinical value of MRI in evaluation of acute spinal cord in-jury. Methods A total of 50 patients with acute spinal cord injury were examined with magnetic reso-nance (MRI), X-ray and CT. Of all, 15 patients for spinal MR imaging were examined with axial and sngittal spin-echo and fast spin-echo imaging and 17 with coronal imaging. All imaging was performed on 1.5T superconducting system (GE/SIEMENS) with spine surface coil. Results Among 50 patients with acute spinal cord injury, spinal cord edema was detected by MRI in 16 patients, by CT in four but none by X-ray. Intraspinal cord hemorrhage was detected by MRI in 21 patients, by CT in 11 but none by X-ray. Compression and dislocation were detected by MRI in 34 patients, by CT in 15 but none by X-ray. Incomplete and complete transection of spinal cord was found by MRI in 10 patients, by CT in three but none by X-ray. Conclusion MRI is superior to CT scanning and X-ray in detection and evaluation of a-cute spinal cord injury and is the optimal method for examination and diagnosis of acute spinal cord injury.
6.Effects of transcutaneous electrical acupoint stimulation assisting sodium nitroprusside controlled hypotension on Glu,ATⅡ and SOD

Objective To observe the effects of transdermal acupoint electric stimulation(TEAS)assisting sodium nitroprus-side induced controlled hypotension on serum glucose (Glu),angiotensin Ⅱ (ATⅡ)and superoxide dismutase (SOD),and to inves-tigate the protective effect of TEAS under controlled hypotension anesthesia.Methods 60 cases undergoing elective endoscopy si-nus surgery by adopting sodium nitroprusside induced controlled hypotension under general anaesthesia maintained the mean arterial pressure(MAP)in 50-60 mm Hg and were randomly and equally divided into two groups.The group Ⅰ conducted TEAS,while the group Ⅱ did not conduct TEAS.The controlled hypotension time and surgery time were recorded in the two groups;Glu and ATⅡ values were detected before anesthesia (T0 ),30 min after hypotension (T1 ),hypotension stopping(T2 );SOD was detected at T0 ,T2 ,30 min after hypotension(T3 ).Results The operation time and controlled hypotension continuous time had no statistically significant difference between the two groups(P >0.05).The Glu value in the group Ⅰ had no statistically significant difference a-mong the 3 time points,while which at T1 ,T2 was higher than that at T0 in the group Ⅱ(P <0.05),and which at T1 ,T2 in the group Ⅱ was higher than that in the group Ⅰ (P <0.05);the ATⅡ value at T1 was higher than that at T0 in the group Ⅰ (P <0.05),while which at T1 ,T2 was higher than that at T0 in the group Ⅱ(P >0.05),which at T1 ,T2 in the group Ⅱwas higher than that in the group Ⅰ (P <0.05);the SOD value at T2 was lower than that at T0 in the group Ⅰ,which at T2 ,T3 in the group Ⅱwas lower than that in the group Ⅰ(P <0.05).Conclusion TEAS assisting sodium nitroprusside controlled hypotension can better in-hibit the stress response.
7.Effects of Rhodiolae Crenulatae Radix ET Rhizoma extracts on the aphrodisiac qualities of male mice

Objective To study the effects of Rhodiolae Crenulatae Radix ET Rhizoma extracts on sexual behavior of male mice.Methods 50 healthy male mice were randomly divided into the low dose, middle dose and the high dose Rhodiola group, theNanbao capsules group and the normal control group, 10 mice per group. The low dose, middle dose and high dose group were drenched with 0.05, 0.20 and 0.80 g/kg Rhodiola diluent respectively. TheNanbao capsules group mice were drenched with 2.00 g/kg turbid liquid. The normal control group were drenched with saline in the same volume. Liquid is drenched two times each day for 21 days. After 21 days, 50 female mice were matched with to the ratio of 1:1. The number of free movement and swimming test were observed before execution. After the execution, the organ indexes were calculated, and then the contents of SOD and MDA in the testis and liver were measured.Results Compared with the normal control group, capturing latency period of low dose group and middle dose group (20.88 ± 19.94 s, 35.40 ± 22.02 svs.78.11 ± 43.33 s) significantly decreased (P<0.05 orP<0.01). Testicular coefficient of the middle dose group (0.72% ± 0.10 %vs. 0.64% ± 0.08%) was significantly increased (P<0.05); the content of SOD in testicular of the middle dose group, the high dose groups and theNanbao capsules group (152.71 ± 38.10 U/mg, 122.32 ± 52.76 U/mg, 94.38 ± 22.20 U/mgvs. 25.30 ± 14.21 U/mg) increased (P<0.01); the content of SOD in liver of the middle dose group and theNanbaocapsules group (77.71 ± 26.35 U/mg, 74.10 ± 26.04 U/mgvs. 57.92 ± 17.17 U/mg) significantly increased (P<0.05).Conclusion Rhodiola extract can improve the ability of sexual behavior of male mice, and improve the antioxidant capacity of testis and liver.
8.Effects of electroacupuncture on the elderly's postoperation cognitive dysfunction after general anesthesia

Objective To study the effects of electroacupuncture (EA) on the elderly's postoperation cognitive dysfunction (POCD) after general anesthesia.Methods Patients who need to have surgical operation done for lumbar spinal stenosis were randomly divided into an observation group (electroacupuncture and routine intravenous anesthesia) and a control group (only routine intravenous anesthesia),and EA was administered to those in the electroacupuncture group for 30 minutes at an interval of 10 minutes,beginning at 30 minutes prior to routine intravenous anesthesia and lasted till the end of the surgery.The differences regarding intraoperative propofol and remifentanil dosage,cognitive function,the levels of serum interleukin-6(IL-6),interleukin-10 (IL-10) and S100-β were compared.Results The intraoperative propofol and remifentanil dosages in observation group were (6.54 ± 3.27) μg/(kg· h) and (3.67 ± 2.53) mg/(kg· h),respectively,while those in control group were (9.70 ± 4.82) μg/kg and (5.62 ± 1.73) mg/kg.The intraoperative propofol and remifentanil dosages in observation group were significantly lower than those in control group (P < 0.05).At 72 hours post-operation the incidence of POCD in observation group was significantly lower than that in control group (1 case,2.5% versus 8 cases,20%),while the MMSE score was higher in observation group than that in control group (25.7 ± 1.9 versus 23.2 ± 2.1).The post-operation level of serum IL-6,IL-10 and S100-β in observation group were significantly lower than that in control group (P < 0.05).Conclusion Electroacupuncture could significantly improve the elderly's postoperation cognitive dysfunction after general anesthesia.It could inhibit the excessive expressions of IL-6,IL-10 and S100-β,alleviate the brain damage and reduce the incidence of POCD.

Objective To detect the in vitro effect of the traditional Chinese medicine on the tachyzoites of Toxoplasma gondii. Methods Supernatant (1^5 ml) of different doses of the traditional Chinese medicine (Changqing capsule) was collected by normal saline immersion and 2^5?10+4 Toxoplasma gondii tachyzoites were added in each paste well for 8 hours. Spiramycin, pyrimethamine and azithromycin in different doses were used as controls. Normal saline was used as negative control. Mice were inoculated with drug-treated tachyzoites intraperitoneally or intragastrically. The normal mice were subcultured after 8 days for 3 generations. 10.Changes of the BDNF in the hippocampus of P77PMC during seizures

AIM: To quest the relationship bwteen BDNF and seizures. METHODS: BDNF and other correlative proteins in the hippocampus of audioepileptic rat (P77PMC) have been detected by immunohistochemistry at different stages of seizures including quiet stage, preconvulsive stage, convulsive stage and postconvulsive stage. Wistar rats were served as controls. RESULTS: At quiet stage, the BDNF in the hippocampus of P77PMC were less than that of control group and stepped up after stimulation, trkB in the hippocampus of P77PMC were high at every stages, but PY20 were increased only after stimulation. NPY in the hippocampus of P77PMC were in a low level before seizures. CONCLUSION: The endogenetic BDNF may alter the excitement of P77PMC by adjusting the expression of NPY in hippocampus and activating its receptor trkB.
