Objective:To study the effects of 17 beta-estradiol (E2) on the proliferation and metastasis potential of salivary gland mucoepidermoid carcinoma M_3SP_4 cells. Methods:The effects of E2 on the potential of proliferation and metastasis of M_3SP_4 cells were investigated by cell counting, MTT assay, clonegenetic assay, in vivo tumour growth and metastasis assay in nude mice. The expression of VEGF,c-erbB-2,Ki-67 and P16 in M_3SP_4 cells was examined by immunohistochemistry assay. Results:E2 increased the proliferation of M_3SP_4 cells at 10~-10-10~-5 mol/L. E2 at 10~-7 mol/L showed the strongest effects. After treatment with E2 at 10~-7 mol/L,the population doubling time of M_3SP_4 cells decreased by 10.8%,clonegenecity increased by 225%, VEGF,c-erbB-2 and Ki-67 expression increased,P16 decreased. In the in vivo assay, the tumour growth increased by 65% and metastasis increased by 609% in nude mice treated with E2 at 0.005 2 mg/d. Conclusion:E2 may stimulate the proliferation and metastasis potential of M_3SP_4 cells.

Objective:To establish a culture method of human salivary gland epithelial cells and to study their growth characteristics in vitro. Methods:Tissue explant technic was employed to culture human salivary gland epithelial cells in serum free medium (SFM),1∶1 DMEM/F12 and 1∶1 DMEM/F12 containing 25 ml/L fetal boven serum (FBS) respectively. The morphology of the cultured cells was observed by phase contract microscope. The cell growth was studied by cell counting. The cells were identified by HE staining,PAS staining and SP staining. Results:Growth of human salivary gland epithelial cells was observed in primary culture in the three kinds of medium in all 10 cases. The cultured cells were epidermoid, positive for cytokeratin, negative for Vimentin and positive for PAS staining. The cells in SFM could be subcultured for five passages, while only for one passage in the other two kinds of medium. Conclusion:SFM is superior to serum free 1∶1 DMEM/F12 or 1∶1 DMEM/F12 containing 25 ml/L FBS for the culture of human salivary gland epithelial cells.

Air pollution and global warming are increasingly deteriorating. Large amounts of polyamides derived from fossil fuel sources are consumed around the world. Cadaverine is an important building monomer block of bio-based polyamides, thus biotechnological processes for these polymers possess enormous ecological and economical potential. Currently, the engineered strains for biological production of cadaverine are Corynebacterium glutamicum and Escherichia coli. We review here the latest research progress of biosynthesis of cadaverine including metabolism of cadaverine in microorganisms, key enzymes and transport proteins in cadaverine synthesis pathway, optimum pathways and cadaverine yields.

International Journal of Cerebrovascular Diseases 2011;19(8):580-584
Objective To investigate the relationship between the C825T polymorphism of G protein β3 subunit (GNB3) and essential hypertension and cerebral infarction patients with a history of hypertension. Methods The C825T polymorphism of GNB3 was determined by polymerase chain reaction-restriction fragment length polymorphism in 110 healthy subjects aged over 40 years, 92 patients with essential hypertension, and 80 cerebral infarction patients with a history of hypertension. Sex, age, family histories of diabetics, smoking and alcoholic consumption were documented, and body mass index, waist-hip ratio, total cholesterol (TC),triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), and fasting blood glucose concentration were measured. A multivariate logistic regression analysis was used to screen the factors associated with the occurrence of cerebral infarction in patients with hypertension. Results The distribution of the C825T polymorphism of GNB3 in the 3 groups all met the Hardy-Weinberg Law of genetic equilibrium. The genotype frequencies of CC, CT and TT in cerebral infarction patients with a history of hypertension were 33%, 57%, and 20%, respectively; those in patients with essential hypertension were 33%, 42%, and 25% ; respectively;and those in the control group were 26%, 54%, and 20%, respectively. There were no significant differences (x2 =4. 030, P =0. 402). The allele frequencies of 825T in the 3 groups were 39%, 40%, and 47%, respectively. There were no significant differences (x2 =0. 832). A multivariate logistic regression analysis showed that TC (odds ratio [ OR] 10. 810, 95% confidence interval [ CI] 2. 64544. 136, P =0. 000), TG (OR 5. 453, 95% CI 1.662-17. 881, P =0. 005),HDL-C (OR, 0. 181, 95% CI 0. 041-0. 795, P = 0. 027), blood glucose (OR 2. 386, 95% CI1.062-5. 363, P =0. 035), and diabetes (OR 7. 156, 95% CI 1.271-40. 291, P =0. 026) were the independent risk factors for cerebral infarction, and the GNB3 genotype and allele did not enter the model. Conclusions The C825T polymorphism of GNB3 may not be associated with cerebral infarction and essential hypertension.

OBJECTIVE:To study the feasibility of overflow dissolution method for evaluating the drug in vitro sustained re-lease performance. METHODS:Overflow dissolution method was established by simulating the drugs elimination in vivo. Using Nifedipine sustained-elease tablets(Ⅰ)from 2 different manufacturers as model drug A,B,concentration-time curve, cumulative release rate- time curve, release velocity-time curve of model drugs in release pool at 3 different overflow speed (0,1.50,3.00 mL/min)were investigated. RESULTS:When overflow speed was 0,the cumulative dissolution was consistent with that of the con-ventional dissolution method. As the overflow speed increased, cmax of drug A,B was decreased [A:(8.89±0.20),(5.21±0.04), (3.51±0.03)μg/mL;B:(7.62±0.05),(4.80±0.09),(2.89±0.04)μg/mL];cumulative release rate was increased [A:(85.47± 2.45)%,(94.29 ± 2.44)%,(96.04 ± 2.56)%;B:(73.28 ± 1.13)%,(78.46 ± 1.94)%,(82.50 ± 1.69)%] ;tmax was ahead (A:1.5,1.0,0.5 h;B:2.0,1.0,0.5 h). CONCLUSIONS:Overflow dissolution method has avoided the inhibition of too large drug concentration on drug release, making complete drug release and more accurate evaluation of in vivo sustained release performance of the preparation.
