1.Expression and correlation analysis of serum soluble myeloidcell trigger receptor 1,interleukin-21,25(OH)vitamin D in children with inflammatory bowel disease

Objective To investigate the expression of serum soluble myeloid cell trigger recep-tor 1(sTREM-1),interleukin-21(IL-21),25(OH)vitamin D[25(OH)D]in children with inflam-matory bowel disease and their relations.Methods A total of 107 children with inflammatory bowel disease were selected as research objects and included in observation group.At the same time,53 healthy people were selected as control group.The patients in the observation group were divided into active stage group(n=54)and remission stage group(n=53)according to disease activity.The ex-pression of sTREM-1,IL-21,25(OH)D were compared between the observation group and the con-trol group and between active stage group and remission stage group.Kendall's tau-b method was used to analyze the correlations of sTREM-1,IL-21 and 25(OH)D with inflammatory bowel disease activi-ty.Receiver operating characteristic(ROC)curve model was used to analyze the area under the curve(AUC)value,sensitivity and specificity of sTREM-1,IL-21 and 25(OH)D in the diagnosis and prediction of inflammatory bowel disease(IBD).Results The serum levels of sTREM-1 and IL-21 in the observation group were higher than those in the control group,but level was lower than that in the control group(P<0.05).The serum levels of sTREM-1 and IL-21 levels in the active stage group were higher than those in the remission stage group,but 25(OH)D level was lower than that in the remission stage group(P<0.05).Kendall's tau-b correlation analysis showed that sTREM-1 and IL-21 were positively correlated with inflammatory bowel disease activity(r=0.460,0.484;P<0.05).25(OH)D was negatively correlated with inflammatory bowel disease activity(r=-0.434,P<0.05).ROC curve analysis showed that the AUC values of sTREM-1,IL-21,25(OH)D and their combination in the diagnosis of inflammatory bowel disease were 0.791,0.852,0.808,0.862,respectively(P<0.05),and the sensitivities were 74.80%,81.30%,75.50%and 81.30%,respectively.The specificities were 75.50%,75.50%,81.30%and 79.20%,re-spectively.The AUC values of sTREM-1,IL-21,25(OH)D and their combination in predicting the active phase of inflammatory bowel disease were 0.821,0.840,0.799,0.840,respectively(P<0.05),the sensitivities were 79.60%,75.90%,77.40%and 87.00%,respectively,and the specificities were 79.20%,75.50%,83.30%and 79.20%,respectively.Conclusion Serum levels of sTREM-1,IL-21,25(OH)D are closely related to the occurrence and development of in-flammatory bowel disease.Dynamic monitoring of their levels is helpful to provide an important ref-erence for clinical diagnosis of inflammatory bowel disease and evaluation of disease progression in children.

3.Optimization of the Purification Technology of Total Triterpenoids of Ligustrum lucidum by Box-Behnken Re- sponse Surface Methodology

OBJECTIVE:To i mprove the transfer rate and purity of oleanolic acid and ursolic acid in total triterpenoids from Ligustrum lucidum ,so as to optimize the purification technology. METHODS :Oleanolic acid and ursolic acid were used as representative components of total triterpenoids ,and their contents were determined by HPLC. The determination was performed on Thermo BDS Hypersil C 18 column with mobile phase consisted of methanol- 0.02% ammonium acetate solution (80∶20,V/V)at the flow rate of 1.0 mL/min. The detection wavelength was set at 210 nm,and column temperature was 30 ℃. The sample size was 20 μ L. In single factor tests,using transfer rate of oleanolic acid and ursolic acid as index ,the effects of water precipitation temperature and time ,the amount of redissolved ethanol on the purification technology was investigated ;using transfer rate and purity of two components as indexes ,the effects of the amount of activated carbon and volume fraction of crystallization ethanol were investigated. Based on it ,using the amount of redissolved ethanol and activated carbon ,volume fraction of crystallization ethanol as factors ,Box-Behnken response surface methodology was used to optimize the purification technology ,and validation tests were performed. RESULTS :The optimal purification technology was adding 4-fold(mL/g,the same below )water in L. lucidum concentrated solution ,placing for 2 hours at 0 ℃(water precipitation );adding 1-fold ethanol to dissolve (redissolution); adding 4% activated carbon (edulcoration);finally adding water to adjust the volume fraction of ethanol to 80%,placing at 4 ℃ for 12 hours(crystallization),centrifuging and drying. The results of 3 times of validation tests showed that the transfer rates of oleanolic acid and ursolic acid in total triterpenoids prepared by optimized technology were 61.11% and 65.78%,the purities of them were 53.44% and 19.79%,and RSDs were both lower than 3%. CONCLUSIONS :The optimized purification technology has high extraction efficiency and simple operation ,which can be used for industrial production of purification of total triterpenoids from L. lucidum and the development of corresponding preparations.
