1.Diagnostic Value of Serum miR-122,ALT and HBV-DNA in Patients with Chronic Hepatitis B

Objective To explore the clinical significance of serum miR-122,ALT and HBV-DNA in patients with chronic hepatitis B(CHB).Methods 900 patients of CHB were selected in Zaozhuang Mining Group Hospital from January 2014 to January 2016,and the serum levels of miR-122,ALT and HBV-DNA were detected,and analysed the relevance among the three and chronic hepatitis B.Results Serum markers of hepatitis B test showed that the HBV-DNA load in patients with chronic hepatitis B or early hepatitis infection was significantly higher than that of other patients (F=12.355,P=0.000),and the ALT content in patients with CHB was significantly higher than that of other patients (F=24.654,P =0.000).There was no significant correlation between ALT and HBV-DNA and HBV-M pattern.With the increase of HBV load,the proportion of hepatitis B patients with ALT > 80 U/L increased gradually within limits (P<0.05).Correlation analysis showed that ALT content was positively correlated with HBV loading (r=0.919,P =0.034).With the increase of HBV load,the level of serum miR-122 in patients with CHB was gradually increased (F=1463.435,P=0.000).Correlation analysis showed that serum miR-122 was positively correlated with HBV (r=0.975,P=0.012).Conclusion There was a correlation between serum miR-122,ALT and HBV-DNA in patients with CHB.The three indicators have good sensitivity and specificity in the diagnosis of CHB,so can use the three joint detection in the diagnosis in patients CHB,in order to improve the diagnostic accuracy and help patients better treatment.

Capillary bronchitis is a common respiratory infectious disease. In the early childhood,it is mainly caused by respiratory syncytial virus,which is closely related with later asthma. The pathogenesis is still not entirely clear. Recent studies have found that glutathione functions in keeping airway epithelial integrity,a-gainst lung injury and inflammation. This article summarizes the protective effect of glutathione in capillary bron-chitis.

Objective To investigate the effects of Puerarin on the expression of hyperphosphorylated-tau (P_ ser404-tau) and choline acetyltransferase (ChAT) in hippocampus of Alzheimer's disease(AD) rats.Methods The rats were divided into sham-operation group, AD group and Puerarin treated group at randomly. The AD rat models were established by injecting amyloid-beta peptide(A?_ 25-35) into right amygdala. The sham-operation rats were injected with trifluoroacetic acid in the same location. The spatial memory function was observed by Y-maze. 4.Lateralization in the hand motor cortex during rehabilitation after acute cerebral infarction

Objective To observe any change in the laterality index (LI) in the active volume of the hand motor cortex during rehabilitation after acute cerebral infarction and to analyze the mechanisms involved in the rehabilitation of motor function.Methods Sixteen patients with acute cerebral infarcts were administered standard but individualized rehabilitation training.Blood oxygenation-dependent functional magnetic resonance imaging (BOLD-fMRI) was used to evaluate the active volume of their hand sensorimotor cortex (SMC) and the LI,at admission and after 14 days of rehabilitation.The Fugl-Meyer motor assessment for the hand (FMA) was used to evaluate hand function.Ten healthy volunteers were recruited as a control group and subjected to a single BOLD-fMRI examination to confirm the location and the volume of the active area when performing the same rehabilitation exercises.Results The baseline LI of affected hand SMC activation was significantly smaller than that of the unaffected hand [(0.010 ±0.808) versus (0.789 ± 0.157)],but no significant difference was observed between the affected and the unaffected hands after treatment.Rehabilitation therapy significantly increased the SMC LI of affected hand activation when compared with the baseline,but no such effect was observed with the unaffected hand.In 12 patients with dysfunction of the right hand as evaluated by the FMA,the baseline LI of the affected hand was smaller than that of the unaffected hand and that of the healthy volunteers.Conclusion Rehabilitation after acute infarction can promote functional recovery.The LI of the affected hand reflects cerebral plasticity during rehabilitation after acute cerebral infarction.
5.BOLD-fMRI quantitive analysis of motor cortex by three hand motor tasks in normal subjects

Objective To identify the neural substrates of three motor tasks (repetitive and sequential fin-get-to-thumb opposition movements in turn, making fates, fingers passive flexion-extension movements in turn) of dominant and subdominant hands by using the whole-brain functional magnetic resonance imaging. Methods Ten right-handed healthy volunteers were scanned while they were performing the movement tasks with their right and left fingers. The motor cortex active volume and intensity was recorded. Quantitive analysis of motor cortex was conducted with paired t test. Results Under the three hand motor tasks, activation volumes in SMC during movements of the subdominant hand were significantly larger than those during movements of the dominant hand (P < 0. 05). Activation volumes during finger-to-thumb opposition movements and passive bendlng-extending fingers movements were significantly larger than those during movements of making fasts (P < 0.05). Activation intensity during passive ben-ding-extending fingers movements was significantly larger than those during movements of making fasts (P < 0.05). Conclusion The representation of the Motor Cortex is related to the complexity of the hand motor exercises. Quantitive criterion as volume and intensity approves the dissymmetry of cortex activation by dominant and subdominant fingers'movements. It is practicable and credible to adopt invariable fingers passive flexion-extension movements in turn in the study on BOLD-fMRI.
6.Affection to differentiation of Th17 cell in bronchiolitis rat models after injectingγ-secretase inhibitor

Objective:To investigate the affection to the differentiation of Th17 cell in rat models of bronchiolitis after blocking Notch signaling by γ-secretase inhibitor and provide rationale to seek new target for bronchiolitis drug treatment. Methods:The rats were randomly divided into normal group,bronchiolitis group andγ-secretase inhibitor group. The model of bronchiolitis was established successfully by nasal dripping,and γ-secretase inhibitor(MW167) was injected into the vena caudalis. The pathological changes of the airway were observed by HE staining;the plasma level of interleukin17 ( IL-17 ) was detected by ELISA;the level of RORγt mRNA in lung tissues and peripheral blood mononuclear cells( PBMCs) was tested by real-time quantitative PCR;the levels of Notch signaling and RORγt protein in lung tissues were examined by Western blot. Results:Compared to the bronchiolitis group, the histopathologic change in MW167 intravenous injection group was significantly alleviated;the plasma level of IL-17 was decreased;the level of RORγt mRNA in lung tissues and PBMCs was lower in MW167-treated group than bronchiolitis group;the levels of Notch signaling and RORγt were decreased. Conclusion:γ-secretase inhibitor through intravenous injection suppresses the differentiation of Th17 cell and relieves the airway inflammation of bronchiolitis in rat models after blocking Notch signaling and has potential therapeutic value for treating bron-chiolitis.
7.Changes in the hand motor cortex after cerebral infarction and during rehabilitation

Objective To observe any change in the active volume of the hand motor cortex during rehabilitation therapy after acute cerebral infarction and analyze the mechanisms involved in motor function rehabilitation.Metbods Of 16 patients with acute brain infarction,8 were administered routine intemal medicine treatment only,while and the other 8 received rehabilitation therapy in addition.Before treatment and after 14 days,the patients were assessed with functional magnetic resonance imaging(fMRI)and the Fugl-Meyer assessment of motor function (FMA).The active volume of the motor cortex was compared between the two groups of patients.Ten healthy volunteers were examined with fMRI to confirm the location and the volume ofthe active area when performing the sanle exercises.Results After treatment,all the 16 patients showed increased motor cortex active volume,and their FMA scores also increased.Those receiving rehabilitation therapy improved tO a significantly greater extent than those treated with internal medicine treatment alone.Conclusion Rehabilitation of patients with acute infarction Can activate a greater volume of the motor cortex and promote functional recovery.
8.Possible molecular mechanism of β2 adrenergic signaling pathway in regulation of inflammation after stroke

Infection is one of the common complications of stroke. It can seriously affect the prognosis of patients. Poststroke infection is closely associated with immune system imbalance, while the excessive activation of sympathetic nerve is an important factor of the occurrence of immunosuppression after stroke. The sympathetic nerve regulates immune cels primarily via β2 adrenergic receptors on the surface of the immune cels. There is evidence to show that β2 adrenergic receptor mediated immune function enhances rather than inhibits under certain conditions, but its specific molecular mechanism is unclear. This article reviews the molecular mechanisms of β2 adrenergic signaling pathway in regulation of inflammation after stroke.
9.Role of microRNA-21 in the proliferation and apoptosis of ovarian epithelial carcinoma cells

ObjectiveTo investigate the role and mechanism of microRNA-21 (miR-21) in the proliferation and apoptosis of ovarian epithelial carcinoma cells. MethodsA short-hairpin RNA specifically targeting miR-21 plasmid was constructed, and the recombinant was identified by restriction endonuclease analysis and DNA sequencing. Three experimental groups were included, transfection group (transfected with pSIREN-miR-21 ), negative control group ( transfected with pSIREN-miR-21-neg) and blank control group (without transfection plasmid ). The expression of miR-21 was detected by stem-loop real-time reverse transcription (RT)-PCR in OVCAR3 cells ,and western blot was used to detect the expression of programmed cell death 4 ( PDCD4 ) protein. Tethyl thiazolyl tetrazolium (MTT) and flow cytometry method were used respectively. ResultsRecombinant plasmid (pSIREN-miR-21) was constructed successfully and identified by restriction endonuclease analysis and DNA sequencing. The relative expression level of miR-21 in cells transfection, negative control and blank control group was 0.26 ± 0.08, 1.26 ± 0.21and 1.00 respectively. The level of miR-21 in the cells in transfection group was significantly lower than those in the negtive control and blank control group(P <0. 01 ). The gray scale of PDCD4. protein was 1443 ±33,858 ± 19 and 846 ± 16 in the transfection group, negative control and blank control group respectively. The value of PDCD4 in transfection group was higher than other control groups, and there were significantly difference among them( P <0. 01 ). Moreover, the optical density of the cells in transfection group was 0. 661 ±0. 015,significantly lower than those in two control groups (0. 848 ± 0. 150 for negative control, 0. 935 ± 0. 133 for blank control,P < 0. 01 ). Forty-eight hours after tranfection, the rate of viable apoptotic cell was significantly higher than negative control and blank control group [(25.821 ± 0. 763 )% vs.(0. 010 ± 0. 003 ) % vs. (0. 238 ± 0. 023) % ; P < 0. 01];72 hours after tranfection, the rates of viable apoptotic cell and necrotic cell were all higher than the two control groups [the rate of viable apoptotic cell was ( 30. 480 ±0. 821 ) %, ( 7. 792 ± 0. 312 ) % and ( 7. 033 ± 0. 257 ) % respectively ( P < 0. 01 ) ; the rate of necrotic cell was (3.558 ±0.211)%, (1.557 ±0.067)% and (1.049 ±0.028)%, respectively (P<0. 01)].ConclusionmiR-21 might play an important role in the proliferation and apoptosis of ovarian epithelial carcinoma cells through negatively control the expression of PDCD4.
10.Apoptosis-promoting effect of Panax notoginseng extracts on MNNG-transformed GES-1 cells

OBJECTIVE: To study the apoptosis-promoting effect of the serum from Panax notoginseng extracts-fed dog on precancerous gastric cells by means of flow cytometry. METHODS: In the experiment, we adopted eternalized human gastric mucosa epithelium GES-1 cells transformed by N-methyl-N'-nitro-N-nitroso-guanidine (MNNG) (MC cells) as the model of precancerous lesions for study in vitro. We took the serum of a dog before and at two different points of time (2 and 6 hours) after feeding the dog with Panax notoginseng extracts for experiment. The MC cells were cultured in mediums with different concentrations of the medicated serum at 2- or 6-hour point of time for 72 hours. By means of flow cytometry, we examined the apoptosis-promoting effects of the serums on the MC cells. RESULTS: The medicated serums at these 2 points of time had significant effects in promoting MC cell apoptosis. The proportions of apoptotic cells in culture mediums with medicated serums had a significant increase as compared with those in culture mediums with non-medicated serums (serum obtained before administration of extracts to the dog) under the same conditions (P<0.05). The number of MC cells in G(0)/G(1)phase was decreased (P<0.05) and that in G(2)/M phase increased (P<0.05), while no consistent changes were observed in S phase. CONCLUSION: The medicated serums obtained at the two different points of time have significant apoptosis-promoting effects on MC cells. They decrease the number of MC cells in G(0)/G(1) phase and increase the number of MC cells in G(2)/M phase. This is probably responsible for the effects of Panax notoginseng extracts in inhibiting the proliferation of MC cells and promoting its apoptosis.
