1.The influences on the structure of rats′aorta with long-term high fat forages diet

Aim To investigate the influences on the aortal structure of rats with long-term high fat forages diet.Methods 14 SD rats were divided into two groups:the control group and the test group. The test rats were fed with high fat forages.12 weeks later, the aortas of the rats were observed with a light microscope, transmission electron microscope(TEM) and scanning electron microscope (SEM).Results In the test group, the aortic tunica intima thickening, endotheliocyte injury and monocyte adhesion were found with a light microscope; the elastic lamina being broken and the smooth muscle cells proliferated. Under TEM, the endothelial cell membrane of the aorta in the test rats was destroyed and appeared to be worm-eclipsed shape.Mitochondria exhibited swelling,vacuole degeneration and its cristae was dissolved, broken or some disappeared.Rough endoplasmic reticula (RER) expanded. The endothelial cell spaces were enlarged and the cell junctions deformed. Monocytes adhering to the endothelial cell stretched out pseudopodia and intruded into the endothelial crevice and the subendothelial layer. Some basement membranes completely sloughed following with endothelial cell. ERE and ribosomes increased in smooth muscle cells. SEM observation showed that the endothelial cells became swelling and the surface of endothelial cell was worm-eclipsed or crater shape. There were deeper crevices between endothelial cells.Conclusions Long-term high fat forages diet can induce injury of the endothelium and elastic lamina, adhesion of the monocytes and its intrusion into endothelial layer and subendothelial layer, proliferation of the subendothelial layer and smooth muscle in the aorta of rats.
2.Efficacy Observation of Urinary Kallidinogenase in the Treatment of Acute Cerebral Watershed Infarct

OBJECTIVE:To observe clinical efficacy of urinary kallidinogenase in the treatment of acute cerebral watershed in-farct (WSI). METHODS:128 patients with WSI were randomly divided into control group and treatment group,each of the 64 cases. Control group was given Shuxuening 15 ml added into 0.9% Sodium chloride 250 ml,ivgtt,qd;treatment group received urinary kallidinogenase 0.15 PNA added into 0.9% Sodium chloride 100 ml,ivgtt,qd. Both groups were treated for consecutive 14 days. Neurologic impairment score(NIHSS)and clinical efficacy were observed in 2 groups before treatment and 3,7 and 14 days after treatment. The blood specimens were collected after 7 and 14 days treatment,to determine serum levels of TCC. RESULTS:After treatment,NIHSS and total effective rate of treatment group were significantly higher than those of control group,with statis-tical significance(P<0.01). There was no statistical significance in TCC between 2 groups before treatment(P>0.05);7 days af-ter treatment,TCC level of 2 groups increased significantly,to 14 days,and a declive;the treatment group was higher than the control group,with statistical significance (P<0.05). CONCLUSIONS:Urinary kallidinogenase can improve clinical efficacy of WSI significantly,and promote neurologic impairment symptom and TCC levels.
3.Effect of interrupted suture and continuous suture on perineal rehabilitation of parturients: a Meta-analysis

Objective:To explore the effect of perineal lateral incision and continuous suture of deep and superficial Ⅱdegree laceration on postoperative perineum rehabilitation of parturients.Methods:Randomized controlled trials of intervention effects on postoperative maternal wound rehabilitation using continuous sutures and interrupted sutures of mucosal, muscle, and skin layers after perineal lateral resection and Ⅱdegree lacerations were searched in Cochrane Library, PubMed, Medline, Embase, Science direct, China Biology Medicine, China National Knowledge Infrastructune, Wanfang Database, VIP Database. The retrieval time limit was from database establishment to October 1, 2020. using RevMan 5.3 software for statistical processing.Results:After searching and screening, 12 articles were finally included. The total sample size was 2 827 cases. After the two groups used different suture techniques, the results of Meta analysis showed that the pains ( MD=1.02, 95% CI=1.02-1.06), the suture time ( MD=-4.59, 95% CI=-7.29 - -1.91) and the healing condition ( MD=-0.55, 95% CI=-0.71 - -0.39) of the two groups were compared, the differences were significant (all P<0.01). Conclusions:In the suture of perineal lateral incision and Ⅱdegree laceration, continuous suture technique is used to suture the mucosal layer, muscle layer, and cortex, which can reduce the suture time, reduce the pain of the parturient, and is beneficial to the healing of the wound. It can be clinically based on the parturient. It can be used in accordance with the neatness and depth of the mouth and the needs of the parturient.
4.Effects of acupuncture and moxibustion combined with limb functional exercise based on continuous nursing management on stroke patients with limb dysfunction

Objective:To explore effects of acupuncture and moxibustion combined with limb functional exercise based on continuous nursing management on stroke patients with limb dysfunction.Methods:A total of 130 stroke patients with limb dysfunction who were admitted to Nanyang First People's Hospital from September 2017 to July 2019 were selected by the convenient sampling method. They were randomly divided into the control group and the observation group, with 65 cases in each group. The control group was given routine nursing combined with limb function exercise while the observation group was given acupuncture and moxibustion treatment and continuous nursing on the basis of the control group. TCM symptom scores, neurological score, Fugl-Meyer Assessment (FMA) Scale score, Postural Assessment Scale for Stroke Patients (PASS) score and 6-Minute Walking Distance were compared between the two groups.Results:After intervention, the score of each dimension of TCM syndrome scores and neurological score of the observation group were lower than those of the control group, and the differences between the two groups were statistically significant ( P<0.01) . The FMA upper and lower limb scores of the observation group were higher than those of the control group, and the differences between the two groups were statistically significant ( P<0.01) . The PASS score of the observation group was higher than that of the control group, and the 6-Minute Walking Distance was more than the control group, the differences between the two groups were statistically significant ( P<0.01) . Conclusions:Acupuncture and moxibustion combined with limb functional exercise based on continuous nursing management can improve the nerve function and limb motor function of patients and improve the balance control ability of their limbs, which is worthy of clinical promotion and application.
5.Effect of PI3Kδ inhibitor CAL-101 on myeloma cell lines and preliminary study of synergistic effects with other new drugs.

OBJECTIVETo investigate the proliferation inhibitory role and mechanism of PI3Kδ inhibitor CAL-101 on multiple myeloma (MM) cells, and to provide new therapeutic options for MM treatment.
METHODSMM cell lines U266 and RPMI8226 cells were treated with various concentrations of CAL-101. MTT assay and CalcuSyn software were performed to determine the inhibitory effect of CAL-101 and the synergistic effect with PCI- 32765, SAHA (suberoylanilide hydroxamic acid), BTZ (Bortezomib) on MM cells. The protein expression level of p-AKT, p-ERK, AKT, ERK and PI3Kδ processed by CAL-101 were analyzed by Western blot.
RESULTSCAL-101 at concentration of 15, 20, 25, 30 and 40 μmol/L could induce significant dose-dependent proliferation inhibition on U266 cells after treatment for 48 hours. The cell proliferation inhibition rates were (33.54 ± 1.23)%, (41.72 ± 1.78)%, (53.67 ± 2.01)%, (68.97 ± 2.11)% and (79.25 ± 1.92)%, respectively. Similar results were found in RPMI8226 cell line. Western blots showed high expression level of p-AKT, p-ERK, AKT, ERK and PI3Kδ in cell lines and MM primary cells. p-AKT and p-ERK protein expression levels were down-regulated significantly by CAL-101 treatment. Synergistic effect has been verified between CAL-101 and PCI-32765, SAHA and Bortezomib in U266 cell line, and PCI-32765, Bortezomib in RPMI8226 cell line with CI values less than 1.
CONCLUSIONCAL-101 could inhibit proliferation of MM cell lines. High levels of p-AKT, p-ERK, AKT, ERK and PI3Kδ protein expression were observed in both cell lines and primary cells. Down-regulation of p-AKT and p-ERK probably related with the mechanism of CAL-101 in MM cell proliferation inhibition. CAL-101 has significant synergistic effect with PCI-32765, SAHA and BTZ.
