1.Bilateral stereotactic radiofrequency amygdalohippocampectomy for the treatment of bilateral medial temporal lobe epilepsy

Objective To retrospectively analyze the effect and safety of bilateral stereotactic radiofrequency amygdalohippocampectomy (SAHE) for treatment of bilateral medial temporal lobe epilepsy (BMTLE). Methods Twelve BMTLE patients were treated with bilateral SAHE under limited coagulations. Clinical parameters were evaluated with the programs of Engel′s classification, Liverpool Seizure Severity Scale (LSSS) 2.0, Wechsler Adult Intelligence Scale-Revised (WAIS-R) and Wechsler Memory Scale-Revised (WMS-R), respectively. Results Five patients (42%) were assessed as EngelⅠwith 12 ~ 62-month follow-up. Seizure severity scores were declined sharply compared with the baseline of the patients with out seizure free. Function of memory and intelligence was transiently declined without statistical significance immediately after operation (P >0.05), but was significantly increasedat 6 months after operation (P < 0.05). Conclusion Bilateral SAHE could terminate seizures or reduce seizure severity in patients with BMTLE. Under the circumstance of limited coagulations, neuropsychological function was improved along with seizure control.
2.The effects of aloe juice on SOD, GSH-PX , MDA and skin wound healing in scalded rats

Objective To observe the effects of aloe juice on skin wound healing in scalded rats and scavenging the oxygen free radical in their bodies. Methods 24 rats were divided into 3 groups,namely, aloe group, burn cream group and blank control group. And burn wounds ( Ⅱ degree deep) were created with a diameter of 2. 6cm in their backs, respectively. Smear Burn Ointment and Aloe Vera juice,were used to observe the time of the appearance of their epithelial tissues and the time of the wound healing,and the content of SOD, GSH-PX and MDA in healed skin surfaces were detected. Results The time of the appearance of epithelial tissues[(4.6 ±0. 56)d ,(16.2±2.6),(5.4±0.6)d,(18. 1 ±3.4)d,(6.8± 0. 3) d , (22. 3 ± 3.2 ) d], the wound healing and the content of SOD [( (98.07 ± 6. 22) vs(83.97 ± 6. 34), (57. 50 ± 9. 43 )], GSH-PX [(243.21 ± 20. 18)vs (208.25 ± 24. 52), ( 139. 88 ± 22. 70)] and MDA [(4. 89 ±2. 12) vs (6. 93 ± 3.05 ), (8.98 ± 2. 14)] in aloe group have significant difference compared with the blank group( P <0.01, 0.001, 0.005), but no significant difference compared with the burn cream group. Conclusion Aloe juice has the effects of promoting the skin wound healing of scalded rats ,scavenging the oxygen free radical in their bodies and protecting cells.
3.Identification of phosphoproteome in mice neurons by isotope-labeling technique combining with chemiluminescence Western blotting method.

This study was aimed to work out a simple, applicable, sensitive and specific protocol for the identification of phosphoproteome. Isotope-labeling, two-dimensional electrophoresis, autoradiography and so on were used to establish a phosphoproteome map of mice neurons, and then chemiluminescence Western blotting was utilized to detect three phosphoproteins PI3Kr3, MEK1 and PKCalpha selectively. The results of comparison showed that the blots of PI3Kr3, MEK1 and PKCalpha on autoradiography map were almost identical with the blots of PI3Kr3, MEK1 and PKCalpha on chemiluminescence Western blotting maps. So this protocol based on isotope labeling and chemiluminescence Western blotting methods has proven to be sensitive and specific in the identification of phosphoproteome.

4.IL-34 Aggravates Steroid-Induced Osteonecrosis of the Femoral Head via Promoting Osteoclast Differentiation

IL-34 can promote osteoclast differentiation and activation, which may contribute to steroidinduced osteonecrosis of the femoral head (ONFH). Animal model was constructed in both BALB/c and IL-34 deficient mice to detect the relative expression of inflammation cytokines. Micro-CT was utilized to reveal the internal structure. In vitro differentiated osteoclast was induced by culturing bone marrow-derived macrophages with IL-34 conditioned medium or M-CSF. The relative expression of pro-inflammation cytokines, osteoclast marker genes, and relevant pathways molecules was detected with quantitative real-time RT-PCR, ELISA, and Western blot. Up-regulated IL-34 expression could be detected in the serum of ONFH patients and femoral heads of ONFH mice. IL-34 deficient mice showed the resistance to ONFH induction with the up-regulated trabecular number, trabecular thickness, bone value fraction, and down-regulated trabecular separation. On the other hand, inflammatory cytokines, such as TNF-α, IFN-γ, IL-6, IL-12, IL-2, and IL-17A, showed diminished expression in IL-34 deficient ONFH induced mice. IL-34 alone or works in coordination with M-CSF to promote osteoclastogenesis and activate ERK, STAT3, and non-canonical NF-κB pathways. These data demonstrate that IL-34 can promote the differentiation of osteoclast through ERK, STAT3, and non-canonical NF-κB pathways to aggravate steroid-induced ONFH, and IL-34 can be considered as a treatment target.
