1.Research progress of a new therapeutic target in hepatocellular carcinoma glypican-3

Glypican-3 ( GPC3 ) plays very important role in the regulation of cell growth and differentiation in hepatocellular car-cinoma ( HCC ) . GPC3 is closely related to the occurrence and development of HCC. A dramatic elevation of GPC3 expression has been reported in a large proportion of HCC, which suggests that GPC3 is remarkably sensitive and specific to the diagnosis of HCC. GPC3 is a potential therapeutic target of HCC. This paper reviews the structure and function of GPC3, the progress of im-munotherapy based on GPC3 of HCC, and discusses the prospect of therapeutic target of liver cancer in the future.
2.Preliminary Study of Immunodiagnosis of Trichinosis by ELISA Using Urea Soluble Antigen of Trichinella spiralis

Urea soluble antigens of Trichinella spiralis infective larva were prepared and the efficiency of the antigens in diagnosis of trichinosis by ELISA are studied. Specific IgG antibodies are detected by ELISA using urea soluble antigens in 91.67%(22/24) of 24 sera and 91.67% (ll/12)of 12 blood spots of filter paper of patients of trichinosis. Specific IgM antibodies are seen in 95.24% (20/21) of 21 sera of patients using the antigens. Sera of patients infected with Schistosoma japonicum have some cross reactions on the antigens. Sera from mice infected with - Schistosoma japonicum, Ancyclostoma caninum larva, and Plasmodium yoelii are negative. The urea soluble antigen has a relatively high sensitivity, specificity and good reproducibility in diagnosis of trichinoisis by ELISA, and has a practical and economic value.
3.Effects of granulocyte-macrophage colony-stimulating factor (GM-CSF) on the anti-fertility efficacy of canine zona pellucida glycoprotein 3 (CZP3) DNA vaccine

Objective To investigate the effects of GM-CSF (granulocyte-macrophage colony-stimulating factor) as an immune adjuvant on the anti-fertility efficacy of canine zona pellucida glycoprotein 3 (CZP3) DNA vaccine.Methods RT-PCR and quantitative real-time PCR were performed to analyze the effects of GM-CSF on the maturation of antigen presenting cells (APCs) and the enrichment of APCs at injection site.Female BALB/c mice were immunized with CZP3 DNA vaccine pcDNA3-CZP3 alone or in combination with genetic adjuvant pcDNA3-GM-CSF by using the method of electrical impulses.ELISA (enzyme-linked immunosorbent assay) was used to detect the levels of IgG and sIgA (secretory IgA) antibodies in serum samples and genital tracts and the levels of IL-4 and IFN-γ in serum samples.MTT method was used to analyze the proliferation of spleen T cells in mice.The binding ability of serum anti-CZP3 antibody to native mouse oocytes was analyzed by indirect immunofluorescence assay.All of the female BALB/c mice were coupled with male mice of the same age six weeks after the first-dose vaccination.Litter size at birth in each group was counted and the differences between different groups were comparatively analyzed.ResultsImmunizing the mice with pcDNA3-CZP3 and GM-CSF significantly promoted the expression of CD80, CD83 and CD86 (P<0.05) and increased the sIgA antibody level in genital tract and IgG level in serum (P<0.01).Moreover, the levels of IL-4 and IFN-γ in serum samples were significantly up-regulated (P<0.05) and the proliferation of spleen T cells was significantly enhanced (P<0.05).Results of the indirect immunofluorescence assay showed that the fluorescence intensity and density on mouse egg surface were positively correlated with the level of antibody in serum.Results of the anti-fertility test suggested that GM-CSF significantly reduced the litter size in mice immunized with pcDNA3-CZP3 vaccine (P<0.05).Conclusion GM-CSF could be used as an effective adjuvant to enhance the anti-fertility efficacy of CZP3 DNA vaccine.
4.The study on gastrointestinal transit and changes of myenteric nerve plexus in acute necrotic pancreatitis rats complicated with hyperlipemia

Objective To explore association between the changes of myenteric nerve plexus and the delayed gastrointestinal transit in acute necrotic pancreatitis (ANP) rat complicated with hyperlipemia. Methods Atotal of 40 male Sprague-Dawley (SD) rats were randomly divided into 4 groups (normal control group, ANP group, hyperlipemia (HL) control group and HL with ANP group). HL rat model was established by fed with high-fat diet for 4 weeks, and ANP rat model was induced by retrograde injection of 3.5% sodium taurocholate into pancreatic duct. Gastrointestina1 transit distance was measured by ink gavage method. The histological changes of cholinergic and nitriergic nerves in myenteric plexus were observed by Karnovsky-Root method and NADPH histochemistry method. Results Pathological injuries were more severe in HL with ANP group than in ANP group (12.8±0.63 vs. 10.8±1.93,P<0.01), gastrointestinal transit was obviously delayed (transmission rate was 27%±5% vs. 38%±6%,P<0.01), the density of cholinergic neurons decreased (4.80±1.23 vs. 5.80±0.79, P<0.05), and the density of nitriergic neurons significantly increased (8.70±0.75 vs. 6.80±1.48, P<0.01). There was a linear regression between changes of cholinergic and nitrievgic nerves in myenteric nerve plexus and gastrointestinal transit (R2=0.531, P<0.01). Conclusion There was significant gastrointestinal motility disorder in the ANP rat complicated with hyperlipemia, which may be closely related with the changes of myenteric nerve plexus.
5.Recent Advances in Research of Antifreeze Proteins

Many overwintering organisms produce antifreeze proteins (AFPs) that can be adsorbed onto the surface of ice crystals and modify their growth. These proteins show great diversity in structures, and they have been found in a variety of organisms. AFPs from insects have higher thermal hysteresis activity than other organisms. Recent studies revealed the structures of AFPs and put forward different ice-binding models. No mechanism, however, can apply to all antifreeze proteins and the molecular interaction between AFPs and ice are not accurately resolved. AFPs can be applied extensively to agriculture, aquaculture and low temperature storage of organs, tissues, as well as cells. To confer transgenic plant cold resistance application of AFPs is essential, while the expression and regulation of antifreeze gene need to be elucidated.
6.Effect of lipide diet on the pharmacokinetics of levofloxacin in Beagle dogs

Objective To study the effect of lipide diet on pharmacokinetics of levofloxacin (Lfox) in beagle dogs .Methods In randomized cross over experiment ,6 Beagle dogs were given Lfox 10 mg/kg at the condition of empty stomach or with lipide diet for 2 week .Plasma concentration of Lfox was determined by HPLC at different time (during 48 h) .The pharmacokinetic parameters were calculated by 3p97 software .Results The plasma concentration-time curves were two compartment modelat the condition of empty stomach or lipide diet .Main pharmacokinetic parameters were as follows :t1/2β:(11 .07 ± 8 .46)and(21 .79 ± 4 .59)h ;tmax :(3 .42 ± 0 .41) and(2 .00 ± 0 .32)h;Cmax :(2 .71 ± 0 .60)and(2 .52 ± 0 .38)μg/mL ;AUC0→ t :(36 .51 ± 8 .66) and(37 .19 ± 8 .21)μg · h-1 · mL -1 ,respectively .There were significant differences in pharmacokinetic parameters of tmax (P<0 .01)except for t1/2β、Cmax and AUC0→ t .Conclusion Lipide diet can quicken tmax ,has no effect on the absorption of Lfox .
7.Relationship Between Arterial Stiffness and Variability of Blood Pressure in Senile Patients With Hypertension

Objective: To investigate the relationship between arterial stiffness and variability of blood pressure (BPV) in senile patients with hypertension.
Methods: A total of 234 senile patients with hypertension were enrolled. The patients were examined by 24h ambulatory blood pressure monitoring (ABPM), BPV was identified by the standard deviation (SD) of blood pressure value. Based on increased percentage of baPWV, the patients were divided into 4 groups: Normal artery group, n=44, Mild arterial stiff group, n=72, Moderate arterial stiff group, n=78 and Severe arterial stiff group, n=40. The 24h ABPM results, BPV, biochemical indexes and their relationship to baPWV were compared among different groups.
Results:①The HDL-C level and 24hDBP, dDBP, 24hSBPSD, 24hDBPSD, dSBPSD, dDBPSD, nDBPSD were significantly different among each group, all P<0.05.②The increased percentage of baPWV was positively related to the age and 24hSBPSD, 24hDBPSD, dSBPSD, dDBPSD, nDBPSD, while negatively related to HDL-C level, all P<0.05.③The age and 24hSBPSD, 24hDBPSD, dSBPSD were the independent risk factors for baPWV elevation.
Conclusion: The arterial stiffness was closely related to BPV in senile patients with hypertension.
8.Apoptosis of human gastric carcinoma AGS cells induced by cecropinXJ

Aim To investigate the apoptosis of human gastric carcinoma AGS cells induced by cecropinXJ. Methods Human gastric carcinoma AGS cells and human normal epithelial cells GES-1 were co-cultured with different concentrations of cecropinXJ ranging from 0. 01 to 1 000 mg·L-1 for 24 h. MTT assay was used to observe the effects of cecropinXJ on the proliferation of AGS cells and GES-1 cells. The ultrastructural changes of the AGS cells were observed by transmission electron microscopy. Hoechst staining was used to de-tect cell apoptosis. The changes of intracellular reac-tive oxygen species ( ROS) and mitochondrial potential were analysed by flow cytometery. The expression of Bax, Bcl-2, caspase-3 and cytochrome C in mRNA level was investigated by qRT-PCR. Western blot was used to determine the protein expression of Bax, Bcl-2, caspase-3 and cytochrome C. Results CecropinXJ significantly suppressed the proliferation of AGS cells in vitro (P<0. 05) in a dose-dependent manner, IC50 =61. 19 mg·L-1 , but had no inhibitive effects on the proliferation of GES-1 cells. After treatment for 24 h, cecropinXJ induced AGS cells nuclear condensation, and increased ROS production, disrupted mitochondri-al integrity. The results of qRT-PCR and Western blot demonstrated cecropinXJ could up-regulate the expres-sion of Bax and down-regulate the expression of Bcl-2 , promote the release of cytochrome C and activate caspase-3. Meanwhile, cecropinXJ promoted caspase-3 activity in a dose-dependent manner, and cell death ratio of AGS cells induced by cecropinXJ was signifi-cantly reduced by caspase-3 and caspase-9 specific in-hibitors treatment. Conclusion CecropinXJ can in-duce apoptosis of AGS cells by downregulating Bcl-2 , upregulating Bax and activating caspase-3 , which may be one of its anti-tumor mechanisms.
9.Clinical analysis of 54 serious severe acute respiratory syndrome patients with multiple organ injury

Objective To study the clinical characteristics of the severe acute respiratory syndrome(SARS) patients who progressed into multiple organ injury Methods Retrospective analysis was performed on the clinical data of 54 severe SARS patients with multiple organ injury in our hospital Results In 54 SARS patients,systemic inflammatory response syndrome(SIRS) developed in all cases and multiple organ injury developed in 49 cases and the mortality of patients with multiple organ injury was 22 2% Among the involved organs,lungs were the most frequently involved organ,followed by liver,heart,brain,kidney and blood system The injury in other organs was rare Conclusions Serious SARS is predisposed to progress into multiple organ injury The mortality was correlated to the number of organs involved Recognization and treatment of multiple organ injury at early stage are critical with improve cure rate of serious SARS with multiple organ injury
10.Myocardial protection of L-carnitine in patients with acute myocardial infarction undergoing primary percutaneous coronary intervention

50% resolution of elevated ST-segment in 1h after PCI was increased in the L-CN treated group. LVEDD was less but LVEF was higher in the treatment group compared with the control group 3 months after PCI. Conclusion L-CN has significant myocardial protection effects in patients with AMI undergoing primary PCI, which is characterized by inhibition of the reperfusion injury , reduction of infarction area and improvement of left ventricular remodeling.
