Objective To investigate the effect of therapeutic dual-plasmid HBV DNA vaccine on specific humoral and cellular immunity in cancrivorous monkey (Macaca fascicularis). Methods The eukaryotic expression plasmids encoding human IL-2 and IFN-? fusion protein (pFP) were constructed to enhance the cellular immunity of therapeutic HBV DNA vaccine (pS2.S) encoding HBV envelope middle protein in the form of dual-plasmid (pS2.S+pFP). Thirty cancrivorous monkeys were randomly divided into 5 groups (6 each) with sex rotio of 1∶1, namely the dual-plasmid DNA vaccine group in high dosage (2000?g/kg), medium dosage (660?g/kg), low dosage (200?g/kg), and EP-mediated pFP (330?g/kg) or PBS administration groups as the controls. The monkeys were vaccinated repeatedly with the dual-plasmid HBV DNA vaccine, and then the immune responses including level of serum anti-HBs and number of IFN-? secreting T-cells induced by HBsAg were examined by means of enzyme-linked immunosobent assay (ELISA) and enzyme-linked immunosobent spot assay (ELISPOT) respectively. Results Ten weeks after immunization of HBV DNA vaccine, various levels of serum anti-HBs was detected in all the monkeys of three different dose groups. Sixteen weeks after administration of EP-mediated HBV DNA vaccine, the positive HBsAg-specific INF-? T cell responses was found in 3, 2 and 3 out of 3 monkeys, respectively in the high, medium and low losage groups, and HBsAg-specific INF-? T cell responses were positive in all the animals with the respective cell count of 30.0?13.5 SFCs/3?105 PBMCs, 30.7?26.3 SFCs/3?105 PBMCs and 17.7?6.4 SFCs/3?105 PBMCs in each corresponding group at the 29th week. However, HBsAg-specific INF-? T cell responses were negative in the pFP and PBS group at the same time. Conclusion Electroporation-mediated vaccination of the HBV DNA vaccine can effectively induce both humoral and cellular HBsAg-specific immune responses in cancrivorous monkeys.

Objective:To explore the relationship between neuroticism and depression, and the mediating effect of coping style. Methods:A total of 274 prision inmates of Fujian province were tested by self-rating depression scale, Eysenck personality questionnaire and coping style questionnaire.SPSS 23.0 was used for data analysis, and Mplus 7.0 was used for structural equation modeling and Bootstrap mediating test.Results:(1) The scores of neuroticism, positive copying style, negative coping style and depression were 11.08±5.51, 32.47±6.45, 28.81±5.96, 41.15±9.07, respectively.Neuroticism was negatively correlated with positive copying style( r=-0.40, P<0.01) and positively correlated with negative coping style( r=0.39, P<0.01) and depression( r=0.69, P<0.01). (2)Neuroticism positively predicted depression and could explain 46.4% variation of depression score( F=46.372, P<0.01). (3)Coping styles (positive coping style and negative coping style) had partial mediating effects between neuroticism and depression. The whole model was well fitted (χ 2/ df=2.364, RMSEA=0.070, NFI=0.993, CFI=0.996, RFI=0.955, IFI=0.996), and the effect size of positive coping style and negative coping style were 0.108(95% CI: 0.061-0.157) and 0.066(95% CI: 0.021-0.109). Conclusion:Neuroticism directly affects depression and indirectly affects depression by means of positive coping style and negative coping style.

Objective To observe the influence of high-salt intake on the blood pressure, weight of left ventricle, myocardial microRNA133a and fibrosis levels by implementing the high-salt diet intervention trial in Dahl salt-sensitive rats and SD rats so as to explore the role of microRNA133a in salt-sensitive hypertensive cardiac fibrosis with high-salt intervention. Methods Sixteen 4-week male Dahl salt-sensitive rats are divided into two groups,8 in the SLS group and 8 in the SHS group.Similarly, sixteen 4-week male SD rats were divided into two groups. Systolic blood pressure of the rats was measured by the indirect tail-cuff method.Left ventricular mass index was measured after the rats were sacrificed. The expression levels of left ventricular myocardial tissue collagen I (LVMI),connective tissue growth factor (CTGF)expression and microRNA133a were observed. Results Blood pressure in high-salt groups both increased (P<0.01).Compared with that of SD rats, blood pressure was more significantly increased in high-salt group than in low-salt group of Dahl salt-sensitive rats (P<0.01).After the intervention of high-salt diet,LVMI,CTGF and collagen I expression in Dahl salt-sensitive rats increased more significantly than the low-salt group, with a significant difference (P<0.01);however,there was no significant difference between high-salt and low-salt groups (P>0.05).After the intervention,microRNA133a expression was significantly decreased in all high-salt groups compared with that in low-salt groups (P<0 .0 1 ),and the expression decreased more significantly in high-salt group of Dahl salt-sensitive rats than in high-salt group of SD rats (P<0.05).Conclusion High-salt diet is probably involved in salt-sensitive hypertension myocardial fibrosis by downregulating the expression of myocardial microRNA133a.

Tianjin Medical Journal 2009;37(12):1026-1028
Objective: To investigate the changes of urinary S100B protein concentrations and their relationship with brain damage in preterm infants there of . Methods: The urinary S100B protein of 84 preterm infants and 26 full term infants, which were used as control, were measured at 24 h and 120 h after birth. At the same time, routine clinical observations, neurologic patterns and ultrasound screens were recorded. The value of urinary S100B protein and brain damage were evaluated in preterm infants with different gestational age. Results: The differences of urinary S100B protein were statistical significance between the different gestations. The levels of urinary S100B protein were higher in preterm infants, whose gestations were lower than 32 W, than those of other groups. The levels of S100B protein were significantly higher in samples of 27 peri-intraventricular hemorrhage (PIVH) and 3 peri-ventricular leukomalacia(PVL) than those in samples without brain damage( P < 0.05). The S100B levels were significantly higher in urine of 10 preterm infants with polycethemia than those in infants without brain damages. In addition, the S100B levels were different in urine of preterm infants with different prognosis. The S100B levels were significantly higher in urine of infants who died or deteriorated than those of others(P < 0.05). Conclusion: There is an evident trend of decrease in urinary S100B protein concentration with increasing gestational age. It will be helpful to identify preterm infants with PIVH,PVL and high risk of brain damages by measurement of S100B protein in urine early after birth, which indicates further inspection, provides protective treatment and enhances follow up.
