1.The effect of folic acid on DNA methylation of tumor-related genes in healthy human peripheral blood mononuclear cells

Objective To investigate the effect of folic acid on the DNA methylation of tumorrelated genes promoters in healthy human peripheral blood mononuclear cells(PBMC). Methods Ten healthy volunteers were divided into two groups, and were randomized to receive either 5 mg folic acid (n=5)or placebo(n = 5) , one time per day for 3 months. The serum folic acid concentration was detected with chemiluminescence enzyme immunoassay kit before and after the intervention. The methylation statuses of five tumor-related genes promoter, including oncogenes c-myc, c-Ha-ras,tumor suppressor genes p16INK4A, E-cadherin and mismatch repair gene hMLH1 in PBMC were detected by bisufite sequencing. Results After folic acid intervention, the level of serum folic acid increased significantly in intervention group (t= -4. 739,P<0. 05) , however no significant difference in control group. After three-month folic acid intervention, the level of methylation of oncogene c-myc promoter increased from 4%, 3. 3%, 4. 1% before intervention, one week after intervention, one month after intervention respectively to 8%(t= -4. 079,P<0. 05), while no significant change in placebo taken group. Before and after the folic acid intervention, there was no significant difference of DNA methylation of other tumor-related genes promoter, including c-Ha-ras、E-cadherin、p16INK4Aand hMLH1. Conclusion Folic acid intervention can up-regulate DNA methylation of oncogene c-myc promoter, but can not affect the promoter methylation status of tumor suppressor genes E-cadherin,p16INK4Aand hMLH1.
2.Clinical analysis of alleviative treatment in 42 patients of advanced gastrointestinal cancer

Objective To study the short-term clinical efficacy of treating patients with advanced gastrointestinal cancer with lentinan injection and javanica oil emulsion injection.Methods Clinical information of 42 patients with advanced gastrointestinal cancer were retrospectively collected.The 42 patients were divided into two groups according to treatments,with 21 case in the control group who were treated with javanica oil emulsion injection,as well as 21 case in the treatment group treated with lentinan injection and javanica oil emulsion injection.The efficacy,quality of life (QOL) and adverse effects were observed after treatment for 3 weeks.Results 81.0% (17/21)of patients in the treatment group improved in QOL,which was much higher than that in the control group 47.6% ( 10/21 ) ( x2 =5.081,P =0.024 ).The objective remission rate was 19.0% (4/21)and 14.3% (3/21)in the treatment group and the control group respectively,with no significant differece bwtween the two groups( x2 =0.171,P =0.679 ).the disease control rate was 85.7% (18/21)in the treatment group,which was significantly higher than that of 61.9% (12/21)in the control group( x2 =4.200,P =0.040 ).The incidence of adverse effect related to hematological toxicity,liver and kidney function,the digestive tract and itching of skin were similar between the two groups (Ps > 0.05 ).Phlebitis in the treatment group was not as frequent as that in the control group(P <0.05).Conclusion Treating patients with advanced gastrointestinal cancer with lentinan injection and javanica oil emulsion injection had high efficacy than treating only with javanica oil emulsion injection,and it improved QOL signifiantly with safety.
3.Experimental occlusal interference induces the expression of protein gene products and substance P in masseter muscles of rats

Objective:To investigate the peripheral mechanism by studying the histological changes of masseter muscles using HE stains and substance P(SP) and protein gene product 9.5(PGP9.5) immunohistochemical stains.Methods: Fifteen male Sprague-Dawley were randomly assigned into occlusal interference group(n=12) and control group(n=3).In occlusal interference group,0.4 mm thick crowns were bonded to the rats'first molar of the maxillary.In the control group,rats were anesthetized and mouths were forced open for about 5 min but restorations were not applied.1,5,10,and 21 d after 0.4 mm occlusal alteration treatment,mechanical pain thresholds of bilateral masseter muscles were quantitatively measured by modified electronic anesthesiometer in control group and occlusal interference group.The rats were euthanized by transcardiac perfusion after deep anesthetization at different time points.The paraffin sections of masseter muscles were made and processed for HE,SP,and PGP9.5 immunohistochemical staining.Results: Decreased head withdrawal threshold to mechanical pressure was detected in masseter muscles on both sides following occlusal interference.Histological stains of masseter muscles presented intact following occlusal interference,and no inflammatory cells were observed in both sides.Intensely stained PGP9.5 was observed at 1 d in occlusal interference groups and maintained until the end of the experiment.SP expression was the most obviously increased at 5 d in both sides and gradually decreased to the level of control.Conclusion: Experimental occlusal interference-induced masticatory muscle pain is associated with peripheral sensitization of nociceptive neurons rather than muscle damage and inflammation.
4.Determination of genpioside,baicalin and berberine hydrochloride in Gonglao Quhuo Tablets simultaneously by RP-HPLC

AIM: To establish a method of determining genpioside,baicalin and berberine in Gonglao Quhuo Tablets(Caulis Mahoniae,Cortex Phellodendri Chinensis,Radix Scutellariae,etc). METHODS: The mobile phase composed of A acetonitrile,B acetonitrile and 0.5% triethylamine(Ph3.1)(10∶90).The flow rate was 1.0 mL/min with the wavelength at 254 nm. RESULTS:The calibration curves were linear in the ranges of 3.22-206 ?g/mL(r=0.999 9) for genpioside,2.95-186 ?g/mL(r=0.999 9) for baicalin and 1.64-104 ?g/mL(r=0.999 9) for berberine hydrochloride,respectively.The average recoveries were not less than 98.4%. CONCLUSION: This HPLC method is convenient,rapid,accurate and could be used for quality control of production of Gonglao Quhuo Tablets.
5.The application of 18F-FDG PET/CT in diagnosis of neuroblastoma in children

Objective To study the clinical value of 18F-FDG PET/CT in childhood neuroblastoma (NB). Methods 18F-FDG PET/CT was performed in 31 children diagnosed with NB. According to the treatment conditions, patients were divided into pre-therapy group, radiation and chemotherapy group, postoperative group, respectively. The positive rate, sites, and im-age characteristics of the primary lesion and metastasis lesion were analyzed through qualitative and quantitative analysis of the image. Results Twenty-one (67.7%) and fourteen (45.2%) patients were found positive in primary sites by CT and PET respectively. All cases (9/9, 100%) in pre-therapy group were found positive in primary lesions by PET, 3 positive cases (75.0%) in radiation and chemotherapy group and 2 positive cases (11.1%) in postoperative group. Twenty-one patients showed metas-tases (67.7%). Lymph nodes (16 cases) and bone (bone marrow) (13 cases) were the most common sites of metastasis followed by pleura, meninges, liver and retrobulbar inifltration, all of which showed increased FDG uptake. Two patients were found lesions in lungs by CT, but had no FDG uptake. SUVmax of primary lesions was signiifcantly different among pre-therapy, chemotherapy and postoperative group (H=13.89, P=0.001), and pre-therapy group had the highest value. Metastases (lymph nodes, bone and bone marrow, pleura, liver and meninges) in pre-therapy group had high SUVmax. Conclusions NB primary tumors are characterized by the increased FDG metabolism. PET can fully detect the distribution of NB metastases in whole body. Except the pulmonary metastasis, metastases in other positions show increased FDG uptake. PET has potential role in evaluating the efifcacy of radiation and chemotherapy, and identifying postoperative residual or recurrence.
6.High-performance liquid chromatographic determination of floxuridine in human serum

OBJECTIVE To develop an HPLC method for the determination of floxuridine in human serum.METHODS With metronidazole as the internal standard,200 μL of serum was extracted by n-propyl alcohol/methyl t-butyl ether in a two-step extraction.The organic layer was evaporated under nitrogen stream and the residual was reconstituted with the mobile plase.A Shim-Pack CLC-ODS column was selected and the mobile phase was consisted of acetonitrile-phosphate buffer-water (75∶100∶900) at a flow rate of 0.6 mL*min-1.The detection wavelength was 268 nm.RESULTS A linearitywas obtained from 0.005 to 0.5 mg*L-1 of floxuridine in serum with a good correlation coefficient (r=0.9999,n=8).The intra-run and inter-run coefficients of variation were less than 4.09%.The mean recoveries were 103.00%,107.00% and 100.88% for the low,middle and high concentrations of check samples,respectively.The limit of detection was 0.001 mg*L-1.CONCLUSION The method was sensitive,specific and simple.It is suitable for clinical pharmacokinetic study.
7.Preliminary analysis of hippocampal volume measurement based on magnetic resonance imaging in 68 cases

Objective To investigate the volume of hippocampal formation in normal adult using coronal magnetic resonance imaging. Methods 3D-fSPGR sequence was used to depict the brain in 68 healthy adult. The volume of hippocampus was calculated by drawing the outline of 10 coronal hippocampal formation images acquired equally form posterior border of rostrum corpus callosum to anterior border of the splenium. Data were analysed using the SPSS 17.0 software. Results In the coronal plane images of normal adult brain, the absolute volume of left and right hippocampal formation were 2 319.63-2 610.73 mm3 and 2 447.52-2 749.50 mm3 respectively . The relative volume of left and right hippocampal formation were 2 319 . 87-2 602 . 47 mm 3 and 2 443.96-2 755.89 mm3. There were no correlation between hippocampal volume and age (r = 0.084, P = 0.549. Significant gender differences (t=2.500, P=0.029) were observed between absolute volume of right hippocampal formation in the youth group. There were significant differences in the absolute volume (t = -2.571, P = 0.022), relative volume (t = 2.600, P = 0.021) among the right and left hippocampal formation. Significant absolute volume differences (P = 0.038) were observed between the middle-aged group and the youth group among the hippocampal formation of women. Conclusion No significant differences were observed in age, gender among the hippocampal volume of normal adult, and there was a significant difference between the left and right hippocampal formation volume.
