4.Effects of pulmonary arterial endothelium cells injuryed by tumor necrosis factor ? on proliferation of pulmonary arterial smooth muscle cells and the interference effects of heat stress response

Objective To investigate the effects of pulmonary arterial endothelium cells(PAEC) injuryed by tumor necrosis factor ? (TNF-?) on the proliferation of pulmonary arterial smooth muscle cells (PASMC) and the influence of heat stress response (HSR) on it .Methods Normal PAEC or PAEC endured with HSR were incubated with TNF-? at the concentrations of 500, 1000 and 2000u/ml in 1 hour, then cultured in DMEM without serum in 24 hours, the upper liquid was collected to prepare the endothelial cell-conditioned medium liquid (EC-CMⅠ or EC-CMⅡ ), in which PASMC were incubated in 24 hours as group Ⅰ or group Ⅱ respectively. The normal endothelial cell-conditioned medium liquid was also prepared to incubate PASMC in 24 hours as group Ⅲ.The PASMC were incubated without the endothelial cell-conditioned medium liquid as group Ⅳ.Flow cytometry was applied to determining the intracellular DNA content of the incubated PASMC. The fractions of different cytocycle phases were calculated according to the areas under the curves of DNA content.Results Compared with those of group Ⅳ, the percentage of PASMC in G0-G1 phases increased markedly ,and in S and G2-M phases decreased significantly in group Ⅲ (P

6.The effect of gingkgo biloba on cytokine levels and clinical efficacy in treatment of patients with asthma

ObjectiveTo observe the effect of extract of ginkgo biloba(EGB) on cytokine levels and clinical efficacy of patients with asthma.Methods A prospective randomized control trial was conducted. 112 patients with asthma in Department of Respiratory Medicine of Affiliated Hospital of Hebei Union University were enrolled. The patients were divided into ginkgo lamina group(58 cases) and conventional therapy group(54 cases) by random number table.According to the Global Initiative for Asthma(GINA) Prevention and Treatment, both groups received conventional therapy, and in addition, every patient in ginkgo lamina group took 2 tablets once of EGB(40 mg/tablet) orally, 3 times a day for 2 weeks. The radioimmunoassay was used to detect the levels of tumor necrosis factor-α(TNF-α), interleukins(IL-6, IL-8, IL-10) in patients of two groups before and after treatment, and the changes of pulmonary functions were observed.Results After treatment, serum IL-6,IL-8 and TNF-α were significantly decreased and IL-10 was increased in patients of both groups, the above changes of indexes in ginkgo lamina group being more obvious compared with those in conventional therapy group〔IL-6(μg/L): 0.15±0.05 vs. 0.23±0.12, IL-8 (μg/L): 0.51±0.24 vs. 1.42±0.54, IL-10(μg/L): 69.18±13.12 vs. 32.61±12.51, TNF-α(μg/L): 1.35±0.59 vs. 2.14±1.52,allP<0.05〕. After treatment, the pulmonary function indexes of patients in the two groups were increased obviously, and the increase in ginkgo biloba group being more significant compared with that in conventional therapy group〔percentage of 1 second forced expiratory volume/predicted value(FEV1%):(68.12±0.38)% vs. (55.32±0.24)%, FEV1/forced vital capacity(FEV1/FVC):(71.32±0.59)% vs.(56.56±0.42)%, percentage of peak expiratory flow rate/predicted value(PEF%):(63.28±0.24)% vs.(52.14±0.24)%, allP<0.05〕. ConclusionGinkgo biloba may improve the airway inflammation in patients with asthma by affecting the levels of serum IL-6, IL-8, IL-10 and TNF-α.
7.Study on pure red cell aplastic lymphocyte subsets characteristics and the effect of cyclosporin A

Objective To detect and analyse acquired pure red cell aplasia (PRCA) T lymphocyte subsets distribution and to assess its condition and cyclosporine immune function of T lymphocytes subsets.Methods Flow cytometry was applicated to detect peripheral blood T lymphocyte subsets of acquired PRCA patients before and after 3 months of treatment with cyclosporine-based immunosuppressive regimen and normal controls.Results Among 22 patients,17 cases of blood returning normal (three cases of bone marrow returning normal),the total effective rate was 95.5 % (3 cases of cure,14 cases of remission,4 cases of improvement,1 case of ineffectiveness).Th (CD3+ CD4+) cells and Th/Ts ratios in acquired PRCA patient group were lower than those in the normal control group,while Ts (CD3+ CD8+) cells was higher than that in the normal control group,the differences were statistically significant (P < 0.05).After 3 months of treatment,Th cells and Th/Ts ratio were higher than those before,and Ts cells were decreased compared with the previous (P < 0.05).Conclusion Disorders of T lymphocyte subsets in acquired PRCA patients lead to immune dysfunction,however,cyclosporine can improve T lymphocyte subsets in patients with imbalance,which is an effective way to treat the disease with significant curative effect and mild adverse reactions.
10.Multiple risk factor analysis of recurrent respiratory tract infection after Mycoplasma pneumoniae pneumonia in children

Objective To investigate the occurrence trend and risk factors of recurrent respiratory tract infection during the ifrst year after Mycoplasma pneumoniae pneumonia (MPP) in children. Methods The clinical data of 133 children complete-ly recoved from MPP and one year follow-up after MPP were included in this study, MPP IgM IgG double antibody titer were measured in different time slots (3 month, 6 months, 9 months, 12 months) after discharge. Information on frequency of recurrent respiratory tract infections, respiratory tract infection site, and drug use within one year were collected. Possible factors affecting the occurrence of recurrent respiratory tract infection were analyzed by means of single factor and multi factor analyses. Results In 133 patients, the recurrent rate was 31.58%in the ifrst year;aged 3-6 years old (OR=2.29,95%CI:1.13~4.64), or continuous positive or negative to positive antibodies (OR=4.47,95%CI:1.47~13.65), or low CD4/CD8 (OR=10.26,95%CI:3.30~31.90), or low IgA (OR=1.90,95%CI:1.06~3.40) is independent risk factor of recurrent respiratory tract after MPP;immune enhancer therapy is an independent protective factor (OR=0.29,95%CI:0.11~0.78). Conclusions Immune function disorders in the ifrst year after MPP were independent risk factors of recurrent respiratory tract infection. MP antibody positive without clinical symptoms and sustained antibody positive can not prevent the recurrence of respiratory tract infection without use of antibiotics. Immune enhancer was advocate to adjust immune function and reduce the incidence of repeated respiratory tract infection.
