Journal of Chinese Physician 2017;19(4):556-559
Objective To explore the contribution of endoplasmic reticulum stress to cardiac myocyte apoptosis in mouse congestive heart failure induced by Doxorubicin. Methods Doxorubicin (DOX)was administered intraperitoneally to rats,2 mg/kg doses over 8 weeks.Echocardiographic and hemodynamics measurements were obtained at 8 weeks after treatment. Then left ventricular (LV) samples were collected. The successfully established rats were divided into 3 groups (Fu Shen Ke Li 40 mg/kg,60 mg/kg,and 80 mg/kg).After 5 weeks post the operation,an echocardiographic was performed on all the rats under narcotism,Western blot to test the B-cell lymphoma-2 (Bcl-2),myeloid cell leukemia-1 (Mcl-1),Bcl-2-Associated X (Bax),Caspase-3,C/EBP homologous protein (CHOP),and 78-kDa glucose-regulated protein(GRP78),and reverse transcription-polymerase chain reaction (RT-PCR) to test the mRNA expression levels of Mcl-1,and NOXA.Results The left ventricular end-diastolic diameter (LVEDD),left ventricular end systolic diameter (LVESD),left ventricular ejection fraction (LVEF) prompted modeling success The left ventricular end-diastolic diameter (LVEDD),left ventricular ejection fraction (LVEF),and fractional shortening (FS) have been improved in high and middle groups of Fu Shen Ke Li.Compared to the model group, higher expression levels of Bcl-2,and Mcl-1 were, lower expressions of Bax,NOXA,CHOP,and GRP78 were (P < O.05) in 3 groups of Fu Shen Ke Li.Compared to the model group, there were higher mRNA expression level of Mcl-1 was, lower mRNA expression of NOXA was (P < 0.05) in 3 groups of Fu Shen Ke Li.Conclusions Fu Shen Ke Li depresses endoplasmic reticulum stress, which can reduce the apoptosis of myocardial cells, and ultimately achieve the purpose of treatment of heart failure.

Objective To investigate the expression of tumor stem cell marker aldehyde dehydrogenase -1 (ALDH-1)in primary tumor and lymph node metastases of lymph node-positive invasive micropapillary carcinoma ( IMPC) of breast and its clinical significance .Methods The expression , location and distribution of ALDH-1 were examined by immunohistochemical staining in 103 cases with lymph node metastasis of IMPC and 110 cases with lymph node metastasis of invasive ductal carcinoma not otherwise specified ( IDC-NOS) without preoperative chemotherapy and radiotherapy , and their relationship with clinicopathological characteristics and prognostic sig-nificance was analyzed .Results ( 1 ) The positive expression rate of ALDH-1 was significantly higher in IMPC group of primary tumor and lymph node metastases (37.9% in primary tumor, 39/103 cases;47.6% in lymph node metastases, 49/103 cases)than in IDC-NOS group(21.8% in primary tumor, 24/110 cases; 23.6% in lymph node metastases, 26/110 cases)(P<0.05).(2)Expression of ALDH-1 had a significantly positive corre-lation with tumor size , histological grade , lymph node metastasis , ER-negative status , PR-negative status , and HER2 overexpression in primary tumor and lymph node metastases of IMPC (P<0.05).(3)The disease-free sur-vival of IMPC group patients was significantly shorter than that of IDC-NOS group patients(P=0.003).In IPMC patients with lymphode metastases , the disease-free survival of patients with ALDH-1 positive in primary tumor and lymph node was significantly lower than patients with ALDH-1 negative expression (P<0.05).In multivari-ate analysis, positive expression of ALDH-1 in metastatic lesions was associated with worse prognosis ( P =0.005).Conclusion ALDH-1 can be used as an independent factor for predicting prognosis of patients with lymph node-positive breast IMPC and the existence of stem cells in IMPC tumor cells may represent high IMPC lymphatic invasion , lymph node metastasis and high resistance .

Objective To compare the clinical efficacy between laparoscopic resection and open resection for rectal cancer and explore the safety and feasibility of laparoscopic resection. Methods The clinical data of 68 patients suffering rectal cancer resection were analyzed retrospectively. The patients were divided into laparoscopic group(31 cases)and open group(37 cases)according to the operation method. Results There was no significant difference in the number of lymph nodes dissection and postoperative complication rate between two groups(P > 0.05).The operation time in laparoscopic group was significantly longer than that in open group [(162.03 ±39.78)min vs.(142.70 ±30.29)min].The bleeding in laparoscopic group was less than that in open group [(153.23 ± 58.94)ml vs.(247.46 ± 92.51)ml].The recovery of intestinal function time and hospital stay in laparoscopic group was shorter than that in open group [(51.39 ±7.28)h vs.(77.81 ±11.68)h,(12.65 ±2.24)d vs.(15.29 ±3.11)d].And there were significant differences between two groups(P < 0.05 or < 0.01).Conclusion Laparoscopic resection for rectal cancer can achieve short-term clinical efficacy similar to the traditional open surgery, and advantages in terms of safety and postoperative recovery worthy of further promotion.

As a cell membrane protein of human body, connexins are closely related to the cell metabolism, proliferation and differentiation, which are highly expressed in lung.Recent studies have shown that connexins play an important role in the lung diseases, such as pulmonary fibrosis, lung injury, lung tumors and so on.This review is to summarize the characteristics of connexins molecules, biological function, the distribution in the lung tissues,as well as the possible regulation mechanisms in the lung diseases.

ObjectiveTo explore the clinical features, etiological factors, diagnosis and treatments in children with hemophagocytic syndromes (HPS).MethodsA retrospective study was carried out to analyze etiological factors, clinical symptoms and sighs, laboratory findings and outcomes of 32 children with HPS in our hospital. Results( 1 ) Etiological factors:virus infection cases accounted for 75.0% (24/32),in which Epstein-Barr virus infection cases were predominant, making up to 68.8% (22/32) ; (2) Main clinical features:included persistent fever, hepatosplenomegaly, lymphadenopathy; ( 3 ) Characteristic laboratory findings:included decrease of three lines or two lines of peripheral blood cells, hyperferrttinemia, abnormal liver function and coagulation function, hypertriglycerdema and decreased fibrinogen, hemophagocytosis in bone marrow, high soluble interleukin-2 receptor level, decreased natural killer cell activity.(4) Treatments and outcomes:twenty-four(24/32) virus infection cases received antivirus and high dose of immunoglobulin treatment.Twenty-two cases were treated with glucocorticoid and cyclophosphamide, resulting in 11 cases improvement,5 cases with complete remission,2 cases giving up and 4 death. The other 10 cases were treated under the guide of HLH-2004 protocol treatmemt with 4 cases improved and 6 cases with complete remission. Conclusion HPS is mainly triggered by infections, especially Epstein-Barr virus infection.Serum ferrifin,interleukin-2 receptor level and natural killer cell activity are biomarkers of the disease,so that they should be monitored during the progress of chemotherapy.HLH-2004 protocol is a good guide for the treatment of this disease.
