1.Effect of Astragalus on T Lymphocyte Immune Function in Immature Rats with Obstructive Jaundice

Objective To evaluate the effect of Astragalus on T lymphocyte immune function in peripheral blood and intestinal mucosa of immature rats with obstructive jaundice. Methods Three-week-old Wistar rats were randomly divided into four groups(n=12,in each):control group, sham operation group, obstructive jaundice(OJ)group,OJ+Astragalus (OJ+A)group.The percentage of CD4 and CD8 T lymphocytes, the ratio of CD4 to CD8 in peripheral blood and intestinal mucosa of immature rats was detected by flow cytometry. Result The percentage of CD4+ cell and the ratio of CD4 to CD8 in the OJ group were lower than those in the control group and the sham operation group,with significant difference(P<0.05).The percentage of CD4 cell and the ratio of CD4 to CD8 in the OJ+A group were higher than those in the OJ group(P<0.05).Conclusion Astragalus can increase T lymphocyte immune function in OJ immature rats.

3.Effect of Danshen-containing serum on expression of SuFu and DYRK2 in HSCs.

To observe the effects of Danshen-containing serum on SuFu and DYRK2 expression in the HSCs stimulated by leptin. SD rats (n = 60) were used to make danshen-containing serum by gastric perfusion for ten days with Danshen water decoction, normal saline and colchicine. The HSCs that were cultured in vitro would be stimulated for 24 hours by leptin (100 μg x L(-1)) except blank control group, after being intervened, the drug serum in each group would be cultured at 37 degrees C in 5% incubator. The cells would be collected after 24 hours, then the effects of danshen-containing serum on the proliferation of HSCs were detected by MTT, the expression of SuFu mRNA and DYRK2 mRNA were detected by RT-PCR, the expression of SuFu and DYRK2 proteins were tested by Western blot. Compared with blank control group, the expression of DYRK2 mRNA and DYRK2 proteins were enhanced obviously after stimulated the HSCs of rats by leptin (P < 0.01), but the expression of SuFu mRNA and SuFu proteins were decreased significantly (P < 0.01). Compared with the model group, after cyclopamine group (Hh pathway inhibitor), Danshen-containing serum and colchicine were interfered, the expression of DYRK2 mRNA and DYRK2 proteins were decreased clearly (P < 0.01), but the expression of SuFu mRNA and SuFu proteins were increased significantly (P < 0.01 or P < 0.05). Compared with model group, adding purmorphamine (Hh pathway agonist) to model group and making it activate could increase the expression of DYRK2 mRNA and DYRK2 proteins, but the expression of SuFu mRNA and SuFu proteins were decreased significantly (P < 0.01). Compared with the model group, using the Danshen-containing serum to interfere the purmorphamine group could make the expression of DYRK2 mRNA and DYRK2 proteins decrease and the expression of SuFu mRNA and SuFu proteins increase significantly (P < 0.01). Danshen-containing serum would inhibition the activation and increment of HSCs by interfering the expression of SuFu and DYRK2 which were induced by leptin.

4.Tankyrase expression in lung bronchiolo-alveolar adenocarcinoma and its relationship with the WNT pathway

Objective To explore the expression of tankyrase (TNKS) and its relationship with WNT/β-catenin signal?ing pathway in lung acinar adenocarcinoma. Methods Seventy-two samples of single subtype alveolar like lung adenocarci?noma (lung adenocarcinoma group) and 67 specimens of normal lung tissue adjacent to carcinoma (adjacent to carcinoma group) were collected. Immunohistochemical method was used to detect expressions of TNKS, beta-catenin (β-catenin) and c-myc protein. The correlation of each protein expression in lung adenocarcinoma tissues was analyzed. The differential ex?pression of TNKS was detected by Western blot assay in two groups. Results Tankyrase protein was mainly expressed in cy?toplasm. The expression ofβ-catenin protein was mainly in cytoplasm and nuclear of lung adenocarcinoma. The expression ofβ-catenin was mainly in cytoplasm, and a small amount was in nuclear of the adjacent group. The c-myc protein was ex?pressed mainly in the nucleus. The positive expression rates of TNKS,β-catenin and c-myc protein were significantly high?er in lung adenocarcinoma group than those of adjacent to carcinoma group (P<0.05). The expression ofβ-catenin in cyto?plasm and nucleus was positively correlated with the expression of TNKS and c-myc (P<0.05). Western blot analysis showed that the relative expression level of TNKS was significantly higher in lung adenocarcinoma group than that of adja?cent to carcinoma group (0.497 ± 0.021 vs. 0.237 ± 0.015, t=13.00, P<0.01). Conclusion Abnormally high expression of TNKS in lung adenocarcinoma may promote the occurrence of lung cancer by regulating the WNT signaling pathways. Inhib?iting TNKS expression may become a new target to treat lung adenocarcinoma.

OBJECTIVES:
To study the value of autotaxin (an autocrine motility factor) level in serum and bronchoalveolar lavage fluid (BALF) in predicting refractory Mycoplasma pneumoniae pneumonia (RMPP) in children and its correlation with interleukin-6 (IL-6), interleukin-8 (IL-8), and C-reactive protein (CRP).
METHODS:
A retrospective analysis was performed on 238 children with Mycoplasma pneumoniae pneumonia who were admitted from January 2019 to December 2021. According to disease severity, they were divided into two groups: RMPP (n=82) and general Mycoplasma pneumoniae pneumonia (GMPP; n=156). The two groups were compared in terms of the levels of autotaxin, IL-6, IL-8, and CRP in serum and BALF to study the value of autotaxin level in serum and BALF in predicting RMPP in children, as well as the correlation of autotaxin level with IL-6, IL-8, and CRP in children with RMPP.
RESULTS:
Compared with the GMPP group, the RMPP group had significantly higher levels of autotaxin, IL-6, IL-8, and CRP in serum and BALF (P<0.05). For the children with RMPP, the levels of autotaxin, IL-6, IL-8, and CRP in serum and BALF in the acute stage were significantly higher than those in the convalescent stage (P<0.05). The receiver operating characteristic (ROC) curve showed that the level of autotaxin in serum and BALF had a good value in predicting RMPP in children, with an area under the curve of 0.874 (95%CI: 0.816-0.935) and 0.862 (95%CI: 0.802-0.924), respectively. The correlation analysis showed that the level of autotaxin in serum and BALF was positively correlated with IL-6, IL-8, and CRP levels (P<0.001).
CONCLUSIONS
The level of autotaxin in serum and BALF increases and is correlated with the degree of disease recovery and inflammatory cytokines in children with RMPP. Autotaxin can be used as a predictive indicator for RMPP in children.

7.Auricular acupuncture for primary insomnia: a systematic review based on GRADE system.

The clinical efficacy and safety of auricular acupuncture (AA) for treatment of primary insomnia was evaluated. After a comprehensive retrieval in domestic and foreign databases, literatures were strictly screened and Revman 5.2 software was applied to perform a Meta-analysis on eligible randomized controlled trials (RCTs). The evidence quality was assessed with GRADE profiler 3.6 software. As a result, 8 articles were included involving 894 patients. Compared among AA and sham AA, placebo AA, blank control, there was significant difference in Pittsburgh sleep quality index (PSQI) [WMD = -3.48, 95% CI (-3.96, -3.00)], sleep latency LWMD = -10.14, 95% CI (-17.16, -3.12)] and sleep awakening times [WMD = -9.98, 95% CI (-1.10,-0.48)]. Compared between AA and western medication, there was significant difference in PSQI [WMD = -3.62, 95% CI (-4.59, -2.65)]. The evidence quality was moderate in AA vs. sham AA, placebo AA or blank control, while that of the rest was extremely low. No reports of adverse events were described in all studies. In conclusion, for the treatment of primary insomnia, AA could effectively improve sleep quality, but due to the low evidence quality, cautious attitude should be taken on this conclusion, and clinical trials with large sample and high quality were needed in the further.

8.Current situation and influencing factors of demoralization syndrome among young and middle-aged female breast cancer patients with radiochemotherapy after radical resection

Objective:To explore the current situation of demoralization syndrome among young and middle-aged female breast cancer patients with radiochemotherapy after radical resection and to analyze its influencing factors.Methods:From January 2019 to September 2019, this study selected 204 young and middle-aged female breast cancer patients of Breast Surgery at a ClassⅢ Grade A hospital in Jilin Province as subjects by convenience sampling. All patients were investigated with the General Information Scale, Demoralization Scale-Mandarin Version (DS-MV) , Medical Coping Modes Questionnaire (MCMQ) and the Social Support Rate Scale (SSRS) . A total of 204 questionnaires were sent out and 192 valid questionnaires were collected.Results:Among 192 young and middle-aged female breast cancer patients, the score of DS-MV was (33.72±14.23) ; the total score of MCMQ was (42.49±8.83) , scores of confronce, avoidance and resignation dimension were (19.12±4.10) , (14.28±3.26) and (9.39±3.48) respectively; the total score of SSRS was (35.39±6.23) . Multiple linear regression analysis showed that the influencing factors of demoralization of breast cancer patients included the age, education level, time after surgery, confronce coping, resignation coping and social support.Conclusions:Demoralization of young and middle-aged female breast cancer patients with radiochemotherapy after radical resection has a high level. Medical staff should provide targeted intervention for patients with young age, low level of education, short time after surgery, facing disease with the resignation coping style and low level of social support so as to improve their physical and mental health.
9.Studies on triterpenoid constituents from Rabdosia japonica var. galaucocalyx.

OBJECTIVETo study the chemical constitutes in Rabdosia japonica var. galaucocalyx .
METHODThe compounds were isolated by nomal phase silica gel chromatography. The structures were identified by physical and spectral data.
RESULTNine triterpenoids were isolated and identified as friedelin (I), 3beta, 28-dihydroxy-ursane (II), ursolic acid (III), 3beta-acetyloxy-ursolic acid (IV), 2alpha, 3alpha-dihydroxy-urs-12-en-28-oic acid (V), 2alpha, 3alpha, 23-trihydroxy-urs-12-en-28-oic acid (VI), oleanic acid (VII), beta-sitosterol (VIII), daucousterol (IX).
CONCLUSIONCompounds I, II, IV, V and VI were isolated from R. japonica var. galaucocalyx for the first time.

10.Preliminary study of 18F-FDG PET-CT in defining lymph node radiation target volume for non-small-cell lung cancer patients.

OBJECTIVETo investigate the value of 18F-FDG PET-CT in detecting metastatic lymph node and radiation planning for patient with non-small-cell lung cancer (NSCLC).
METHODSThe data of 58 NSCLC patients were retrospectively analyzed. Preoperatively, Both 18F-FDG PET-CT and CT scan were performed for all patients. Blinded interpretation of these images was then carried out. The gross tumor volume defined by radiation oncologist based on imaging results of either PET-CT or CT alone was compared with pathological results eventually.
RESULTSThe sensitivity, specificity, accuracy, positive predictive value and negative predictive value of detecting metastatic lymph node in the mediastinum was 56.0%, 54.2%, 54.8%, 38.9%, 70.3% by CT alone, and 88.0%, 85.4%, 86.3%, 75.9%, 93.2% by PET-CT, respectively. There was a statisticalty significant difference between two methods(P < 0.05). However, statisticalty no significant difference in detecting hilar lymph nodes was observed. The radiation target volume defined by CT was identical to that by PET-CT in 31(53.5%) cases, but changed in the other 27 (46.5%) cases when 18F-FDG PET data was added. The accuracy was 75.9% in detecting metastatic lymph nodes in the radiation target volume defined by PET-CT, while it was much lower by CT alone (48.3%) with a statistically significant difference between two methods (P < 0.05).
CONCLUSION18F-FDG PET-CT is more accurate than CT alone in assessment of mediastinal lymph nodes for NSCLC patients. It may be recommended as a method in defining the radiation target volume.
