1. Expression of cyclooxygenase-2, urokinase plasminogen activator and thrombospondin-1 in gastric carcinoma and surrounding peripheral lymph nodes

Objective: To investigate the expression of cyclooxygenase-2 (COX-2), urokinase plasminogen activator(u-PA), and thrombospondin-1 (TSP-1) in gastric carcinoma and the surrounding lymph nodes, so as to discuss their roles in the invasion and metastasis of gastric carcinoma. Methods: The expression of COX-2, u-PA, and TSP-1 in the samples was examined by tissue microarray and S-P immunohistochemistry staining. Results: The positive rates of COX-2, u-PA, and TSP-1 in gastric carcinoma were 67.7%, 78.1%, and 78.6%, respectively, which were significantly higher than those in the surrounding lymph nodes (40.0%, 6.7%, and 40.0%, respectively, P<0.05). The expression of COX-2 was significantly correlated with the depth of invasion (P<0.05). The expression of u-PA was significantly correlated with lymph node metastasis and depth of invasion (P< 0.05). The expression of TSP-1 was neither correlated with lymph node metastasis nor depth of invasion (P>0.05). COX-2, u-PA, and TSP-1 expression was not significantly different between metastatic lymph nodes and corresponding gastric carcinoma. COX-2 expression was positively correlated with u-PA, and TSP-1 expression. TSP-1 expression was not correlated with u-PA expression. Conclusion: COX-2, u-PA, and TSP-1 are highly expressed in the gastric carcinoma. COX-2 expression is correlated with depth of invasion. U-PA expression is significantly related with depth of invasion and lymph node metastasis.
2.Main methods of computational pharmacology and their application in research on traditional Chinese medicine pre-scriptions for treating chronic liver diseases

Traditional Chinese medicine (TCM)has certain advantages in the treatment of chronic liver diseases,especially chronic inflam-matory injury and liver fibrosis.However,the studies on medicinal components and action mechanism of TCM prescriptions are confronted with a great challenge.The computational pharmacology methods used in recent years,mainly including virtual screening technique,omics technologies,and network pharmacology approaches,as well as their application in TCM research,are presented;given the previous re-search work,the progress in research on active ingredients and action mechanism of TCM prescriptions using computational pharmacology methods is reviewed.It is thought that computational pharmacology techniques combined with conventional molecular biological and pharma-cological methods are an important research strategy for the material basis and action mechanism of TCMprescriptions for treating chronic liv-er diseases.
3.Mapping and analysis of the methylation variable positions profiles of hMLH1 promoter CpG islands in human sporadic colorectal carcinoma

Objective To map the complete methylation status of the hMLH1 promoter in sporadic colorectal carcinoma and analyze the relationship between MVPs (methylation variable positions) of hMLH1 promoter and the expression of hMLH1. Methods Methylation status of hMLH1 promoter was measured by bisulfite sequencing. hMLH1 protein expression was detected by immunohistochemistry. Results Out of the 30 sporadic colorectal carcinoma specimens, the hypermethylation of CGIs (CpG islands) 1 was in 6 and that of CGIs 2 in 4. The hMLH1 protein was detected in 15 specimens. Chi square test showed the methylation of CGIs 1 was closely related to loss of hMLH1 protein expression (P0.05). Conclusion In CGIs 1, CpG positions from 1 to 28 are the critical region that could influence the expression of hMLH1.

5.Study on the Saccharification Processes of Lignocellulose Brought about by Ultrasonic Wave

Both pretreatment of lignocellulose and its saccharification process are treated with ultrasonic wave.The morphology,structure and crystal performance of the original and treated lignocellulose sample were characterized by SEM and FTIR.Moreover,the changes of raw materials caused by different pretreatment ways and the affect of saccharification rate brought about by ultrasonic wave were also studied.The result shows that the ultrasonic wave decreases the crystallinity of lignocellulose destroying the intermolecular hydrogen bonding effectively and improves the degradation rate of lignin and the saccharification rate of zymohydrolysis availably.The mechanism of activation of ultrasonic wave in zymohydrolysis process was discussed primarily.
6.Feasibility of regional platform for wound care technical training

Objective To study the feasibility of regional network platform for wound care technical training.Methods Micro-letter pubic number and internet technologies were used to develop a regional network platform of wound care which has one center with multiple branches,training nurses how to take care of wound by the educational column.Results There were 383 nurses had browsed the web site and took part in the learning since regional network platform was working 3 months,931 nurses paid close at-tention to Micro-letter pubic number.Performance examination was improved obviously after training,the percent of pass was 17.8% before training and 80.7% after training,the difference was with statistical significance (χ2=303.629,P<0.01).Conclu-sion The regional network platform of wound care based on micro-letter and internet technologies has a promising application.
7.The establishment of emergency preplan for workplace violence in emergency department

Objective To prevent the occurrence of violence in the emergency department,to reduce the damage to medical personnel,and to maintain the normal medical order in emergency department.Methods A retrospective analysis of medical incidents of violence in the emergency department has been made.A root cause analysis (RCA) group has been established to explore the root cause of the incident and to make the violence emergency preplan of emergency department.Results Medical personnel have been trained according to the plan.It has improved the awareness of health care workers.The incidence of violent injury incidents decreased.Conclusions The establishment and training of emergency preplan can improve the awareness and prevention ability of emergency medical staff to violence injury.The safety of medical staff in emergency department can be guaranteed.
8.Establishment of experimental model of doxorubicin extravasation injury in mice

Objective To establish animal models in order to provide an experimental study basis for both the pathogenesis study and taking effective prevention scheme for doxorubicin extravasation injury. Methods A total of 20 Kunming mice for experiments on doxorubicin extravasation injury were divided in-to four groups, I.e., high dose group(2 g/L), medium dose group(1 g/L), low dose group(0.5 g/L) and the control group (injection with water). Dosages were administered with subcutaneous injection on both sides of mice abdomen. The adverse reaction of body, damage areas of extravasation injury, recovery period were observed and histopathologic slides of animal models on both 4 days and 11 days after experiment were performed and compared. Results No significant adverse body reaction was observed after injection for all groups. The damage areas due to extravasation injury were dosage and concentration dependent. In addi-tion, significant differences in recovery period were observed for mice in different groups, that is, the higher injection concentration and dose led to the longer recovery period. Results from the histopathologic study in-dicated that the putrescence of damage area was developed in high dose group mice, and the ulcer occurred after 4 d of dosage in medium dose group mice, respectively. In contrast, no ulcer was observed in low dose group mice. Conclusions It would be feasible to establish a prevention model for mice on doxorubicin extravasation injury by subcutaneous injection at a dosage of 0.05ml(1 g/L).
