1.Relationship between ionization and the expression of HIF-1α and P53 in anoxia condition

Objective To investigate the effect of ionization to the expression of HIF-1α and p53 of nasopharyngeal carcinoma (NPC) in anoxia condition. Method Nasopharyngeal carcinoma cell line CNE-1 was divided into three groups, control group, radiation group, and hypoxia plus radiation group. The cell viability was analyzed by MTT method, Immunofluorescent technique was performed to de-termine the expression location of HIF-1α in CNE-1. And the expression of HIF-1α and P53 protein were measured with flow cytometry. Re-sults The results of MTr detection showed that the survival fraction of the control group was the highest. The minimum survival fraction was found in the radiation group (P<0.05). There was no HIF-1αprotein expression in cytoplasm except the radiation plus hypoxia group. With the flow cytometer technique, the expression of HIF-1αprotein in the radiation plus hypoxia group was higher than that in the radiation group. No statistical significance was found between the radiation group and the control group. When detecting the expression of P53 protein with the same technique, the expression of P53 was highest in the hypoxia plus radiation group and lowest in the control group (P<0.05). Conclusion The expression of HIF-1α inducted by Hypoxia can prevent NPC cell from the damages of radiation. The HIF-1α decreases the radiosensitivlty through inducing p53 expression in NPC CNE-1 cell line.
2.Expression of hypoxia inducible factor-1? in HepG2 cells cultured under hypoxia and its relationship with vascularization and apoptosis-related protein

Objective:To investigate the expression of hypoxia inducible factor-1?(HIF-1?) in HepG2 cells cultured under hypoxia for different time periods, and to assess its relationship with vascularization and cell apoptosis. Methods: HepG2 cells were cultured in a medium containing cobalt chloride (125 ?mol/L) for different time periods ( 0, 1, 2, 4, 6, and 8 hours ).RT-PCR was used to measure the expression of HIF-1? mRNA,VEGF mRNA,and bcl-2 mRNA, and bax mRNA at the above time points; Western blot was used to determine the expression of Caspase-3 protein and the activity of Caspase-3 enzyme. Results: After 1-2 hour culture under hypoxia, the expression of HIF-1? mRNA,VEGF mRNA,and bcl-2 mRNA began to increase gradually, peaked at 4 h, and then decreased, but were still higher than that before culture. There was no obvious change in the expression of bax mRNA and the ratio of bcl-2 to bax mRNA expression had a similar change with that of bcl-2 mRNA expression. The changes of Caspase-3 protein/mRNA and Caspase-3 enzyme activity were contrary to that of HIF-1?. Conclusion: HIF-1? may inhibit the apoptosis of hepatic cancer cells through regulating the expression of VEGF, bcl-2, bax, and Caspase-3, and the inhibition is related to the severity of hypoxia.
3.Nuclear Factor Kappa B Involed in Immunologic Function of Critically Ill Newborns

Objective To observe the immunologic function of critically ill newborn and the relative function of nuclear factor kappa B(NF-?B).Methods The critically ill group contained 50 cases,and 25 cases from healthy newborns were used as control group.Blood samples were collected in each case,levels of cytokine interleukin(IL)-4,interferon(IFN)-?,tumor necrosis factor(TNF)-? and NF-?B were detected.Result Compared with control group,NF-?B of the critically ill newborn activated and the cytokine were disorder,and IL-4 and TNF-? increased,but IFN-? decreased.Conclusions Critically ill newborn exist immune functional disorder.Furthermore,NF-?B activation may be involved in the process in infants with critically illness.
4.Genetic polymorphism of HIF-1α G1790A associated with radiosensitivity of nasopharyngeal carcinoma

Objective To explore the association between the genetic polymorphism of hypoxia inducible factor 1 α (HIF-1α) G1790A and the radiosensitivity of nasopharyngeal carcinoma.Methods A total of 189 patients with nasopharyngeal carcinoma treated with radical radiotherapy were followed-up for 3 years.The patients were divided into cured group with 135 cases and recurrence group with 54 cases by clinical follow-up results.PCR-RFLP was used to determine the mononucleotide genotypes of HIF-1α G1790A.Results The observed genotype frequencies of HIF-1α gene 1790 (G→A) for GG, GA and AA were 70.04% , 20.74% , 2.22% in cured group and 59.26% , 38.89% , 1.85% in recurrence group, respectively.The allele frequencies for G and A were 87.4% , 13.9% in cured groups and 78.7% ,21.3% in recurrence group, respectively, without significant difference in distribution of allele frequencies between the two groups(x2 =6.919, P =0.077).Conclusions The genetic polymorphisms of HIF-1α G1790A might be related with the radiosensitivity of nasopharyngeal carcinoma.
5.The therapeutic effects and long-term follow-up of memory alloy embracing fixator for the treatment of the multiple fracture of ribs

Objective To explore the therapeutic effects of Ni-Ti memory alloy embracing fixator on the multiple fracture of ribs and flail chest. Method From January 2006 to December 2009, the effects of operation and long-term follow-up after internal fixation operation with memory alloy embracing fixator in patients with multiple fracture of ribs and flail chest were observed. Results The hospital stay of patients treated with memory alloy embracing fixator was 10.31 ± 3.14 days and pain was significantly reheved after treatment ( P= 0.02) .The rate of post-operative complications was 17.65% such as atelectasis, pulmonary infection, etc. There were fewer longterm complications and less influence to daily activity of life. Conclusions It is a advanced and practical method for patients with the multiple fracture of ribs by using Ni-Ti memory alloy embracing fixator with slighter trauma,simple, safe, reliable, good histocompatibility and fewer post-operative complications.
6.Effect of Pingwei Xiaodao Capsule on Gastric Motility of Functional Dyspepsia Model Rats

Objective To observe the effect of leptin on glucose oxidation with increased mRNA expression of complete length long intracellular domain of leptin receptor ( OBRb) in primary cultured skeletal muscle cells. Methods Skeletal muscle cells were isolated from SD sucking rat and cultured in four groups (recombinant transfection, empty plasmid transfection, non-transfection and control).The recombinant with complete length OBRb cDNA or the empty plasmid were introduced into skeletal muscle cells by clonfectin when the cultured cells were 70% conlluency, leptin and regular insulin were applied into wells and incubated for 1 h, then D-[ U-I4C]-glucose was added into wells and incubated for 2 h. mRNA in ARE and GW9662 group were 2.276 ±0.534 and 0.362 ±0.026,respectively.Compared with control group,PPARγmRNA level in both of ARE and GW9662 group reached statistical significance (t =4.785,P =0.001 ;t =2.395,P =0.044).PPARγprotein expression in ARE group,GW9662 +ARE group and control group were 27 688.33 ±3 593.06,21 816.00 ±1 644.07,17 716.33 ±2 273.95,respectively,which was higher in ARE group than that in control and GW+ARE group (t =5.159,P =0.001 ;t =3.038,P =0.016). NF-κB p65 mRNA expression in GW9662 +ARE group was 0.346 ±0.149,which in ARE group and GW9662 group were 0.392 ±0.1 87 and 1 .720 ±0.338,respec-tively.The differences of NF-κB p65 mRNA expression level between ARE,and control or GW9662 group were statistically significant (t =3.592,P =0.007;t =7.851 ,P =0.000).While,the differences of Caspase-3 mRNA and protein expression levels among the four groups were not statistically significant (F =1 .1 81 ,P =0.376;F =0.647,P >0.05).Conclusion ARE may restrain NF-κB through up-regulating PPARγto inhibit the proliferation and invasive potential of LLC in vitro, which suggests that PPAR-γmay be a novel therapeutic target for lung cancer.
9.Expression of IP-10 mRNA in peripheral blood mononuclear cells is correlated with serum levels of HBsAg and HBV DNA and of value for prognosis in patients with HBV-related acute on chronic liver failure

Objective To investigate the correlations of expression of interferon inducible protein 10 ( IP-10) mRNA in peripheral blood mononuclear cells ( PBMCs) with serum levels of HBsAg and HBV DNA in patients with HBV-related acute on chronic liver failure ( HBV-ACLF ) , and to assess its value in predicting disease prognosis .Methods Eighty patients with HBV-ACLF, 60 patients with chronic hepatitis B (CHB), and 25 healthy subjects were enrolled during October 2013 and February 2015.IP-10 mRNA in PBMCs was measured by real time quantitative polymerase chain reaction ( RT-qPCR) , and the difference in IP-10 mRNA expression among three groups was compared by ANOVA .Serum levels of HBsAg and HBV DNA were also measured in patients with HBV-ACLF, and Pearson correlation test was performed to analyze the correlations of IP-10 mRNA with HBsAg and HBV DNA levels .t test was used to analyze the differences in IP-10 mRNA, HBsAg and HBV DNA levels between fatal and surviving cases after 3-month entecavir therapy in HBV-ACLF group.Receiver operating characteristic curves ( ROC) was used to evaluate the prognostic value of IP-10 mRNA, HBsAg and HBV DNA in HBV-ACLF patients .Results IP-10 mRNA level in HBV-ACLF group was 0.998 ±0.186, which was higher than those in CHB patients and healthy controls (0.641 ±0.083 and 0.412 ±0.062, t=3.841 and 16.661, P<0.01).Pearson correlation analysis showed that IP-10 mRNA level was negatively correlated with HBsAg level in HBV-ACLF patients (r=-0.576, P<0.01), but positively correlated with HBV DNA level (r=0.547, P<0.01).After 3-month entecavir therapy , IP-10 mRNA level in surviving cases of HBV-ACLF group was 0.894 ±0.164, which was lower than that in fatal cases ( 1.103 ±0.177, t =-4.328, P <0.01 ); HBsAg level in surviving cases was higher than that in fatal cases (3.303 ±0.565 vs.2.605 ±0.844, t =3.251, P<0.01).The area under ROC of IP-10 mRNA in evaluating prognosis of HBV-ACLF was 0.820, which was higher than those of HBsAg (0.663) and HBV DNA (0.570).Conclusions IP-10 mRNA in patients with HBV-ACLF is over-expressed and is correlated with HBsAg and HBV DNA levels .It may be used for predicting the prognosis of patients with HBV-ACLF.

Now symptomatic extracranial and intracranial stenting usually uses bare metal stents, but the incidence of restenosis is higher.The studies of drug-eluting stems for the treatment of coronary atherosclerosis have suggested that the incidence of restenosis decreases significantly. Although extracranial and intracranial drug-eluting stem placement appears to be safe,further large,prospective,randomized,controlled trials are needed to demonstrate its safety and effectiveness.
