Objective To explore the contribution of endoplasmic reticulum stress to cardiac myocyte apoptosis in mouse congestive heart failure induced by Doxorubicin.Methods Doxorubicin (DOX)was administered intraperitoneally to rats,2 mg/kg doses over 8 weeks.Echocardiographic and hemodynamics measurements were obtained at 8 weeks after treatment.Then left ventricular (LV) samples were collected.The successfully established rats were divided into 3 groups (Fu Shen Ke Li 40 mg/kg,60 mg/kg,and 80 mg/kg).After 5 weeks post the operation,an echocardiographic was performed on all the rats under narcotism,Western blot to test the B-cell lymphoma-2 (Bcl-2),myeloid cell leukemia-1 (Mcl-1),Bcl-2-Associated X (Bax),Caspase-3,C/EBP homologous protein (CHOP),and 78-kDa glucose-regulated protein(GRP78),and reverse transcription-polymerase chain reaction (RT-PCR) to test the mRNA expression levels of Mcl-1,and NOXA.Results The left ventricular end-diastolic diameter (LVEDD),left ventricular end systolic diameter (LVESD),left ventricular ejection fraction (LVEF) prompted modeling success The left ventricular end-diastolic diameter (LVEDD),left ventricular ejection fraction (LVEF),and fractional shortening (FS) have been improved in high and middle groups of Fu Shen Ke Li.Compared to the model group,higher expression levels of Bcl-2,and Mcl-1 were,lower expressions of Bax,NOXA,CHOP,and GRP78 were (P ＜ O.05) in 3 groups of Fu Shen Ke Li.Compared to the model group,there were higher mRNA expression level of Mcl-1 was,lower mRNA expression of NOXA was (P ＜ 0.05) in 3 groups of Fu Shen Ke Li.Conclusions Fu Shen Ke Li depresses endoplasmic reticulum stress,which can reduce the apoptosis of myocardial cells,and ultimately achieve the purpose of treatment of heart failure.

Objective To screen the proteins interacting with AMP-activated protein kinase ?2(AMPK?2)in rat brain cDNA library by bacterial two-hybrid system,and to investigate the biological role and the regulatory mechanism of AMPK?2 in brain.Methods The recombinant pBT-AMPK?2 was used as the bait to screen a rat fetal brain cDNA library by bacterial two-hybrid system.The plasmids of positive colonies were extracted and analyzed by DNA sequencing and BLAST search in GenBank.Results Seven proteins binding to AMPK?2 were screened out,including polyubiquitin,small heat shock protein 8,phosphofructokinase,cytochrome C oxidase subunit Ⅰ(COXⅠ),HLA-B-associated transcript 3(BAT3)isoform 1,protein tyrosine phosphatase receptor type D(Ptprd)and islet-brain 1(IB1).Conclusion Polyubiquitin,HSP8,phosphofructokinase,COXⅠ,BAT3 isoform 1,Ptprd and IB1 can interact with AMPK?2 in brain.

Objective To understand the relationship between Toxoplasma gondii infection and pet-breeding and hygienic status. Methods One or two districts were chosen from Jining City, Za-ozhuang City and Linyi City as study areas and the questionnaire surveys were carried out for hygienic status, etc. The pet-breeders and non-pet-breeders were paired by sex, age, living place and occupation so on in a ratio of one to one. Blood samples taken from each were tested for antibodies against T. gondii with ELISA in a double-blind method. Results The total infection rate was 14. 25%, much higher than that investigated ten years ago (1. 53%). Objective:To express and purify recombinant human collagen type Ⅲ and evaluate its properties.Methods:The recombinant genetic engineering strain pET30a(+)-1880/pACYCDuet-hy726/bL21(DE3) was constructed to stably co-express recombinant human type Ⅲ collagen (rhCol) and prolyl hydroxylase. rhCol was prepared and purified by E. coli high-density fermentation, salting out and column chromatography protein purification technology. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis was used to determine the purity of rhCol. The N-terminal amino acid sequences of rhCol were determined by automatic protein polypeptide sequencing instrument. The hydroxyproline content of rhCol was determined by ultraviolet spectrophotometry. The cellular compatibility of rhCol was evaluated by MTT assay. Results:The final wet weight of high-density fermentation was about 200 g/L. The expression level was about 3 g/L. The purity of rhCol by affinity chromatography was over 95%. The results showed that the hydroxyproline content of rhCol was 11.44%, and the rhCol products have good water solubility and cell compatibility.Conclusions:RhCol can be widely applied to the field of skin care and biomedicine as an excellent biological material.

OBJECTIVETo analyze the influence of retinoic acid (RA) on the undifferentiated state and EB formation abilities of human embryonic stem cells.

METHODSThe biological characteristics of H9 ESCs after RA treatment were characterized by real-time PCR, MTS proliferation assay and immunofluorescence staining. The expression of three germ layers markers, osteogenic differentiation markers and adipogenic differentiation markers in H9-differentiated embryoid bodies (EBs) with RA treatment were quantified by real time PCR.

RESULTSThe proliferation of H9 ESCs in the early logarithmic growth phase was accelerated by RA treatment. In addition, RA induced differentiation of H9 ESC coupled with morphology changes, decreased expression of undifferentiated markers Oct4, Nanog, Sox2 and OCT4 mRNA binding protein Lin28 at mRNA level, and reduced expression of Oct4 at protein level. RA induced formation of cavities in EBs. Real time PCR results showed that the expressions of ectodermal markers: NeuroD1, Noggin; mesodermal markers: Brachyury, Twist and endodermal markers: AFP, GATA-4 were significantly increased (P < 0.05), especially for AFP (P < 0.01), by RA treatment in a dose-dependent manner. In addition, the expression of adipogenic differentiation marker C/EBPalpha was increased while the osteogenic differentiation marker OPN was decreased in EBs after RA treatment for 5 days.

CONCLUSIONSHigh concentrations of RA induced the loss of stemness in H9 ESCs and excessive differentiation in EBs, and damaged the balance between osteogenic and adipogenic differentiation during early EB differentiation, which may be relevant to the congenital malformations.

Objective:To analyze adverse drug reactions(ADRs)associated with proprietary Chinese medicine occurring in Beijing Hospital.Method:The ADRs associated with proprietary Chinese medicine between 2002 and 2006 were collected and the associated information was retrospectively analyzed.Result:a total of 179 ADRs were analyzed. Most of them occurring in women patients(76.1%)with hyperplasia of mammary glands who took Xiaojin pills and Xi- huang pills(22.1% & 12%).The herbal medicine injections were the other most associated ones.The clinical appearances of ADRs were rash and bodily manifestation of hypersensitiveness.Condusion:The investigation of Chinese medicine ADR plays a very important role in the modernization of Chinese medicine.

Objective To study the characteristics of MRI Lesions in posterior circulation cerebral infarction and the correlation of the syndrome types of traditional Chinese medicine.Methods 82 patients with posterior circulation cerebral infarction from May 2012 to May 2015 were recruited, two observers estimated cranial MRI infarct size, location, and TCM syndrome type in the hospital medical records of the patients separately.Results Of all patients with posterior circulation cerebral infarction, patients with meridians involved infarction in the distant part of posterior circulation is far more than other parts of the posterior circulation(21 patient,P=0.006), nevertheless,patients with apoplexy involving Zang-fu organs,cerebral infarction volume is larger (14.78 ± 5.68 mlvs. 9.12 ± 6.67 ml,P=0.001).Conclusion Of all patients with posterior cerebral infarction, patients with meridians be involved, infarction in the distant part of posterior circulation is far more than other parts of the posterior circulation, nevertheless, in patients with apoplexy involving Zang-fu Organs, cerebral infarction volume is larger.
